Several approaches for the inactivation of bacteriophage lambda, including UV germicidal irradiation (UVGI) and the chemical agents Virkon-S, Chloros, Decon-90, and sodium hydroxide (NaOH), were compared. PlsY is the essential first step in membrane phospholipid synthesis of Gram-positive pathogens. PlsY catalyzes the transfer of the fatty acid from acyl-phosphate to the 1-position of glycerol-3-phosphate to form the first intermediate in membrane biogenesis. A series of non-metabolizable, acyl-sulfamate analogs of the acyl-phosphate PlsY substrate were prepared and evaluated as inhibitors of Staphylococcus aureus PlsY and for their Gram-positive antibacterial activities. From this series phenyl (8-phenyloctanoyl) sulfamate had the best overall profile, selectively inhibiting S. aureus phospholipid biosynthesis and causing the accumulation of both long-chain fatty acids and acyl-acyl carrier protein intermediates demonstrating that PlsY was the primary cellular target. Bacillus anthracis was unique in being more potently inhibited by long chain acyl-sulfamates than other bacterial species. However, it is shown that Bacillus anthracis PlsY is not more sensitive to the acyl-sulfamates than S. aureus PlsY. Metabolic profiling showed that B. anthracis growth inhibition by the acyl-sulfamates was not specific for lipid synthesis illustrating that the amphipathic acyl-sulfamates can also have off-target effects in Gram-positive bacteria. Nonetheless, this study further advances PlsY as a druggable target for the development of novel antibacterial therapeutics, through the discovery and validation of the probe compound phenyl (8-phenyloctanoyl) sulfamate as a S. aureus PlsY inhibitor. 相似文献

Self-assembly of purified apolipoproteins and phospholipids results in the formation of nanometer-sized lipoprotein complexes, referred to as nanolipoprotein particles (NLPs). These bilayer constructs are fully soluble in aqueous environments and hold great promise as a model system to aid in solubilizing membrane proteins. Size variability in the self-assembly process has been recognized for some time, yet limited studies have been conducted to examine this phenomenon. Understanding the source of this heterogeneity may lead to methods to mitigate heterogeneity or to control NLP size, which may be important for tailoring NLPs for specific membrane proteins. Here, we have used atomic force microscopy, ion mobility spectrometry, and transmission electron microscopy to quantify NLP size distributions on the single-particle scale, specifically focusing on assemblies with 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) and a recombinant apolipoprotein E variant containing the N-terminal 22 kDa fragment (E422k). Four discrete sizes of E422k/DMPC NLPs were identified by all three techniques, with diameters centered at approximately 14.5, 19, 23.5, and 28 nm. Computer simulations suggest that these sizes are related to the structure and number of E422k lipoproteins surrounding the NLPs and particles with an odd number of lipoproteins are consistent with the double-belt model, in which at least one lipoprotein adopts a hairpin structure. 相似文献

The anatomically sophisticated visual system of the cubozoan jellyfish Carybdea marsupialis is described. Individual cubomedusae have eight complex eyes, each with a cornea, lens, and retina of ciliated photoreceptor
cells, eight slit ocelli, and eight dimple ocelli. The photoreceptor cells of the complex eyes are bipolar and resemble vertebrate
rod cells. Each photoreceptor has an outer cylindrical light-receptive segment that projects into a vitreous space that separates
the lens and the retina, an inner segment rich in pigment granules, and a basal region housing the nucleus. The outer segment
is a modified cilium with a 9 + 2 arrangement of microtubules plus stacks of membrane. These stacks of membrane form numerous
discs that are oriented transversely to the long axis of the cell. The outer segment is connected to the inner segment by
a slender stalk. The basal end of each photoreceptor forms an axon that projects into an underlying layer of interneurons.
Each ocellus is composed of ciliated photoreceptor cells containing pigment granules. Rhodopsin-like and opsin-like proteins
are found in the membrane stacks of the outer segments of the photoreceptors of the complex eyes. An ultraviolet-sensing opsin-like
protein is present in the inner segments and basal regions of some of the photoreceptors of the complex eyes. Rhodopsin-like
proteins are also detected in the photoreceptors of the slit ocelli. The cellular lens, composed of crystallin proteins, shows
a paucity of organelles and a high concentration of homogeneous cytoplasm. Neurons expressing RFamide (Arg-Phe-amide) comprise
a subset of interneurons found beneath the retinas of the complex eyes. RFamide-positive fibers extend from these neurons
into the stalks of the rhopalia, eventually entering into the subumbrellar nerve ring. Vision may play a role in the navigation,

We examined mitochondrial cytochrome b sequence variation in masked Sula dactylatra , red-footed S. sula , and brown S. leucogaster boobies sampled from islands in the central and eastern Pacific Ocean and in the Caribbean Sea. Each species showed a different phylogeographic pattern. Whereas haplotypes in masked and red-footed boobies were shared across the central and eastern Pacific (i.e., across the Eastern Pacific Basin), brown booby haplotypes were not shared across the Eastern Pacific Basin. Although most masked booby haplotypes from the Pacific were distinct from those in the Caribbean, one haplotype was shared across the Isthmus of Panama. Red-footed and brown boobies, however, did not share haplotypes across the Isthmus of Panama. We estimate that divergence of these regional populations occurred within the last 560,000 years. Thus, the Isthmus of Panama and the Eastern Pacific Basin (albeit to a lesser degree) appear to have played a role in the diversification of these species. 相似文献
