The product of the cloned recA+ gene of Proteus mirabilis substitutes for a defective recA protein in Escherichia coli recA- mutants and restores recombination, repair, and prophage induction functions to near normal levels (Eitner, G., Adler, B., Lanzov, V. A., and Hofemeister, J. (1982) Mol. Gen. Genet. 185, 481-486). In this paper, we report the purification to near homogeneity of the P. mirabilis recA protein (recApm). The polypeptide has a molecular weight similar to that of E. coli recA protein (recAec) and shows partial identity with recAec when reacted against antibodies specific for the E. coli recA protein. recApm catalyzes the hydrolysis of ATP in the presence of single-stranded but not double-stranded DNA. We have compared the recombination-like activities of recApm with those of recAec and found them to be similar. In the presence of ATP and Mg2+, stoichiometric amounts of recApm promote the complete reciprocal exchange of strands between gapped circular and linear duplex DNA molecules. The enzyme also efficiently promotes the formation of D-loops from circular duplex DNA and homologous single-stranded fragments. However, although recApm and recAec share the above physical and functional similarities, they differ in their ability to interact with the E. coli single strand binding protein to catalyze the transfer of one DNA strand from a linear duplex to a single-stranded circle. 相似文献

We measured the distribution of molecular forms of acetylcholinesterase (AChE) in muscles of a song bird, the zebra finch, and found a pattern similar to those reported in other vertebrates. As in other species, the most rapidly sedimenting form of the enzyme decreases to barely detectable levels following denervation. In the muscles of the syrinx, castration causes a large decrease in AChE activity, but has little or no effect on the relative abundance of AChE forms. This suggests that the number of AChE catalytic sites is changing without affecting the distribution of catalytic sites among the molecular forms. This is in marked contrast with the effect of denervation in the syrinx, which causes changes in the distribution of activity, as well as in total activity. 相似文献

Six geographical isolates of Rhodochorton purpureum (Lightfoot) Rosenvinge (Rhodophyta, Nemalionales) formed tetrasporangia only in short days at 10°C. For most isolates, the critical day-length increased with latitude of origin from 9.5 h for an isolate from California to 14.5 h for one from Antarctica. Tetrasporangium production could be induced by 9–15 short-day cycles followed by a further 22–28 cycles in long days. A night-break consisting of 1 h of white light in the middle of a 16-h dark period inhibited the short-day response of isolates from low latitudes, but not those from higher latitudes. When a similar night-break was given in the middle of a 14-h dark period, however, the response of all isolates was at least partially inhibited. Night-breaks given at any time in the central 7 h of a 14-h dark period were equally inhibitory. Broad-band red light (0.3–0.4 mmol m-2), given as a night-break, caused 50% inhibition of the short-day response. At a slightly higher photon exposure (0.6 mmol m-2, given as 1 μmol m-2 s-1 for 10 min), narrow-band red (662 nm) and blue (448 nm) light caused similar inhibition, but green (547 nm) and far-red (731 nm) were ineffective as night-breaks. The inhibitory effect of a 10-min night-break with red light could not be reversed by subsequent exposure to an equal photon exposure of far-red light. These results add to the existing evidence that the pigments mediating photoperiodic responses among algae are more varied than those among flowering plants. 相似文献

Using cell permeabilization, a technique which allows addition of exogenously supplied radiolabeled sugar nucleotides to serve as direct glycosyl donors, oligosaccharide biosynthesis was examined in fibroblasts obtained from normal and cystic fibrosis (CF) subjects. Incubation of logarithmically growing cells with either radiolabeled leucine or xylose has indicated that there was a difference in the synthetic rate between the cell types. Protein synthesis in normal cells made permeable with 50 m?g/ml lysolecithin (LL) was demonstrated to be absent, and could not be induced to take place by adding exogenous components, including energy sources and amino acids, normally required for protein synthesis. Thus radiolabeled sugars were being added to peptide acceptors which were already present at the time of LL addition. Both permeable and intact fibroblasts were exposed to labeled UDP-xylose, UDP-galactose, and UDP-glucuronic acid, all donors of mucopolysaccharide precursors. The uptake of xylose into protein was the same for both normal and CF cells, but permeable CF fibroblasts incorporated statistically greater amounts of sugar from UDP-galactose and UDP-glucuronic acid. Intact CF cells were also labeled using these two sugar nucleotides. Trypan blue exclusion indicated CF and normal fibroblasts were equally intact. This and the fact that preincubation of CF cells with the appropriate cold sugar nucleotide eliminated the differences in incorporation between the normal and CF cells suggested that CF fibroblasts had more cell surface acceptor than the normal cells. 相似文献

Summary Kinetic studies of the uptake of hydroquinone--D-glucoside (arbutin) by excised roots of barley demonstrated that this compound is actively transported. Similar studies on the uptake of hydroquinone indicated that the latter compound enters the root tissues by diffusion. A concentration gradient favouring diffusion is maintained for at least three hours by the conversion of the aglucone to its correspondings glucoside. A radioimmunoassay for human plasma corticosterone has been developed. Antisera were obtained by immunizing rabbits with corticosterone-21-hemisuccinate-BSA. An antiserum titer of 1:4000 was used for standard curves ranging from 0–1000 pg. Interfering steroids were removed from plasma extracts by paper chromatography. Plasma blanks obtained from adrenalectomized or Addisonian patients ranged from 29 to 42 ng/dl. Recovery of radioactive corticosterone through the entire method was 67.6 ± 5.2%. The coefficient of variation within assays was 19% and between assays 13%. The average 8 a.m. value in males was 396 ± 228 ng/dl and in females it was 655 ± 271 ng/dl. Corticosterone was found to be secreted episodically, in parallel with cortisol. Secretion of this steroid was suppressed by dexamethasone and stimulated by ACTH infusion. 相似文献

Concentrated whole extracts of the immature fruit of carob and 3 fractions derived from this extract have been shown to inhibit the gibberellic acid induced growth of pea seedlings. The inhibition can be completely reversed by increasing the amount of gibberellic acid. The inhibitors do not reduce the endogenous growth of seedlings but only that induced by gibberellic acid.
