A novel sordarin derivative, moriniafungin (1), containing a 2-hydroxysebacic acid residue linked to C-3' of the sordarose residue of sordarin through a 1,3-dioxolan-4-one ring was isolated from the fungus Morinia pestalozzioides. Isolation of moriniafungin employed a highly specific bioassay consisting of a panel of Saccharomyces cerevisiae strains containing chimeric eEF2 for Candida glabrata, Candida krusei, Candida lusitaniae, Crytpococcus neoformans, and Aspergillus fumigatus as well as wild type and human eEF2. Moriniafungin exhibited an MIC of 6 microg/mL versus Candida albicans and IC(50)'s ranging from 0.9 to 70 microg/mL against a panel of clinically relevant Candida strains. Moriniafungin was shown to inhibit in vitro translation in the chimeric S. cerevisae strains at levels consistent with the observed IC(50). Moriniafungin has the broadest antifungal spectrum and most potent activity of any natural sordarin analog identified to date. 相似文献

High levels of factor XI (FXI) increase the risk of thromboembolic disease. However, the genetic and environmental factors regulating FXI expression are still largely unknown. The aim of our study was to evaluate the regulation of FXI by microRNAs (miRNAs) in the human liver. In silico prediction yielded four miRNA candidates that might regulate FXI expression. HepG2 cells were transfected with miR-181a-5p, miR-23a-3p, miR-16-5p and miR-195-5p. We used mir-494, which was not predicted to bind to F11, as a negative control. Only miR-181a-5p caused a significant decrease both in FXI protein and F11 mRNA levels. In addition, transfection with a miR-181a-5p inhibitor in PLC/PRF/5 hepatic cells increased both the levels of F11 mRNA and extracellular FXI. Luciferase assays in human colon cancer cells deficient for Dicer (HCT-DK) demonstrated a direct interaction between miR-181a-5p and 3′untranslated region of F11. Additionally, F11 mRNA levels were inversely and significantly correlated with miR-181a-5p levels in 114 healthy livers, but not with miR-494. This study demonstrates that FXI expression is directly regulated by a specific miRNA, miR-181a-5p, in the human liver. Future studies are necessary to further investigate the potential consequences of miRNA dysregulation in pathologies involving FXI. 相似文献

Simulation is used to facilitate new learning in a variety of situations. One application of simulation could be to help therapists gain therapeutic skills prior to seeing clients. This particular study was interested in measuring changes in stress response by looking at subjective and objective measures of distress (as measured by SUDS, HR, and HRV) over three sessions of simulated therapy. 16 second year psychology students participated in three sessions, and had their HR and HRV measured by Polar watches. Over the three sessions, there was a decrease in perceived distress, as measured by SUDS ratings. During and between sessions, there was inconclusive change in physiological parameters. 相似文献

In addition to virion formation, the coat protein (CP) of Alfalfa mosaic virus (AMV) is involved in the regulation of replication and translation of viral RNAs, and in cell-to-cell and systemic movement of the virus. An intriguing feature of the AMV CP is its nuclear and nucleolar accumulation. Here, we identify an N-terminal lysine-rich nucleolar localization signal (NoLS) in the AMV CP required to both enter the nucleus and accumulate in the nucleolus of infected cells, and a C-terminal leucine-rich domain which might function as a nuclear export signal. Moreover, we demonstrate that AMV CP interacts with importin-α, a component of the classical nuclear import pathway. A mutant AMV RNA 3 unable to target the nucleolus exhibited reduced plus-strand RNA synthesis and cell-to-cell spread. Moreover, virion formation and systemic movement were completely abolished in plants infected with this mutant. In vitro analysis demonstrated that specific lysine residues within the NoLS are also involved in modulating CP-RNA binding and CP dimerization, suggesting that the NoLS represents a multifunctional domain within the AMV CP. The observation that nuclear and nucleolar import signals mask RNA-binding properties of AMV CP, essential for viral replication and translation, supports a model in which viral expression is carefully modulated by a cytoplasmic/nuclear balance of CP accumulation. 相似文献

Activation of NMDA receptors leads to activation of cAMP-dependent protein kinase (PKA). The main substrates phosphorylated by PKA following NMDA receptor activation remain unidentified. The aim of this work was to identify a major substrate phosphorylated by PKA following NMDA receptor activation in cerebellar neurones in culture, and to assess whether this phosphorylation may be involved in neuronal death induced by excessive NMDA receptor activation. The main PKA substrate following NMDA receptor activation was identified by MALDI-TOFF fingerprinting as the nuclear protein, matrin 3. PKA-mediated phosphorylation of matrin 3 is followed by its degradation. NMDA receptor activation in rat brain in vivo by ammonia injection also induced PKA-mediated matrin 3 phosphorylation and degradation in brain cell nuclei. Blocking NMDA receptors in brain in vivo with MK-801 reduced basal phosphorylation of matrin 3, suggesting that it is modulated by NMDA receptors. Inhibition of PKA with H-89 prevents NMDA-induced phosphorylation and degradation of matrin 3 as well as neuronal death. These results suggest that PKA-mediated phosphorylation of matrin 3 may serve as a rapid way of transferring information from synapses containing NMDA receptors to neuronal nuclei under physiological conditions, and may contribute to neuronal death under pathological conditions. 相似文献

