This study is the first to investigate the effects of different doses of nandrolone decanoate (ND) upon uterine tissue and fertility, and if the reproductive alterations can be restored after cessation of the treatment. Wistar female rats were treated with ND at doses of 1.87, 3.75, 7.5, and 15 mg/kg body weight, diluted in vehicle (n = 30/group), or received only mineral oil (control group, n = 45). The animals were divided into three periods of study: ND‐treated receiving a daily subcutaneous injection for 15 consecutive days (1), and treatment with ND followed by 30‐day recovery (2), and 60‐day recovery (3). At the end of each period, five females per group were induced to death to histopathological analysis and the others were allowed to fertility evaluation (at 19th gestational day). Animals that received ND followed by 30‐day recovery exhibited persistent diestrous and marked suppression of reproductive capacity. Conversely, after 60‐day recovery, only lowest doses females (1.87 and 3.75 mg/kg) exhibited restoration of normal estrous cyclicity. Uterine weights were increased after ND treatment similarly to that of the controls after 60‐day recovery. The ND‐treated groups showed histopathological changes in the endometrium, myometrium, and perimetrium, and an increase in the thickness of both muscular and serous layers. Notably, the recovery of uterine tissue after ND treatment was dose‐ and period‐dependent. Among the skin disorders of dogs, scabies is notable for its high occurrence rate and the need for veterinary interventions. There are two obstacles to making this diagnosis through direct investigation under a microscope (DIM): the continual need to make new slides when the results are negative and the long time needed for reading these slides. Thus, the objective of the present study was to compare efficacy between DIM and the technique of centrifugation-flotation in sucrose solution (CFSS) in samples from dogs in the semiarid region of the state of Paraíba, Brazil. Samples from 136 dogs were used, and three slides were made for each examination (DIM and CFSS). The readings were halted in cases of positivity. Positive samples were obtained from 56.6% of the dogs (77/136), of which 76.6% (59/77) were positive through both techniques, 13% (10/77) only through DIM and 10.4% (8/77) only through CFSS. The positivity rate did not differ statistically between the techniques. CFSS showed higher quality of readings, due to the considerably fewer artifacts on the slides, thereby optimizing the reading time. Sensitivity (85.6%), specificity (88.1%), accuracy (86.8%), positive predictive value (88.1%) and negative predictive value (85.1%) were obtained and the kappa coefficient (0.73) was considered substantial. It was concluded that CFSS showed high diagnostic capacity for scabies, similar to that of DIM, with optimized reading time, fewer artifacts and better display of mites.
This work aims to evaluate the fermentability of cellulosic hydrolysates obtained by enzymatic saccharification of sugarcane
bagasse pretreated by hydrothermal processing using Candida guilliermondii FTI 20037 yeast. The inoculum was obtained from yeast culture in a medium containing glucose as a carbon source supplemented
with rice bran extract, CaCl2·2H2O and (NH4)2SO4 in 50 mL Erlenmeyer flasks, containing 20 mL of medium, initial 5.5 pH under agitation of an orbital shaker (200 rpm) at
30°C for 24 h. The cellulosic hydrolysates, prior to being used as a fermentation medium, were autoclaved for 15 min at 0.5 atm
and supplemented with the same nutrients employed for the inoculum, except the glucose, using the same conditions for the
inoculum, but with a period of 48 h. Preliminary results showed the highest consumption of glucose (97%) for all the hydrolysates,
at 28 h of fermentation. The highest concentration of ethanol (20.5 g/L) was found in the procedure of sugarcane bagasse pretreated
by hydrothermal processing (195°C/10 min in 20 L reactor) and delignificated with NaOH 1.0% (w/v), 100°C, 1 h in 500 mL stainless

The purpose of this study was to assess the intensity of official Futsal matches, expressed in different ways. Fourteen male professional Futsal players from a First Division Brazilian team volunteered to participate in this study. Maximal oxygen uptake (VO(2)max) and the heart hate (HR) and oxygen uptake (VO(2)) correlation were determined for each player. The match intensity was estimated from the players' average HR measured during 13 National Futsal League matches. The HR measurements were obtained while the players were in the court but the values recorded while the players were sitting on the bench were not considered. In addition, these HR values were used to estimate the intensity of the effort expressed as a percentage of the maximal HR (% HRmax), percentage of VO(2)max (% VO(2)max), kilocalories per minute (kcal·min(-1)), and total caloric expenditure. The mean intensity of the matches was 86.4 ± 3.8% HRmax, 79.2 ± 9.0% VO(2)max, 18.0 ± 2.2 kcal·min(-1), and 313 ± 9.3 kcal. It was concluded that official Futsal matches have high intensity when expressed in the different ways used in this study. The information provided by this research can be used for planning the athletes' workouts, diets, and resting periods. 相似文献
The aim of this study was to develop and evaluate a floating multiparticulate gastroretentive system for the modified release
of zidovudine (AZT). AZT was used as a model drug water-soluble at therapeutic doses. The floating gastroretentive system
was obtained by co-precipitation, after solvent diffusion and evaporation. The proposed system was evaluated in vitro for particle morphology, lag time and floating time, loading rate, release profile, and the release kinetic of AZT release.
