Arabinogalactan type I from potato was partially degraded by endo-galactanase from Aspergillus niger. High-performance anion-exchange chromatography revealed that several of the oligomeric degradation products eluted as double peaks. To investigate the nature of these products, the digest was fractionated by Bio-Gel P2 chromatography. The pool that contained tetramers was treated with a beta-D-Galp-(1-->4)-specific galactosidase from Bifidobacterium adolescentis to obtain a dimer with deviating linkage type, which was further purified by BioGel P2 chromatography. By obtaining all (1)H and (13)C chemical shifts and the presence of intra residual scalar coupling (HMBC) it could be concluded that the dimer contained a beta-(1-->3)-linkage instead of the expected beta-(1-->4)-linkage. Using the same NMR techniques as for the dimer, it was found that the pool of tetramers consisted of the following two galactose tetramers: beta-Galp-(1-->4)-beta-Galp-(1-->4)-beta-Galp-(1-->4)-alpha/beta-Galp-OH and beta-Galp-(1-->4)-beta-Galp-(1-->4)-beta-Galp-(1-->3)-alpha/beta-Galp-OH. The fact that the deviating beta-(1-->3)-linked galactose was found at the reducing end of the dimer showed that this deviating linkage is present within the backbone. The beta-(1-->3)-galactosyl interruption appeared to be a common structural feature of type I arabinogalactans with a frequency ranging from approximately 1 in 160 (potato, soy, citrus) to 1 in 250 (onion).  相似文献

The production of dextran in potato tubers and its effect on starch biosynthesis were investigated. The mature dextransucrase (DsrS) gene from Leuconostoc mesenteroides was fused to the chloroplastic ferredoxin signal peptide (FD) enabling amyloplast entry, which was driven by the highly tuber-expressed patatin promoter. After transformation of two potato genotypes (cv. Kardal and the amylose-free (amf) mutant), dextrans were detected by enzyme-linked immunosorbent assay (ELISA) in tuber juices of Kardal and amf transformants. The dextran concentration appeared two times higher in the Kardal (about 1.7 mg/g FW) than in the amf transformants. No dextran was detected by ELISA inside the starch granule. Interestingly, starch granule morphology was affected, which might be explained by the accumulation of dextran in tuber juices. In spite of that, no significant changes of the physicochemical properties of the starches were detected. Furthermore, we have observed no clear changes in chain length distributions, despite the known high acceptor efficiency of DSRS.  相似文献

Two endoglucanases of Trichoderma viride, endoI and endoIV, were assayed for their activity toward alkali-extracted apple xyloglucans. EndoIV was shown to have a 60-fold higher activity toward xyloglucan than endoI, whereas carboxymethyl cellulose and crystalline cellulose were better substrates for the latter. The enzymic degradation of cellulose embedded in the complex cell-wall matrix of apple fruit tissue has been studied using cellobiohydrolase (CBH) and these two different endoglucanases. A high-performance liquid chromatographic method (Aminex HPX-22H) was used to monitor the release of cellobiose and oligomeric xyloglucan fragments. Synergistic action between CBH and endoglucanases on cell-wall-embedded cellulose was, with respect to their optimal ratio, slightly different from that reported for crystalline cellulose. The combination of endoIV and CBH solubilized twice as much cellobiose compared to a combination of endoI and CBH. Apparently, the concomitant removal of the xyloglucan coating from cellulose microfibrils by endoIV is essential for an efficient degradation of cellulose in a complex matrix. Cellulose degradation slightly enhanced the solubilization of xyloglucans. These results indicate optimal degradation of cell-wall-embedded cellulose by a three-enzyme system consisting of an endoglucanase with high affinity toward cellulose (endoI), a xyloglucanase (endoIV), and CBH.  相似文献
