Genotype and water deficit effects on leaf 2-DE protein profiles of two Populus deltoides × Populus nigra, cv. ‘Agathe_F’ and ‘Cima’, were analysed over a short-term period of 18 days in glasshouse using 4-month-old rooted cuttings and over a long-lasting period of 86 days in open field using 4-year-old rooted cuttings. Leaf proteomes were analyzed using two-dimensional gel electrophoresis, and proteins were identified after database searching from MS peptide spectra.A reliable genotype effect was observed in the leaf proteome over experiment locations, water regimes and sampling dates. Quantitative differences between genotypes were found. Most of them corresponded to proteins matching isoforms or post-translational modification variants. However, ‘Cima’ displayed the highest abundance of antioxidant enzymes.In response to water deficit, about 10% of the reproducible spots significantly varied regardless of the experiment location, among which about 25% also displayed genotype-dependent variations. As a whole, while ‘Cima’ differed from ‘Agathe_F’ by increased abundance of enzymes involved in photorespiration and in oxidative stress, ‘Agathe_F’ was mainly differentiated by increased abundance of enzymes involved in photosynthesis.  相似文献

Plant height, light-saturated rates of photosynthesis (A max) and foliar nitrogen concentration (N 1) were measured forBartsia trixago under field conditions in Mallorca. All three variables were postively correlated, and were also positively related to the abundance of nitrogen-fixing legumes in the associated vegetation (putative host species).A max forB. trixago ranged from 7.7 to 18.8 mol m-2 s-1; similar rates were measured for a second hemiparasiteParentucellia viscosa, and both species were within the range of rates measured for six putative hosts (10.6–19.2 mol m-2 s-1). Fertilization of unattachedB. trixago plants with inorganic nitrogen (ammonium nitrate) elicited neither the growth nor the photosynthetic responses observed in plants considered to be parasitic on legumes and in receipt of an enriched organic nitrogen supply. Both hemiparasites had high diurnal leaf conductances (g s) (469–2291 mmol m-2 s-1) and were at the upper end of the range of those measured in putative hosts (409–879 mmol m-2 s-1). In contrast with the latter, high nocturnal rates ofg s were also recorded for the two hemiparasites (517–1862 mmol m-2 s-1). There was no clear relationship between eitherA max orN 1 and eitherg s, transpiration (E) or water use efficiency (A max/E) inB. trixago plants. The economics of water loss appear to be independent of both the supply of nitrogen from the host and autotrophic carbon fixation.  相似文献

The lipid dynamics of the adrenocortical microsomal membranes was studied by monitoring the fluorescence anisotropy and excited state lifetime of a set of anthroyloxy fatty acid probes (2-, 7-, 9- and 12-(9-anthroyloxy)-stearic acid (AP) and 16-(9-anthroyloxy)palmitic acid (AS). It was found that a decreasing polarity gradient from the aqueous membrane interface to the membrane interior, was present. This gradient was not modified by the proteins, as evidenced by comparison of complete membranes and derived liposomes, suggesting that the anthroyloxy probes were not in close contact with the proteins. An important change of the value of the mean rotational relaxation time as a function of the position of the anthroyl ring along the acyl chain was evidenced. In the complete membranes, a relatively more fluid medium was evidenced in the C16 as compared to the C2 region, while the rotational motion appeared to be the most hindered at the C7–C9 level. In the derived liposomes, a similar trend was observed but the mobility was higher at all levels. The decrease of the mean rotational relaxation time was more important for 12-AS and 16-AP. Temperature dependence of the mean rotational relaxation time of 2-AS, 12-AS and 16-AP in the complete membranes revealed the existence of a lipid reorganization occurring around 27°C and concerning mainly the C16 region. The extent to which the acyl chain reacted to this perturbation at the C12 level depended on pH. The presence of proteins increased the apparent magnitude of this reorganization and also modified the critical temperature from approx. 23°C in the derived liposomes to approx. 27°C in the complete membranes. Thermal dependence of the maximum velocity of the 3-oxosteroid Δ54-isomerase, the second enzyme in the enzymatic sequence, responsible for the biosynthesis of the 3-oxo4-steroids in the adrenal cortex microsomes, was studied. The activation energy of the catalyzed reaction was found to be low and constant (2–5 kcal · mol?1) in the temperature range 16–40°C at pH 7.5, 8.5 and 9, corresponding to the minimum, intermediate and maximum rate, respectively. A drastic increase of the activation energy (20 kcal · mol?1) was observed at temperature below 16°C at pH 7.5. A correlated change of the pKESapp as function of temperature was detected; at 36°C pKESapp = 8.3 while at 13°C the value shifted to 8.7. The pH range of the group ionization was narrower at 13°C. In contrast with the behaviour of the 3β-hydroxy5-steroid dehydrogenase, the 3-oxosteroid Δ54-isomerase was apparently unaffected by the lipid reorganization at 27°C. It is suggested that this enzyme possesses a different and more fluid lipid environment than the bulk lipids.  相似文献

