This study investigated endocrine control of branchial ionoregulatory function in Nile tilapia (Oreochromis niloticus) by prolactin (Prl188 and Prl177), growth hormone (Gh) and cortisol. Branchial expression of Na+/Cl? cotransporter (ncc) and Na+/K+/2Cl? cotransporter (nkcc) genes were employed as specific markers for freshwater- and seawater-type ionocytes, respectively. We further investigated whether Prl, Gh and cortisol direct expression of two Na+, K+-ATPase (nka)-α1 subunit genes, denoted nka-α1a and nka-α1b. Tilapia transferred to fresh water following hypophysectomy failed to adequately activate gill ncc expression; ncc expression was subsequently restored by Prl replacement. Prl188 and Prl177 stimulated ncc expression in cultured gill filaments in a concentration-related manner, suggesting that ncc is regulated by Prl in a gill-autonomous fashion. Tilapia transferred to brackish water (23 ‰) following hypophysectomy exhibited a reduced capacity to up-regulate nka-α1b expression. However, Gh and cortisol failed to affect nka-α1b expression in vivo. Similarly, we found no clear effects of Gh or cortisol on nkcc expression both in vivo and in vitro. When considered with patterns previously described in euryhaline Mozambique tilapia (O. mossambicus), the current study suggests that ncc is a conserved target of Prl in tilapiine cichlids. In addition, we revealed contrasting dependencies upon the pituitary to direct nka-α1b expression in hyperosmotic environments between Nile and Mozambique tilapia.  相似文献

During experimental cerebral malaria (ECM) mice develop a lethal neuropathological syndrome associated with microcirculatory dysfunction and intravascular leukocyte sequestration. The precise spatio-temporal context in which the intravascular immune response unfolds is incompletely understood. We developed a 2-photon intravital microscopy (2P-IVM)-based brain-imaging model to monitor the real-time behaviour of leukocytes directly within the brain vasculature during ECM. Ly6Chi monocytes, but not neutrophils, started to accumulate in the blood vessels of Plasmodium berghei ANKA (PbA)-infected MacGreen mice, in which myeloid cells express GFP, one to two days prior to the onset of the neurological signs (NS). A decrease in the rolling speed of monocytes, a measure of endothelial cell activation, was associated with progressive worsening of clinical symptoms. Adoptive transfer experiments with defined immune cell subsets in recombinase activating gene (RAG)-1-deficient mice showed that these changes were mediated by Plasmodium-specific CD8+ T lymphocytes. A critical number of CD8+ T effectors was required to induce disease and monocyte adherence to the vasculature. Depletion of monocytes at the onset of disease symptoms resulted in decreased lymphocyte accumulation, suggesting reciprocal effects of monocytes and T cells on their recruitment within the brain. Together, our studies define the real-time kinetics of leukocyte behaviour in the central nervous system during ECM, and reveal a significant role for Plasmodium-specific CD8+ T lymphocytes in regulating vascular pathology in this disease.  相似文献

Nasal colonization is a major risk factor for S. aureus infections. The mechanisms responsible for colonization are still not well understood and involve several factors on the host and the bacterial side. One key factor is the cell wall teichoic acid (WTA) of S. aureus, which governs direct interactions with nasal epithelial surfaces. We report here the first receptor for the cell wall glycopolymer WTA on nasal epithelial cells. In several assay systems this type F-scavenger receptor, termed SREC-I, bound WTA in a charge dependent manner and mediated adhesion to nasal epithelial cells in vitro. The impact of WTA and SREC-I interaction on epithelial adhesion was especially pronounced under shear stress, which resembles the conditions found in the nasal cavity. Most importantly, we demonstrate here a key role of the WTA-receptor interaction in a cotton rat model of nasal colonization. When we inhibited WTA mediated adhesion with a SREC-I antibody, nasal colonization in the animal model was strongly reduced at the early onset of colonization. More importantly, colonization stayed low over an extended period of 6 days. Therefore we propose targeting of this glycopolymer-receptor interaction as a novel strategy to prevent or control S. aureus nasal colonization.  相似文献

A new ligand based on a ruthenocene moiety appended by a diyrromethene (Rc-dpm) was synthesized. Two copper complexes, Cu(Rc-dpm)2 (3) and Cu(Rc-dpm)acac (4), and one cobalt complex, Co(Rc-dpm)3, 5, were prepared from the ligand. These complexes were characterized by a combination of UV-Vis spectroscopy, elemental analysis, and X-ray crystallography. Copper(II) complex 3 was found to be coordinated by two dipyrromethene ligands in a distorted square planar environment around the copper while complex 4 had one dipyrromethene ligand and an acac coordinated in a nearly idealized square planar copper geometry. The cobalt(III) complex 5 has an octahedral geometry around the cobalt via the coordination of three dipyyromethene ligands.  相似文献

