Urease is present in considerable quantity in the cotyledonsof Citrullus, though elsewhere in the plant it is present onlyin traces or is absent; urease activity in the cotyledons changesduring growth, showing an initial rise followed by an abruptdrop almost to zero. These changes, under a wide variety ofconditions, are not correlated with those in the major nitrogenfractions; they are, however, closely correlated with cell extensionand the associated changes in water content and respiration.A connexion with chlorophyll formation is possible but unlikely.It is suggested that the changes in cotyledonary urease constitutemerely one aspect of the protoplasmic differentiationthat takes place as a cell matures. 相似文献

We present here characterization data for seven new microsatellite markers designed from new microsatellite loci isolated from a microsatellite‐enriched DNA library from Vitis vinifera. The observed heterozygosity varied from 0.73 up to 0.93 and the number of alleles per locus ranged from 12 to 26. This high polymorphism makes these new markers interesting for use in genotyping studies and completing the set of microsatellite markers already available for V. vinifera. Additionally these seven new markers appear to be conserved in four other Vitis species and 15 Vitis hybrids used as rootstocks for V. vinifera cultivation. 相似文献

The Parvidrilidae Erséus, 1999 constitute the most recently described family of oligochaete microdriles. Prior to this study, Parvidrilus strayeri Erséus, 1999, and Parvidrilus spelaeus Martínez‐Ansemil, Sambugar & Giani, 2002, found in groundwaters of the USA (Alabama) and Europe (Slovenia and Italy), respectively, were the only two species in this family. In this paper, six new species – Parvidrilus camachoi , Parvidrilus gianii , Parvidrilus jugeti , Parvidrilus meyssonnieri , Parvidrilus stochi , and Parvidrilus tomasini – and Parvidrilus gineti (Juget, 1959) comb. nov. are added to the family. With all species being stygobiont, the Parvidrilidae is unique in being the only family of oligochaetes worldwide comprising taxa that are restricted to groundwater habitats. Parvidrilids are exceedingly small worms whose principal morphological characteristics are the presence of hair setae in ventral bundles, the markedly posterior position of setae within the segments, the presence of mid‐dorsal glandular pouches in mesosomial segments, the lateral development of the clitellum, the presence of a single male pore in segment XII, and the presence (or absence) of a single spermatheca. The phylogenetic relationships of the Parvidrilidae within the Clitellata were investigated using the nuclear 18S rRNA gene, and the most representative and taxonomically balanced data set of clitellate families available to date. The data were analysed by parsimony, maximum likelihood, and Bayesian inference. Irrespective of the method used, Parvidrilidae were placed far from Capilloventridae, one family once suggested to be closely related to parvidrilids. Although closer to Enchytraeidae than Phreodrilidae, two other suggested putative sister families, the exact position of Parvidrilidae within Clitellata still remained uncertain in the absence of branch support. The examination of reproductive structures, together with the similarity of other important anatomical traits of the new species herein described, reinforced the idea that phreodrilids were the best candidate to be the sister group to parvidrilids on morphological grounds. A fragment of the mitochondrial cytochrome oxidase I gene, used as a barcode, also genetically characterized a few Parvidrilus species. The observation that two species diverge from each other by high genetic distances, even though their type localities are more or less only 100 km apart, is interpreted in the context of low dispersal abilities of inhabitants of the subterranean aquatic ecosystem, and habitat heterogeneity. A key goal for the perceptual system is to optimally combine information from all the senses that may be available in order to develop the most accurate and unified picture possible of the outside world. The contemporary theoretical framework of ideal observer maximum likelihood integration (MLI) has been highly successful in modelling how the human brain combines information from a variety of different sensory modalities. However, in various recent experiments involving multisensory stimuli of uncertain correspondence, MLI breaks down as a successful model of sensory combination. Within the paradigm of direct stimulus estimation, perceptual models which use Bayesian inference to resolve correspondence have recently been shown to generalize successfully to these cases where MLI fails. This approach has been known variously as model inference, causal inference or structure inference. In this paper, we examine causal uncertainty in another important class of multi-sensory perception paradigm – that of oddity detection and demonstrate how a Bayesian ideal observer also treats oddity detection as a structure inference problem. We validate this approach by showing that it provides an intuitive and quantitative explanation of an important pair of multi-sensory oddity detection experiments – involving cues across and within modalities – for which MLI previously failed dramatically, allowing a novel unifying treatment of within and cross modal multisensory perception. Our successful application of structure inference models to the new ‘oddity detection’ paradigm, and the resultant unified explanation of across and within modality cases provide further evidence to suggest that structure inference may be a commonly evolved principle for combining perceptual information in the brain. 相似文献

