Each plant genome contains a repertoire of β-mannanase genes belonging to glycoside hydrolase family 5 subfamily 7 (GH5_7), putatively involved in the degradation and modification of various plant mannan polysaccharides, but very few have been characterized at the gene product level. The current study presents recombinant production and in vitro characterization of AtMan5-1 as a first step towards the exploration of the catalytic capacity of Arabidopsis thaliana β-mannanase. The target enzyme was expressed in both E. coli (AtMan5-1e) and P. pastoris (AtMan5-1p). The main difference between the two forms was a higher observed thermal stability for AtMan5-1p, presumably due to glycosylation of that particular variant. AtMan5-1 displayed optimal activity at pH 5 and 35 °C and hydrolyzed polymeric carob galactomannan, konjac glucomannan, and spruce galactoglucomannan as well as oligomeric mannopentaose and mannohexaose. However, the galactose-rich and highly branched guar gum was not as efficiently degraded. AtMan5-1 activity was enhanced by Co2+ and inhibited by Mn2+. The catalytic efficiency values for carob galactomannan were 426.8 and 368.1 min?1 mg?1 mL for AtMan5-1e and AtMan5-1p, respectively. Product analysis of AtMan5-1p suggested that at least five substrate-binding sites were required for manno-oligosaccharide hydrolysis, and that the enzyme also can act as a transglycosylase. 相似文献

Background
The Gram negative anaerobic bacterium Porphyromonas gingivalis has long been recognized as a causative agent of periodontitis. Periodontitis is a chronic infectious disease of the tooth supporting tissues eventually leading to tooth-loss. Capsular polysaccharide (CPS) of P. gingivalis has been shown to be an important virulence determinant. Seven capsular serotypes have been described. Here, we used micro-array based comparative genomic hybridization analysis (CGH) to analyze a representative of each of the capsular serotypes and a non-encapsulated strain against the highly virulent and sequenced W83 strain. We report on the identification, molecular cloning, and characterization of
an alpha1,3 fucosyltransferase (alpha1,3FT) expressed by the nematode,
Caenorhabditis elegans . Although C. elegans glycoconjugates do not express
the Lewis x antigen Galbeta1-- >4[Fucalpha1-->3]GlcNAcbeta-->R,
detergent extracts of adult C.elegans contain an alpha1,3FT that can
fucosylate both nonsialylated and sialylated acceptor glycans to generate
the Lexand sialyl Lexantigens, as well as the lacdiNAc-containing acceptor
GalNAcbeta1-->4GlcNAcbeta1-- >R to generate GalNAcbeta1-->4
[Fucalpha1-->3]GlcNAcbeta1-->R. A search of the C.elegans genome
database revealed the existence of a gene with 20-23% overall identity to
all five cloned human alpha1,3FTs. The putative cDNA for the C.elegans
alpha1,3FT (CEFT-1) was amplified by PCR from a cDNA lambdaZAP library,
cloned, and sequenced. COS7 cells transiently transfected with cDNA
encoding CEFT-1 express the Lex, but not sLexantigen. The CEFT-1 in the
transfected cell extracts can synthesize Lex, but not sialyl Lex, using
exogenous acceptors. A second fucosyltransferase activity was detected in
extracts of C. elegans that transfers Fuc in alpha1,2 linkage to Gal
specifically on type-1 chains. The discovery of alpha-fucosyltransferases
in C. elegans opens the possibility of using this well-characterized
nematode as a model system for studying the role of fucosylated glycans in
the development and survival of C.elegans and possibly other helminths.

The present article examines whether the expression of certain surface molecules that trigger immune responses shows a circadian rhythm. We also analyzed the rhythms in the number and percentage of lymphocyte subpopulations, in the leukocyte differential counts, and in the total red and white blood cell counts. Blood samples obtained from rats at 2-h intervals for 24 h were stained with several mouse monoclonal antibodies directed against lymphocyte surface molecules and processed by flow cytometry. The number of B, total T, Tgammadelta, Th, and Ts/c cells followed a 24-h rhythm with a peak in the first half of the resting period. The expression of CD45, CD5, CD3, and CD4 followed a circadian rhythm. Their acrophases suggested temporal association between CD45 and CD5 at the end of the active phase and between CD4 and CD3 at the beginning of this phase. This temporal organization could have an important role for immune cell function. 相似文献

Comparison of the effects of concentration, pH and anion species on astringency and sourness of organic acids 总被引:3,自引:1,他引:3
The separate effects of concentration, pH and anion species on intensity of
sourness and astringency of organic acids were evaluated. Judges rated
sourness and astringency intensity of lactic, malic, tartaric and citric
acid solutions at three levels of constant pH varying in normality and at
three levels of constant concentration varying in pH. To assess the
comparative sourness and astringency of the organic acid anions of study,
binary acid solutions matched in pH and titratable acidity were also rated.
As pH was decreased in equinormal solutions, both sourness and astringency
increased significantly (P < 0.001). By contrast, as the normality of
the equi-pH solutions was increased, only sourness demonstrated significant
increases (P < 0.001) while astringency remained constant or decreased
slightly. At the lowest normality tested, all solutions were more
astringent than sour (P < 0.05). Although lactic acid was found to be
significantly more sour than citric acid (P < 0.05), no other sourness
or astringency differences among the organic acid anions were noted. This
study demonstrates for the first time that astringency elicited by acids is
a function of pH and not concentration or anion species, and confirms that
sourness is independently influenced by concentration, pH and anion species
of the acid.