Degraded grasslands resulting from intensive land use appear to be highly resistant to tree invasion due to interactions between land use, climate, grazing and fire. We describe long-term patterns of tropical montane forest regeneration into degraded grasslands and analyze their relationships with historical changes in rainfall, grazing and fire in Los Toldos valley (Northwest Argentina), cloud forest life zone (1600 m asl). We used dendrochronological techniques to reconstruct spatial and temporal patterns of Podocarpus parlatorei establishment (the dominant tree species in secondary forests) and grassland fires for the last 150 yr. We assessed current livestock spatial distribution along the valley through feces sampling. Inferred tree establishment patterns ( i.e ., considering age structure and mortality) were analyzed in relation to temporal and spatial patterns of grazing and fire derived from our own analyses and from government statistics, and to rainfall patterns derived from previous dendrochronological reconstructions. Current grazing intensity was higher close to the local township. Fire occurrence increased with periods of above-average rainfall (higher fuel productivity), and tended to increase with distance to township (less grazing). Tree establishment in grasslands was spatially associated with high grazing intensity and low fire frequency, and temporally associated with periods of high grazing intensity and below-average rainfall. Despite climatic and land-use changes leading to conditions potentially favorable for trees ( i.e ., more rainfall, less grazing), grasslands persist in this study area, likely due to the direct (saplings burning) and indirect (soil degradation and desiccation) effects of recurrent fires, enhanced by decreasing grazing and increasing rainfall.

Background

Direct cellular contact with stimulated T cells is a potent mechanism that induces cytokine production in human monocytes in the absence of an infectious agent. This mechanism is likely to be relevant to T cell-mediated inflammatory diseases such as rheumatoid arthritis and multiple sclerosis. Microparticles (MP) generated by stimulated T cells (MPT) display similar monocyte activating ability to whole T cells, isolated T cell membranes, or solubilized T cell membranes. We previously demonstrated that high-density lipoproteins (HDL) inhibited T cell contact- and MPT-induced production of IL-1β but not of its natural inhibitor, the secreted form of IL-1 receptor antagonist (sIL-1Ra).

Methodology/Principal Findings

Labeled MPT were used to assess their interaction with monocytes and T lymphocytes by flow cytometry. Similarly, interactions of labeled HDL with monocytes and MPT were assessed by flow cytometry. In parallel, the MPT-induction of IL-1β and sIL-1Ra production in human monocytes and the effect of HDL were assessed in cell cultures. The results show that MPT, but not MP generated by activated endothelial cells, bond monocytes to trigger cytokine production. MPT did not bind T cells. The inhibition of IL-1β production by HDL correlated with the inhibition of MPT binding to monocytes. HDL interacted with MPT rather than with monocytes suggesting that they bound the activating factor(s) of T cell surface. Furthermore, prototypical pro-inflammatory cytokines and chemokines such as TNF, IL-6, IL-8, CCL3 and CCL4 displayed a pattern of production induced by MPT and inhibition by HDL similar to IL-1β, whereas the production of CCL2, like that of sIL-1Ra, was not inhibited by HDL.

Conclusions/Significance

HDL inhibit both MPT binding to monocytes and the MPT-induced production of some but not all cytokines, shedding new light on the mechanism by which HDL display their anti-inflammatory functions.  相似文献

We used dendroecological techniques to analyze the effects of rainfall and grazing on fire regime and its implications for tree regeneration in subtropical mountains of northwestern Argentina during the 20th century, a period characterized by increasing rainfall and decreasing land-use intensity. We dated fire scars and establishment of Alnus acuminata (the dominant tree species) in six watersheds along a 600 km latitudinal range. We correlated fire frequency with rainfall records and performed Superposed Epoch Analyses to assess the relationship between rainfall and fire events during the century, and in two sub-periods: 1930–1965 (low rainfall, high grazing) and 1966–2001 (high rainfall, low grazing). We performed permutation analyses to assess the association between fire events and tree establishment, and to describe the spatial distribution of fires and forests in relation to hillslope aspect. Rainfall was associated with regional fires at interannual and decadal scales: fire probability increased after growing seasons with above-average rainfall and through the century, in concurrence with rainfall increase. The climatic control of fire was stronger under lower land-use intensity. Tree establishment was temporally associated with fire events, which occurred mainly in north facing slopes, where grassland cover is more extensive and forest colonization more likely. These results suggest that fire is limited by the availability of fine fuels, which is enhanced by high rainfall and reduced grazing; and tree establishment is limited by the competition with grasses. Consequently, increasing rainfall and decreasing grazing favored higher fire frequency, thus promoting forest encroachment during 20th century.  相似文献