The yeastSaccharomyces cerevisiaewas used for heterologous expression of the human CHIP28 water Aquaporin-1 channel (Aquaporin-1). A nine-amino-acid epitope of the influenza hemagglutinin protein (HA epitope), recognized by the monoclonal antibody 12CA5, was chosen to tag CHIP28 at its N-terminus. Epitope-tagged CHIP28 was purified from yeast extracts by immunochromatography on protein A/12CA5-coupled beads, after KI extraction and detergent solubilization, then concentrated by anion exchange chromatography. Purified protein was reconstituted in proteoliposomes and was shown to function as a water channel by stopped-flow spectrophotometry. This study demonstrates that the yeast has the capacity to produce functional aquaporins at levels sufficient for biochemical and biophysical analyses.  相似文献

CRISPR assisted homology directed repair enables the introduction of virtually any modification to the Saccharomyces cerevisiae genome. Of obvious interest is the marker-free and seamless introduction of point mutations. To fulfill this promise, a strategy that effects single nucleotide changes while preventing repeated recognition and cutting by the gRNA/Cas9 complex is needed. We demonstrate a two-step method to introduce point mutations at 17 positions in the S. cerevisiae genome. We show the general applicability of the method, enabling the seamless introduction of single nucleotide changes at any location, including essential genes and non-coding regions. We also show a quantifiable phenotype for a point mutation introduced in gene GSH1. The ease and wide applicability of this general method, combined with the demonstration of its feasibility will enable genome editing at an unprecedented level of detail in yeast and other organisms.  相似文献

Processing of lignocellulosic materials to fuels such as methane and bioethanol may involve several processing steps including pretreatment, saccharification, fermentation, and anaerobic digestion. The amounts of substrate used in these processes are usually based on dry matter content, and the processes themselves typically lead to a change in dry matter content. Thus, it is of great importance to be able to measure dry matter accurately. Dry matter content is commonly determined by measuring loss of water during oven drying. We have used Karl Fischer (KF) titration to measure the water content in a wide range of biomass fractions and have compared these data to results obtained by oven drying. This revealed considerable differences for all tested materials. For lignocellulosic materials, oven drying tends to overestimate dry matter content for untreated material. On the other hand, oven drying generally underestimates dry matter content in pretreated materials due to loss of organic volatiles. These differences have major consequences for the calculation of mass balances and yields in bioprocessing. The KF method gives more accurate water determination than oven drying due to the unique selectivity of the analysis. The method is suitable for the analysis of lignocellulosic biomasses and is particularly useful for determination of water content in pretreated materials, where oven drying usually underestimates the dry matter content due to loss of volatiles.  相似文献

Anaerobic digestion is a mature wastewater treatment technology, with worldwide application. The predominantly applied bioreactor designs, such as the upflow anaerobic sludge blanket and expanded granular sludge bed, are based on the spontaneous formation of granular sludge. Despite the exploitation of granular reactors at full-scale for more than two decades, the mechanisms of granulation are not completely understood and numerous theories have been put forward to describe the process from a biological, ecological and engineering point of view. New technological opportunities are emerging for anaerobic digestion, aided by an improved understanding of microbiological and environmental factors affecting the formation and activity of anaerobic granular sludge.  相似文献
