Angiotensin II (Ang II) AT(2) receptors are abundantly expressed in rat fetal tissues where they probably contribute to development. In the present study we examine the effects of Ang II type 2 receptor stimulation on SHP-1 activation. Ang II (10(-7) M) elicits a rapid and transient tyrosine phosphorylation of SHP-1, maximal at 1 min, in a dose-dependent form, blocked by the AT(2) antagonist, PD123319. SHP-1 phosphorylation is followed in time by tyrosine dephosphorylation of different proteins, suggesting a sequence of events. Ang II induces association of SHP-1 to AT(2) receptors as shown by co-immunoprecipitation, Western blot and binding assays. SHP-1 activity was determined in immunocomplexes obtained with either anti-AT(2) or anti-SHP-1 antibodies, after Ang II stimulation (1 min), in correlation with the maximal level of SHP-1 phosphorylation. Interestingly, following receptor stimulation (1 min) c-Src was associated to AT(2) or SHP-1 immunocomplexes. Preincubation with the c-Src inhibitor PP2 inhibited SHP-1 activation and c-Src association, thus confirming the participation of c-Src in this pathway. We demonstrated here for the first time the involvement of c-Src in SHP-1 activation via AT(2) receptors present in an ex vivo model expressing both receptor subtypes. In this model, AT(2) receptors are not constitutively associated to SHP-1 and SHP-1 is not constitutively activated. Thus, we clearly establish that SHP-1 activation, mediated by the AT(2) subtype, involves c-Src and precedes protein tyrosine dephosphorylation, in rat fetal membranes.  相似文献

Question: As it has been found that stress promotes positive interactions mediated by physical amelioration of the environment, is it possible that interactions may turn positive with increasing chronic anthropogenic disturbance (CAD) intensity? Also, is it possible that species that do not tolerate disturbance may require environmental amelioration by their neighbours in disturbed areas, whereas tolerant species may not? Location: The semi‐arid grassland in Concepción Buenavista, Oaxaca, southern Mexico. Methods: We assessed interaction intensity and importance through a neighbour removal experiment along a CAD gradient for three species differing in disturbance tolerance. Water potential was monitored on vegetated and bare soil. Results: A shift from competitive effects in low CAD sites to positive interactions in degraded sites was found. The disturbance‐tolerant species did not respond to CAD, whereas the less tolerant species changed its interactions drastically in terms of growth and reproduction. The species with medium tolerance had an intermediate response. Neighbours promoted germination in all species. Vegetation removal reduced soil humidity. Conclusions: Positive interactions seemingly resulted from the amelioration of the abiotic stresses induced by vegetation removal. The dependence on neighbours to germinate, grow, or reproduce suggests that if CAD eliminates the plant cover, vegetation will hardly recover. Irreversible changes are known to occur in communities where positive interactions predominate, but CAD may set the conditions for irreversible shifts even in communities where interactions are normally competitive.  相似文献

Perturbations in the normal sequence of ventricular activation can create regions of early and late activation, leading to dysynchronous contraction and areas of dyskinesis. Dyskinesis occurs across the left ventricular (LV) wall, and its presence may have important consequences on cardiac structure and function in normal and failing hearts. Acutely, dyskinesis can trigger inflammation and, in the long term (6 wk and above), leads to LV remodeling. The mechanisms that trigger these changes are unknown. To gain further insight, we used a canine model to evaluate transumural changes in myocardial function and inflammation induced by epicardial LV pacing. The results indicate that 4 h of LV suprathreshold pacing resulted in a 30% local loss of endocardial thickening. Assessment of neutrophil infiltration showed a significant approximately fivefold increase in myeloperoxidase activity in the epicardium versus the midwall/endocardium. Matrix metalloproteinase-9 activity increased ~2 fold in the epicardium and ROS generation increased ~2.5-fold compared with the midwall/endocardium. To determine the effects that electrical current alone has on these end points, a group of animals was subjected to subthreshold pacing. Significant increases were observed only in epicardial myeloperoxidase levels. Thus, the results indicate that transmural dyskinesis induced by suprathreshold epicardial LV activation triggers a localized epicardial inflammatory response, whereas subthreshold stimulation appears to solely induce the trapping of leucocytes. Suprathreshold pacing also induces a loss of endocardial function. These results may have important implications as to the nature of the mechanisms that trigger the inflammatory response and possibly long-term remodeling in the setting of dysynchrony.  相似文献

Background

Bacterial interactions with the environment- and/or host largely depend on the bacterial glycome. The specificities of a bacterial glycome are largely determined by glycosyltransferases (GTs), the enzymes involved in transferring sugar moieties from an activated donor to a specific substrate. Of these GTs their coding regions, but mainly also their substrate specificity are still largely unannotated as most sequence-based annotation flows suffer from the lack of characterized sequence motifs that can aid in the prediction of the substrate specificity.

