Comparative effect of the DNAse from rat liver chromatin and Neurospora crassa endonuclease S1 on closed circular superhelical DNA of PM-2 phage and Simian Virus 40 is studied. It is shown that both of them--the DNAse from chromatin proteins and endonuclease S1--are specific to single-stranded regions in DNA molecular. It is suggested that chromatin protein DNAse participates in reparation processes.  相似文献

The sea urchin embryo nuclei which retained their ability to maintain the DNA synthesis in an in vitro system were isolated. The DNA synthesis isolated nuclei was shown to be an ATP-dependent process which is inhibited by low concentrations of actinomycin D, a polymerase alpha araCTP inhibitor. The newly synthesized DNA is represented by short fragments of about 4S. After addition of Ca2+, Mg2+-dependent DNAase to sea urchin embryo nuclei, the synthesis of short DNA fragments is enhanced. This stimulating effect of Ca2+, Mg2+-dependent DNAase is ATP-dependent and is observed only within a narrow range of enzyme concentrations (of the order of 1-5 units of DNAase activity per ml of incubation sample). The increase in the enzyme concentration to 10 or more units of activity results in the depression of DNA synthesis. It is concluded that DNA replication in sea urchin embryo nuclei depends on the presence of active DNAases as well as on the number of accessible initiation sites of DNA replication.  相似文献

Endocellular and exocellular ribonucleases were studied in Bacillus intermedius. Two fractions of ribonucleases (Rf 0.72 and 0.96) were found to be associated with the cellular surface and seven fractions (Rf 0.1, 0.17, 0.33, 0.45, 0.72, 0.82 and 0.96) were detected in the cytoplasm. RNAase with Rf 0,096 had the highest activity and was repressed by inorganic orthophosphate. This RNase accumulated in the cell during the stationary growth phase just as the free enzyme form did in the culture medium. The immunological characteristics of these enzymes were identical as was shown by immunochemical analysis.  相似文献

Virus encoded RNA-silencing suppressors (RSSs) are the key components evolved by the viruses to counter RNA-silencing defense of plants. Whitefly-transmitted begomoviruses infecting tomato crop code for five different proteins, ORF AC4, ORF AC2 and ORF AV2 in DNA-A component, ORF BV1 in DNA-B and ORF βC1 in satellite DNA β which are predicted to function as silencing suppressors. In the present study suppressor function of ORF βC1 of three betasatellites Tomato leaf curl Bangalore betasatellite ToLCBB-[IN:Hess:08], Cotton leaf curl Multan betasatellite CLCuMB–[IN:Sri:02] and Luffa leaf distortion betasatellite LuLDB-[IN:Lu:04] were examined. Agroinfiltration of GFP-silenced Nicotiana tabaccum cv. Xanthi with the cells expressing βC1 protein resulted in reversal of silenced GFP expression. GFP-siRNA level was more than 50-fold lower compared to silenced plants in plants infiltrated with βC1 gene from ToLCBB. However, in the case of 35S-βC1 CLCuMB and 35S-βC1 LuLDB construct, although GFP was expressed, siRNA level was not reduced, indicating that the step at which βC1 interfere in RNA-silencing pathway is different.  相似文献

During a gross anatomy dissection, a piece of the calvarial bone was found between aponeuroses of the anterior abdominal wall in an 82-year-old male cadaver. It corresponded closely in size to the calvarial defect after the neurosurgery performed 5 years before death, showing that very little resorption had occurred over the years. Moreover, 13 percent of the transplant was new bone, indicating that membranous bone can, in the long term, induce new bone formation in humans.  相似文献

A pulse incorporation of radioactive precursors into the cells was used in studying the rate of RNA and protein synthesis during postgerminative development of spores ofBacillus cereus. It was found that the extractable pool of the spores can be enriched by these precursors during swelling and preelongation. During this period of differentiation of a spore to a primary cell,14C-uracil and to a smaller extent14C-adenine are more rapidly used for RNA synthesis. Labelled14C-leucine and36S-methionine are found mostly in the extractable fraction of the cells. However, their rate of incorporation into proteins is not higher. Differences in utilization of precursors for the synthesis of macromolecules are apparently caused by different availability for the metabolic pool of the cell. Of the precursors tested14C-uracil seems to be selectively incorporated into RNA with a higher rate. This can be explained by a selective permeability into metabolic pool or by an increased need of uracil during preelongation period.  相似文献