AZT’s physico-chemical characteristics were evaluated by differential scanning calorimetry (DSC), X-ray diffraction (XDR)
and infrared spectroscopy (IR). The particles obtained were sphere-shaped, hollow, and had porous walls. The floating was
immediate, and floating time was higher than 12 h. The loading rate was 34.0 ± 9.0%. The system obtained had an extended release.
DSC and XDR results showed a modification in AZT’s solid state. IR spectroscopy revealed that the chemical structure of the
AZT was unchanged. The hollow microballoons presented gastroretentive, floating, and extended-release properties. 相似文献
Non-destructive methods of collecting DNA from small fish species can be problematic, as fin clips can potentially affect behaviour or survivorship in the wild. Swabbing body mucus may provide a less invasive method of DNA collection. However, risk of contamination from other individuals in high density groups could give erroneous genotyping results. We compared multilocus microsatellite genotypes from the same individuals when collected at low and high density and compared this with fin clips. We found no differences between these categories, with a genotyping error rate of 0.42%, validating the use of body mucus swabbing for DNA collection in fish. 相似文献

Th1 cytokines are essential for the control of M. tuberculosis infection. The role of IL-10 in tuberculosis is controversial and there is an increasing body of evidence suggesting that the relationship between Th1 cytokines and IL-10 is not as antagonistic as it was first believed, and that these cytokines may complement each other in infectious diseases.
Methods
The present study evaluated the activating capacity of CD4+ and CD8+ T cell repertoire in response to antigen stimulation through the expression of CD69 using Flow Cytometry, as well as the functionality of PBMCs by determining the cytokine profile in patients with active tuberculosis and in clinically cured patients after in vitro stimulation using ELISA. Treated patients were subdivided according to time after clinical cure (<12 months or >12 months post-treatment).
Results
We observed that T cell activation was higher in TB-treated patients, especially CD8+ T cell activation in TB-Treated >1 year. Th1 cytokines were significantly higher in TB-Treated, and the levels of IFN-γ and TNF-α increased continuously after clinical cure. Moreover, IL-10 production was significantly higher in cured patients and it was also enhanced in cured patients over time after treatment. Th17, Th2 and Th22 cytokines showed no statistically significant differences between Healthy Donors, Active-TB and TB-Treated.
Conclusions
This study describes a scenario in which potentiation of CD4+ and CD8+ T cell activation and increased Th1 cytokine production are associated with the clinical cure of tuberculosis in the absence of significant changes in Th2 cytokine production and is accompanied by increased production of IL-10. According to FAO and WHO, probiotics are defined as live microorganisms that, when administered in adequate amounts, confer a health benefit on the host. Most probiotic bacteria used today belong to the genera Lactobacillus and Bifidobacterium and are of animal or human origin. The fundamental characteristic routinely evaluated in potential probiotics strains is their limited viability loss during gastrointestinal transit (GIT), but to date, no studies reported whether probiotics, besides viability, still also maintain their beneficial properties intact. To study this aspect, we considered two strains, Lactobacillus rhamnosus DTA 79 and L. paracasei DTA 83, previously characterised for the presence of some probiotic properties, isolated from faeces of 7- to 21-day-old babies. Here, we examined some additional properties, namely antibiotic resistance, resistance to lysozyme, presence of haemolytic activity and inhibition of pathogen biofilm formation. We then tested the effect of in vitro GIT on all these features and our results show evidence that this procedure had in some cases limited and in others no significant effects on them. Additionally, we examined the gastrointestinal resistance of the strains after skim milk fermentation and successive storage of the product for 20 and 40 days at refrigeration temperature, to see whether prolonged storage could weaken cell resistance to GIT. Our results demonstrate that a protracted refrigeration period before in vitro GIT did not affect or influenced very weakly this essential probiotic property.
