The laparoscopy technique was used to examine the effects of feeding 2 concentrations of energy and protein on the reproductive parameters in the doe rabbit. Forty-one does were laparoscopized on day 12 of gestation in order to estimate the ovulation rate and embryo number. Diets 1 and 2 contained 13.0 and 9.7 MJ digestible energy per kg DM and 159 and 135 g crude protein per kg DM respectively. The ovulation rate, number of implanted embryos at 12 d and total litter size at birth were similar for diets 1 and 2 (13.4 vs 14.2, 12.6 vs 12.8 and 9.1 vs 10.8), but the live litter size at birth was higher for diet 2 (7.1 vs 9.8). Laparoscopy is invaluable in determining the effects of diets on oestrus and birth rates in living and sexually active does.  相似文献

In recent decades, advances in computational methods and experimental biophysical techniques have improved our understanding of protein folding. Although some of these advances have been remarkable, the structural variability of globular proteins usually encountered makes it difficult to extract general features of their folding processes. To overcome this difficulty, experimental and computational studies of the folding of repeat (or modular) proteins are of interest. Because their native structures can be described as linear arrays of the same, repeated, supersecondary structure unit, it is possible to seek  a possibly independent behavior of the different modules without taking into account the intrinsic stability associated with different secondary structure motifs. In this work we have used a Monte Carlo-based simulation to study the folding equilibrium of four repeat proteins belonging to the tetratricopeptide repeat family. Our studies provide new insights into their energy profiles, enabling investigation about the existence of intermediate states and their relative stabilities. We have also performed structural analyses to describe the structure of these intermediates, going through the vast number of conformations obtained from the simulations. In this way, we have tried to identify the regions of each protein in which the modular structure yields a different behavior and, more specifically, regions of the proteins that can stay folded when the rest of the chain has been thermally denatured.  相似文献

Transparent exopolymer particles (TEP) compose an important pool of particulate organic matter (POM) in aquatic systems. However, no studies of TEP contribution to C export to sediment exist for freshwaters. We quantify the contribution of TEP to C sinking fluxes in an oligotrophic reservoir (Quéntar, Southern Spain) by monitoring TEP in the water column and TEP, particulate organic carbon (POC) and dry weight in sedimentation traps. TEP sinking fluxes ranged from 0.73 to 183.23 mg C m?2 day?1 and from 0.51 to 177.04 mg C m?2 day?1 at the surface and at the bottom layer, respectively. These values represent that, over an annual basis, 5.59 Ton TEP-C (over 61.32 Ton POC) are exported, on an average, from the water column to the sediment of Quentar reservoir. TEP concentrations (average = 48.0 μg XG eq l?1) were lower than the scarce data reported for freshwaters. No significant relationships between TEP and Chl a concentrations or BA were observed. Average value for daily sedimentation flux (6.63 g Dry Weight m?2 day?1) in the study reservoir was higher than that documented for low productive natural aquatic ecosystems as a consequence of the high amount of allochthonous material input characterizing reservoirs. TEP contributed to C export to sediment with a value that range from 0.02 to 31%. Our results show that even in man-made systems, which are predominantly controlled by allochthonous inputs, TEP may be relevant for explaining POM settling fluxes.  相似文献

Studies on linkage disequilibrium (LD) across the genome and populations have been used in recent years with the main objective of improving gene mapping of complex traits. Here, we characterize the patterns of genetic diversity of HLA loci and evaluate LD (D'') extent in three genomic regions: Xq13.3, NRY and HLA. In addition, we examine the distribution of DXS1225-DXS8082 haplotype diversity in Azoreans and mainland Portuguese. Allele distribution has demonstrated that the São Miguel population is genetically very diverse; haplotype analysis revealed 100% discriminatory power for X- and Y-markers and 94.3% for HLA markers. Standardized multiallelic D'' in these three genomic regions shows values lower than 0.33, thereby suggesting there is no extensive LD in the São Miguel population. Data regarding the distribution of DXS1225-DXS8082 haplotypes indicate that there are no significant differences among all the populations studied, (Azorean geographical groups, the Azores archipelago and mainland Portugal). Moreover, in these as well as in other European populations, the most frequent DXS1225-DXS8082 haplotype is 210-219. Even though São Miguel islanders and Azoreans do not constitute isolated populations and show LD for only very short physical distances, certain characteristics, such as the absence of genetic structure, the same environment and the possibility of constructing extensive pedigrees through church and civil records, offer an opportunity for dissecting the genetic background of complex diseases in these populations.  相似文献

