Molecular analysis of the heterodimeric T-cell antigen receptor of insulin-specific class II-restricted T-cell hybridomas (THys) derived from C57BL/6 (B6) wild-type and B6.C-H-2bm12 (bm12) mutant mice revealed that such T cells use a diverse V gene repertoire. Analysis of three THys that use related V genes, however, showed a number of novel features. Two THys that share major histocompatibility complex restriction use V alpha genes that are 98.6% homologous. Two THys sharing the same antigen fine specificity use a particular germ line V beta D beta J beta combination. A 21-base-pair deletion in the 5' segment of the J beta gene occurs in one THy, suggesting a novel mechanism for generating diversity in T-cell antigen receptor beta genes. The first amino acid encoded by N sequences at the V-D junction is conserved in a pair of T cells which recognize identical antigenic epitopes. The implications of these findings for the structural mechanisms underlying major histocompatibility complex-restricted antigen-specific T-cell recognition are discussed. 相似文献

The wind dispersal and survival of the first instars of a native polyphagous geometrid, Ectropis excursaria (Guenée) on Citrus sinensis (L.), Cupressus macrocarpa Hartw., Eucalyptus camaldulensis Dehn., Medicago sativa (L.) and Pinus radiata D. Don., were studied. A positive correlation existed between the percentage of caterpillars remaining on the foliage in dispersal experiments and the percent survival of caterpillars on these host-plants. The results are discussed in relation to the host-selection strategy of the first instars and the advantages of dispersal are compared for adult females and young larvae.
Résumé Les femelles d'E. excursaria ne pondent pas là où les chenilles de premier stade s'alimentent. La dispersion par le vent, a été examinée comme l'un des éléments de la sélection de la plante-hôte par les chenilles du premier stade. Cette dispersion était influencée par la nature du végétal et par l'âge du feuillage. La dispersion à partir de 3 plantes de valeurs alimentaires différentes n'a pas été modifiée par le jeûne du premier stade. Le taux de survie, sur feuilles coupées et sur plantes en pot, a été aussi influencé par la nature du végétal. Il y a eu une corrélation positive significative entre le pourcentage de chenilles de premier stade restant sur le feuillage de l'hôte lors d'expériences de dispersion et le taux de survie sur ce feuillage. On peut suggérer que le comportement alimentaire polyphage des chenilles et la fécondité élevée des femelles peuvent avoir contribué à la stratégie de dispersion larvaire, comme processus de sélection larvaire, au détriment d'une sélection du lieu de ponte par les femelles.相似文献

Persistent activation of steroidogenesis in adrenocortical cells by photoaffinity labeling of corticotropin receptors 总被引:2,自引:0,他引:2
Photolysis of rat adrenocortical cells in the presence of the photoreactive derivative [(2-nitro-5-azidophenylsulfenyl)Trp9]-adrenocorticotropic hormone (2,5-NAPS-ACTH) at 24 degrees C resulted in persistent activation of corticosterone production. The basal rate of steroidogenesis became maximal when photolysis was performed at 24 degrees C but remained the same as that of control cells when irradiation was performed at 0 degrees C. No increase in basal rate was observed with dark controls or cells photolyzed with [(2,4-dinitrophenylsulfenyl)Trp9]ACTH, a photoresistant analog of the hormone. Prephotolyzed 2,5-NAPS-ACTH failed to induce persistent activation. Both ACTH and 2,4-(dinitrophenylsulfenyl)Trp9-ACTH blocked the photo-induced activation of steroidogenesis elicited by 2,5-NAPS-ACTH. Under photolysis conditions which caused the basal rate of steroidogenesis to become maximal, a 3-fold increase in the basal rate of cAMP formation was observed. 相似文献

The single tryptophan residue in the pituitary hormone adrenocorticotropin was modified selectively by reaction with a variety of substituted o-nitrophenylsulfenyl chlorides. In addition to quantitative modification of the tryptophan residue, the reaction invariably resulted in partial oxidation of the methionine residue to the sulfoxide. The methionine sulfoxide derivative could be separated from the desired product by partition chromatography on Sephadex G-50 in the solvent system 1-butanol-pyridine-0.1% acetic acid (5:3:11). Thus, the 2,4-dinitrophenylsulfenyl, 2-nitro-4-carboxyphenylsulfenyl, and 2-nitro-4-carbamidophenylsulfenyl derivatives of adrenocorticotropin were prepared and characterized. Modifications in the isolation of adrenocorticotropin from ovine pituitaries are also described. The melanocyte stimulating activities of the native hormone and the analogues are discussed. 相似文献

Variation of the NH-C alpha-H coupling constant with dihedral angle in the NMR spectra of peptides 总被引:6,自引:0,他引:6
Proton magnetic resonance data and conformational calculations of a series of model compounds containing a NH-CαH group substituted as in peptides have been used to generate a proton–proton coupling constant–dihedral angle relation for the peptide unit. For those substances used in which the dihedral angle about the N-Cα bond is not fixed, the angle distribution was calculated from conformational theory. Using eight examples in which the number of theoretical assumptions were least, the best values of the coefficients A, B, and C in the expression J(θ) = Acos2θ + B cosθ + Csin2θ were found by a least-squares procedure to be 7.9, ?1.55, and 1.35, respectively. This relation gives reasonable values for the dihedral angles ? in cyclic oligopeptide structures for which the availability of both NMR data and other structural information allow comparison. When applied to N-acetylamino acid N-methylamides having side chains extending beyond Cβ, however, agreement with the calculated conformational distribution was found for Leu, Met, and Trp, but observed values of J were larger than expected for Val, He, Phe, and Tyr, These disagreements are considered to be the result of interactions not yet taken into account in the usual conformational calculations. 相似文献