Fiber mats with average fiber diameter ranging between 80 and 250 nm of polyvinyl alcohol (PVA)/water solution having a concentration of 4 wt.% have been prepared by electrospinning method. The influence of applied voltage, flow rate, and needle-to-collector distance on the fiber morphology and diameters has been studied. Scanning electron microscopy and atomic force microscopy are used to characterize the fibers. It has been observed that bead-free fibers of 4 wt.% PVA can be obtained at lower voltages (9 kV). Also, density and the deposition area of the fiber mats showed a clear dependence on the applied voltage, flow rate, and collector distance. 相似文献

The influence of preexisting immunity to viral vectors is a major issue for the development of viral-vectored vaccines. In this study, we investigate the effect of preexisting vaccinia virus immunity on the immunogenicity and efficacy of a DNA/modified vaccinia Ankara (MVA) SIV vaccine in rhesus macaques using a pathogenic intrarectal SIV251 challenge. Preexisting immunity decreased SIV-specific CD8 and CD4 T cell responses but preserved the SIV-specific humoral immunity. In addition, preexisting immunity did not diminish the control of an SIV challenge mediated by the DNA/MVA vaccine. The peak and set point viremia was 150- and 17-fold lower, respectively, in preimmune animals compared with those of control animals. The peak and set point viremia correlated directly with colorectal virus at 2 wk postchallenge suggesting that early control of virus replication at the site of viral challenge was critical for viral control. Factors that correlated with early colorectal viral control included 1) the presence of anti-SIV IgA in rectal secretions, 2) high-avidity binding Ab for the native form of Env, and 3) low magnitude of vaccine-elicited SIV-specific CD4 T cells displaying the CCR5 viral coreceptor. The frequency of SIV-specific CD8 T cells in blood and colorectal tissue at 2 wk postchallenge did not correlate with early colorectal viral control. These results suggest that preexisting vaccinia virus immunity may not limit the potential of recombinant MVA vaccines to elicit humoral immunity and highlight the importance of immunodeficiency virus vaccines achieving early control at the mucosal sites of challenge. 相似文献

A survey of 322 proteins showed that the short polar (SP) side chains of four residues, Thr, Ser, Asp, and Asn, have a very strong tendency to form hydrogen bonds with neighboring backbone amides. Specifically, 32% of Thr, 29% of Ser, 26% of Asp, and 19% of Asn engage in such hydrogen bonds. When an SP residue caps the N terminal of a helix, the contribution to helix stability by a hydrogen bond with the amide of the N3 or N2 residue is well established. When an SP residue is in the middle of a helix, the side chain is unlikely to form hydrogen bonds with neighboring backbone amides for steric and geometric reasons. In essence the SP side chain competes with the backbone carbonyl for the same hydrogen-bonding partner (i.e., the backbone amide) and thus SP residues tend to break backbone carbonyl-amide hydrogen bonds. The proposition that this is the origin for the low propensities of SP residues in the middle of alpha helices (relative to those of nonpolar residues) was tested. The combined effects of restricting side-chain rotamer conformations (documented by Creamer and Rose, Proc Acad Sci USA, 1992;89:5937-5941; Proteins, 1994;19:85-97) and excluding side- chain to backbone hydrogen bonds by the helix were quantitatively analyzed. These were found to correlate strongly with four experimentally determined scales of helix-forming propensities. The correlation coefficients ranged from 0.72 to 0.87, which are comparable to those found for nonpolar residues (for which only the loss of side-chain conformational entropy needs to be considered). 相似文献