In previous experiments, we found that rats raised in constant light (LL) manifested a more robust circadian rhythm of motor activity in LL and showed longer phase shifts after a light pulse in constant darkness (DD) than those raised under constant darkness. In addition, we observed that the effects produced by constant light differed depending on the time of postnatal development in which it was given. These results suggest that both sensitivity to light and the functioning of the circadian pacemaker of the rat could be affected by the environmental conditions experienced during postembryonic development. Thus, the present experiment aimed to study whether postnatal exposure to light could also affect the circadian system of the mouse. Three groups of mice were formed: One group was raised under constant darkness during lactation (DD group), the second under constant light (LL group), and the third under light-dark cycles (LD group). After lactation, the three groups were submitted first to constant light of high intensity, then to LD cycles, and finally to constant darkness. In the DD stage, a light pulse was given. Finally, mice were submitted to constant light of low intensity. We observed that the circadian rhythm of the DD group was more disturbed under constant light than the rhythm of the LL group, and that, when light intensity increased, the period of the rhythm of the DD group lengthened more than that of the LL group. No significant differences among the groups were found in the phase shift induced by the light pulse. Therefore, it appears that DD mice are more sensitive to light than their LL counterparts. However, at present there is no evidence to affirm that the light environment experienced by the mouse during postnatal development affects the circadian pacemaker. Bacterial colony morphology can reflect different physiological stages such as virulence or biofilm formation. In this work we used transposon mutagenesis to identify genes that alter colony morphology and cause differential Congo Red (CR) and Brilliant Blue G (BBG) binding in Shewanella algae, a marine indigenous bacterium and occasional human pathogen. Microscopic analysis of colonies formed by the wild-type strain S. algae CECT 5071 and three transposon integration mutants representing the diversity of colony morphotypes showed production of biofilm extracellular polymeric substances (EPS) and distinctive morphological alterations. Electrophoretic and chemical analyses of extracted EPS showed differential patterns between strains, although the targets of CR and BBG binding remain to be identified. Galactose and galactosamine were the preponderant sugars in the colony biofilm EPS of S. algae. Surface-associated biofilm formation of transposon integration mutants was not directly correlated with a distinct colony morphotype. The hybrid sensor histidine kinase BarA abrogated surface-associated biofilm formation. Ectopic expression of the kinase and mutants in the phosphorelay cascade partially recovered biofilm formation. Altogether, this work provides the basic analysis to subsequently address the complex and intertwined networks regulating colony morphology and biofilm formation in this poorly understood species. 相似文献

Vibrational communication is important for successful mating in various stink bugs species. The vibrational signals from males and females of Dichelops melacanthus Dallas (Hemiptera: Pentatomidae) are recorded from a nonresonant substrate (i.e. a loudspeaker membrane) to characterize the temporal and spectral properties of these vibrational signals, as well as on a resonant substrate (i.e. bean plants) to obtain information about how these signals are altered when they are transmitted through the plants. On the loudspeaker membrane, D. melacanthus males and females emit only one male or one female song, respectively. However, when the insects are placed on bean leaves, a more complex repertoire is recorded, with three different songs for each sex. The first female and male songs appear to have calling functions and the third male and female songs are emitted during courtship. The second female and male songs are emitted after the first song, although their functions in mating behaviour are not clear. The identified repertoire is similar to those of other Neotropical stink bugs, starting with songs 1 and 2 and developing into song 3. Frequency modulation is observed in the female songs recorded from the loudspeaker membrane and the plants. The signals recorded from plants present higher harmonic peaks compared with the signals recorded from the loudspeaker membrane. The presence of species and sex‐specific songs during mating confirms the important role of vibrational communication in mate location and recognition. The temporal and spectral characteristic signals are influenced by the substrate used to record the songs emitted by D. melacanthus. 相似文献

A polyclonal antibody against the allatostatin BLAST-3 (AGSDGRLYSFGL-NH2) of the cockroach Blattella germanica (L.) (Dictyoptera, Blattellidae) has been raised and characterized, and an ELISA (enzyme-linked immunosorbent assay) for allatostatin quantification has been developed. Allatostatin contents in brain, midgut and haemolymph have been measured in females of B. germanica during the first gonadotropic cycle. Brain allatostatin content increases steadily from adult emergence to the formation of the first ootheca. The values range from 2 ng/brain on the day of adult emergence to 25 ng/brain when the insect forms the ootheca 8 days later. In the midgut, the pattern is similar but the values are about half those of the brain. Allatostatin concentrations in the haemolymph after HPLC separation are in the nanomolar range. The occurrence of allatostatins in the haemolymph suggests that these peptides can act through a humoral pathway, as well as via nerves. The allatostatin content of both brain and midgut are high while the female is transporting the ootheca, which suggests that these peptides could be related to the low metabolic status characterising the period of oothecal transport. 相似文献
