During experiments studying dietary effects on phosphorylation/dephosphorylation of MAP-2 we found that incubation of microtubules with alkaline phosphatase resulted in extensive proteolysis of MAP-2 but not of tubulin or Tau proteins. In the absence of tubulin, when microtubule-associated proteins (MAPs) were incubated with alkaline phosphatase, MAP-2 was not proteolyzed. This suggests that binding to tubulin induces a conformational change in MAP-2 which makes it more susceptible to proteolysis. The proteolysis of MAP-2 by alkaline phosphatase was prevented by inhibitors of serine proteases, suggesting that the commercial preparation of the enzyme is contaminated by a serine protease and/or that the enzyme also has a weaker proteolytic activity. In addition, selective proteolysis of MAP-2 can be obtained with the metalloprotease collagenase. Brain homogenates are shown to contain a Ca2+-dependent protease which selectively degrades MAP-2 bound to tubulin. These results suggest that selective proteolysis of tubulin-bound MAP-2 could play a role in the regulation of microtubule dynamics in response to extracellular signals.  相似文献

The histology of the digestive tract of the amberjack ( Seriola dumerili , Risso) was studied using light and scanning electron microscopy. The anterior oesophagus mucosa displays primary and secondary folds lined with a stratified squamous epithelium with fingerprint-like microridges which is substituted, on the top of the oesogaster folds, by a simple columnar epithelium with short microvilli. Only primary folds are present in the stomach. The anterior portion is rich in simple tubular glands, whereas the oesogaster and the pyloric region are devoid of them. Pyloric caeca and anterior and middle intestine mucosa display the same pattern of folding. The dominant cell type is the enterocyte, which exhibits larger and thinner microvilli in the caeca than in the intestine. The columnar epithelium of the rectum is replaced, in the anal sphincter, by a stratified flattened epithelium. Goblet cells are numerous throughout the whole length of the tract with the exception of the initial part of the oesophagus, the oesogaster, the stomach and the anal sphincter. Mucosubstances have been shown to vary in the different regions of the gut: acid mucines are found in the oesophagus, pyloric stomach, caeca, intestine and rectum, whereas neutral mucosubstances dominate in the anterior portion of the stomach. The muscularis is well developed throughout the length of the tract: two layers of striated muscle at the oesophageal level; two layers of smooth muscle in the stomach wall and three at the intestinal level.  相似文献

Imaginal pattern duplications caused by hypomorphic expression of the segment polarity gene costal-2 are described. These affect the anteroposterior coordinate of the imaginal disc. A very small part of the pattern is deleted and a large number of additional pattern elements arise in a progressive order, anterior-most first followed by more and more posterior structures. Mosaic analyses show that the duplications arise nonautonomously in the larval stages but that the costal-2 gene is not required after early embryogenesis. Arguments that the duplications are the result of cell interactions and intercalary growth that themselves arise from an abnormal polarity of the embryonic segment are presented.  相似文献

Treatment of the killifish, Fundulus heteroclitus, with ovine thyrotropin (oTSH) produced elevations in serum thyroxin (T4) that varied seasonally with a strong inverse correlation to environmental photoperiod (P less than 0.001). At 20 degrees C, oTSH (0.2 IU) increased serum T4 to levels ranging from 1.5 micrograms/100 ml in midsummer to 7.5 micrograms/100 ml in midwinter despite relatively stable resting levels throughout the two years of study. Similar rates of clearance of T4 in summer and winter suggest that an alteration in thyroid response to oTSH accounts for this change. The serum T4 response to oTSH is both photoperiod- and temperature-dependent. In laboratory studies, fish exposed to a short artificial photoperiod (LD 8:16) for one month responded to oTSH with T4 levels about twice as high as those in fish exposed to long days (LD 14:10). On the other hand the T4 response to oTSH was sharply reduced at 5 degrees C from that seen at 20 degrees C. This suggests that, in nature, seasonal changes in photoperiod and temperature have opposite effects on thyroidal responsiveness to TSH. Overall, the annual variation in the T4 response to oTSH appears to be driven by photoperiod, inasmuch as the alterations preceded major temperature changes in the wild and can be shown to occur at constant temperature in captivity. Photoperiodic induction of changes in thyroid sensitivity may aid in the maintenance of basal T4 levels under changing thermal conditions.  相似文献

Summary The conditions affecting the immunohistochemical identification of albumin in livers of male NMRI-mice were investigated by light microscopy. In normal livers albumin is randomly distributed, revealing a pancytoplasmic nearly homogen reaction in groups of hepatocytes or single parenchymal cells. However, combined autoradiographic studies after pulse labelling with 3H-valin and perfusion experiments with human albumin indicate that this distribution is caused by albumin from blood plasma and does not reflect true protein synthesis. After perfusion of the livers followed by immunohistochemical amplification techniques which allowed to dilute the primary antibody up to 1:30,000, albumin could be detected nearly in all liver parenchymal cells as granular deposits decreasing in its density from periportal fields towards the terminal hepatic venules.In regenerating livers due to partial hepatectomy no remarkable differences in granular albumin deposits between G1- and S-phase of the cell cycle could be detected as was demonstrated by combined immunohistochemistry and 3H-dThd-autoradiography. This investigation provides direct evidence that rat pineal gland contains ATP:L-methionine S-adenosyltransferase. The specific activity of the enzyme is about 25 times greater than that of hydroxyindole-O-methyltransferase, the unique marker enzyme in this tissue. The major product of the reaction catalyzed by ATP:L-methionine S-adenosyltransferase has been identified as S-adenosyl-methionine which can function as a methyl group donor for the formation of melatonin. The specific activity of pineal ATP:L-methionine S-adenosyltransferase is at least eight times greater than that of brain, and one-half that of the liver enzyme.  相似文献

Conditions for the conversion of palmitate into CO2 and acetoacetate by liver homogenates and isolated liver mitochondria are described. In this system, using liver homogenates, adenosine inhibited the conversion of palmitate into CO2 and acetoacetate. The inhibition was not observed if the homogenate was substituted by mitochondria or if palmitate was substituted by palmitoyl CoA or palmitoyl carnitine. Intraperitoneal injection of adenosine produced a marked decrease in the level of acetoacetate and β-hydroxybutyrate in plasma, without changing the concentration of serum free fatty acids. Thus, the nucleoside depressed in vivo the oxidation of long chain fatty acids in liver by inhibiting the extramitochondrial acyl CoA synthase(s). The paramount importance of the extramitochondrial activation of fatty acids as a key control in their oxidation and in the production of ketone bodies is discussed.  相似文献

The kinetics of cell division have been studied in a strain of Escherichia coli which has an amber mutation in the ftsA gene and which also carries a temperature sensitive amber suppressor. This strain is therefore temperature sensitive for the synthesis of the ftsA protein. Cells of this strain were able to divide only if the synthesis of this protein took place during a specific part of the cell cycle. This was a short period (roughly 10 min in duration) immediately before the normal time of cell division.  相似文献