Background  

The origin and prevalence of the prehispanic settlers of the Canary Islands has attracted great multidisciplinary interest. However, direct ancient DNA genetic studies on indigenous and historical 17th–18th century remains, using mitochondrial DNA as a female marker, have only recently been possible. In the present work, the analysis of Y-chromosome polymorphisms in the same samples, has shed light on the way the European colonization affected male and female Canary Island indigenous genetic pools, from the conquest to present-day times.  相似文献

The zoonotic potential of Ascaris infecting pigs has stimulated studies of molecular epidemiology with internal transcribed spacer 1 (ITS1) as the target. The aim of this study was to determine the value of Ascaris ITS1 as a molecular marker through assessing the intra-individual genetic diversity of Ascaris isolates from two geographical areas of Brazil. DNA was extracted from single isolated eggs, ITS1 PCR was performed, and the PCR products were cloned and sequenced. Clone analysis showed high ITS1 intra-individual variability revealed by 2–4 ITS1 genotypes/haplotypes per sample (egg). Two genotypes, G1 and G6, and 13 new haplotypes were detected and characterized. The most prevalent in humans, G1 and/or the Brazilian G6, were detected in all samples. Except for genotype G1, no relationship was observed between Brazilian ITS1 genotypes/haplotypes and those previously described in China, Bangladesh, Japan, United Kingdom, Australia, and Denmark, with respect to geographic origin or host affiliation. However, an association between the two geographically separated Brazilian ITS1 isolates was observed. The ITS1 intra-individual variability revealed in this study indicated that the use of this genetic region to discriminate human and pig Ascaris genotypes should be reconsidered.  相似文献

Question: Do abiotic constraints maintain monospecific woodlands of Juniperus thurifera? What is the role of biotic (livestock) versus abiotic (climate) drivers in the recruitment and growth of the different tree species? Location: Cabrejas range, Soria, north‐central Spain, 1200 m altitude. Methods: Stand history was reconstructed using dendro‐ecology and spatial pattern analysis, combined with historical data of livestock abundances and climatic records. Results: J. thurifera establishment occurred in two distinct pulses, with a tree component establishing in the late 1800s to early 1900s. Quercus ilex and Pinus sylvestris establishment was evident only from the late 1970s onward. Recruitment events were related to reductions in livestock browsing. J. thurifera spatial structure was clumped and Q. ilex showed a short‐scale aggregation to J. thurifera trees and saplings. Radial growth trends of J. thurifera saplings, Q. ilex and P. sylvestris were negatively related to livestock density. Summer drought limited the radial growth of all the study species, and P. sylvestris and Q. ilex grew faster than J. thurifera even after considering an age effect. Conclusions: The differences in radial growth patterns and recruitment pulses between species indicate that livestock browsing and not abiotic factors is the main factor controlling plant succession and structural development. In this process, J. thurifera acts as a nurse plant, facilitating the establishment of other tree species. Under the current low pressure from herbivores, formerly pure J. thurifera woodlands will change towards dense stands of mixed species composition.  相似文献

Spittlebugs annually infest pastures and cause severe damage, representing a serious problem for the tropical American beef cattle industry. Spittlebugs are an important biotic constraint to forage production and there is a lack of cytogenetic data for this group of insects. For these reasons, we conducted this work, in which the spermatogenesis and nucleolar behavior of Deois flavopicta, Mahanarva fimbriolata and Notozulia entreriana were studied. The males possessed testes in the shape of a "bunch of grapes"; a variable number of testicular lobes per individual and polyploid nuclei composed of several heteropycnotic bodies. A heteropycnotic area was located in the periphery of the nucleus (prophase I); the chiasmata were terminal or interstitial; metaphases I were circular or linear and anaphase showed late migration of the sex chromosome. The chromosome complement had 2n = 19, except for N. entreriana (2n = 15); the spermatids were round with heteropycnotic material in the center and elongated with conspicuos chromatin. The analysis of testes after silver nitrate staining showed polyploid nuclei with three large and three smaller nucleolar bodies. Early prophase cells had an intensely stained nucleolar body located close to the chromatin and another less evident body located away from the chromatin. The nucleolar bodies disintegrated during diplotene. Silver staining occurred in two autosomes, in terminal and subterminal locations, the latter probably corresponding to the nucleolus organizer regions (NORs). The spermatids were round with a round nucleolar body and silver staining was observed in the medial and posterior region of the elongated part of the spermatid head.  相似文献