Results

In this work, we developed an analysis flow that uses sequence-based strategies to predict novel GTs, but also exploits a network-based approach to infer the putative substrate classes of these predicted GTs. Our analysis flow was benchmarked with the well-documented GT-repertoire of Campylobacter jejuni NCTC 11168 and applied to the probiotic model Lactobacillus rhamnosus GG to expand our insights in the glycosylation potential of this bacterium. In L. rhamnosus GG we could predict 48 GTs of which eight were not previously reported. For at least 20 of these GTs a substrate relation was inferred.

Conclusions

We confirmed through experimental validation our prediction of WelI acting upstream of WelE in the biosynthesis of exopolysaccharides. We further hypothesize to have identified in L. rhamnosus GG the yet undiscovered genes involved in the biosynthesis of glucose-rich glycans and novel GTs involved in the glycosylation of proteins. Interestingly, we also predict GTs with well-known functions in peptidoglycan synthesis to also play a role in protein glycosylation.

Objective

This study compared the performance of 12 brands of cartomizer style electronic cigarettes (EC) using different puffing protocols and measured the concentrations of nicotine in each product.

Methods

Air flow rate, pressure drop, and aerosol absorbance were measured using two different protocols, first 10 puffs and a modified smoke-out protocol.

Results

First 10 puff protocol: The air flow rate required to produce aerosol ranged between brands from 4–21 mL/s. Pressure drop was relatively stable within a brand but ranged between brands from 14–71 mmH2O and was much lower than the earlier classic 3-piece models. Absorbance, a measure of aerosol density, was relatively consistent between puffs, but varied between brands. With the modified smoke-out protocol, most brands were puffed until 300 puffs. The pressure drop was relatively stable for all brands except three. Absorbance of the aerosol decreased as the number of puffs increased. Although there was some uniformity in performance within some brands, there was large variation between brands. The labeled and measured nicotine concentrations were within 10% of each other in only 1 out of 10 brands.

Conclusions

Over 10 puffs, the cartomizers all perform similarly within a brand but varied between brands. In smoke-out trials, most brands lasted at least 300 puffs, and performed similarly within brands with respect to pressure drop and absorbance. For five brands, products purchased at different times performed differently. These data show some improvement in performance during evolution of these products, but nevertheless indicate problems with quality control in manufacture.  相似文献

To examine the role of the renal vascular receptor in the control of renin secretion in the rat, a denervated, nonfiltering kidney model (DNFK) was developed. The left kidney was subjected to a 2-hr period of total renal ischemia followed by ureteral ligation and section Denervation was accomplished by stripping all visible nerves and painting the renal vessels with 5% phenol. Forty-eight hours later lissamine green dye was injected iv and failed to appear in either the cortical or medullary tubules, indicating that glomerular filtration had ceased. Histological study of these kidneys revealed diffuse tubular necrosis with extensive intratubular cast formation. Norepinephrine content of the DNFK was reduced 91% compared to the contralateral normal kidney (P less than 0.001). In another group of anesthetized rats with a single DNFK, 15 min of suprarenal aortic constriction (SAC) increased plasma renin activity (PRA) from 3.4 +/- 0.6 to 11.5 +/- 1.6 ng AI/ml/hr; in a time control series, PRA was unchanged. To exclude the influence of adrenal catecholamines in this response, bilateral adrenalectomy was performed in a separate group of animals with a DNFK. In this series, SAC also markedly increased PRA. The present data indicate that in the rat the macula densa, the renal nerves, and adrenal catecholamines were not essential for the hyperreninemia induced by a reduction in renal perfusion pressure.  相似文献