A linear model and three nonlinear models (Logan type III, Lactin and Brière) were applied to Macrolophus pygmaeus (Rambur) (Hemiptera: Miridae) at constant temperatures and validated under diel temperature variation, and field conditions. Complete development from egg to adult, with >80% survivorship, occurred at nine constant temperatures between 15 and 32 °C. Total developmental time decreased from a maximum at 15 °C (68.48 days) to a minimum at 30 °C (18.69 days) and then increased at 32 °C (23.44 days). Optimal survival and the highest developmental rate occurred within the range of 27–30 °C. The adjusted determination coefficients were high for linear and nonlinear models (>0.89). Field validation showed high levels of accuracy in all models (≥93.4%). These valid mathematical models contribute to optimal application, field management, and mass rearing of M. pygmaeus for its applicability to biological control. 相似文献

Materials and methods: Rectangular bars of L were made (60 × 6 × 2 mm) and relined (2 mm) with the relining resins Ufi Gel Hard (UH) and Tokuso Rebase Fast (TR). Specimens relined with L and intact L, TR and UH specimens were also made (60 × 6 × 4 mm), for comparison. Before relining, the L surface was left untreated or wetted with methyl methacrylate monomer and/or the bonding agents (BA) supplied by manufacturers of the reline resins. V‐notches were machined at the midpoint of the length of all specimens. The notches were made either across the width (Nw) or across the thickness of the specimens (Nth). The Charpy impact test was performed using a 0.5‐J pendulum, which had been specially designed and constructed. Data were analysed separately for each notch position using one‐way analysis of variance and Tukey honestly significant difference post‐hoc test (p = 0.05). Results: The IS of L was similar to that of L/L. For the Nw notch, treating the denture base L with TR BA and relining with TR reline material produced the highest IS. Conclusion: The IS of specimens made from heat polymerising acrylic resin Lucitone 550 was increased after relining using the hard chairside reline resin TR with its proprietary BA. 相似文献

Abstract: Partial purification of soluble guanylate cyclase on DEAE-Sephacel yields two separate peaks of guanylate cyclase activity. After 10-fold purification of the soluble enzyme, guanylate cyclase is markedly inhibited by micromolar concentrations of dopamine (I50= 0.2 μm). Dopamine inhibition is observed whether the reaction is conducted with Mn21 or with Mg2+, under atmosphere or N2(g), and using enzyme from either peak from the DEAESephacel column. Other catecholamines also inhibit partially purified guanylate cyclase with an order of potency at 1 μm of: dopamine =l -DOPA > norepinephrine = isoproterenol = adrenochrome > epinephrine. The structural requirements for inhibition are two free hydroxyl groups on the phenyl ring and an ethylamine side chain. Dopamine also inhibits the Triton X-100-solubilized microsomal guanylate cyclase after partial purification on DEAESephacel. Neither chlorpromazine, propranolol, nor phentolamine at 20 μm effectively block the dopamine inhibition of partially purified soluble guanylate cyclase. Micromolar concentrations of the reducing agents dithiothreitol and glutathione also inhibit partially purified guanylate cyclase, but unlike these agents, catecholamines can inhibit whether added in the reduced or the oxidized forms. Inhibition of enzyme activity by micromolar concentrations of dopamine, adrenochrome, or dithiothreitol is rapidly reversed by dilution and the dopamine inhibition is competitive with MgGTP. Inhibition does not appear to involve covalent binding or to result from the ability of catecholamines to reduce the concentrations of oxygen or free radicals in solution. 相似文献

Adolescent fertility displays a trend that does not follow—and even runs contrary to—the general temporal fertility pattern. Multiple factors determine this trend, including socioeconomic conditions and cultural patterns at both immigrants’ place of origin of and their destination. This article analyzes adolescent fertility in Spain with regard to country of maternal origin using records of deliveries (1980–2008) and information from countries with high immigration rates to Spain. After 1980, deliveries to adolescents diminished; after 1996, only Spanish-born adolescents continued this downward trend, but not immigrants. The factors responsible for these differences are diverse and related to the characteristics of the immigrants themselves, in addition to the situation of immigrants in the Spanish context. 相似文献

Mowing is known to favor plant diversity and influence ecosystem functioning in semi-natural grasslands. This effect could be influenced by climate variability, especially in regions with harsh climate, such as subalpine zones. In particular, short-term extreme weather fluctuations may induce rapid plant responses, affecting in turn the response to mowing. We tested the effects of concomitant summer weather manipulation and mowing on a subalpine grassland in the Central French Alps for two consecutive years. We addressed two questions: (1) How is a subalpine grassland affected by extreme summer weather? (2) Does extreme summer weather alter mowing effects on the grassland plant diversity and functioning? We used a multi-level, integrative approach assessing the responses of six abundant plant species, as well as effects on plant community structure, biomass production, and litter decomposition rates. Extreme summer weather was simulated by increasing summer temperature by 1.1°C, and decreasing summer rainfall by 80%—resulting in a 30% decrease in total annual precipitation. In addition, a heat-wave event was simulated during the first year of the experiment. This weather manipulation was combined with a late-summer mowing treatment (mown vs. unmown). Extreme summer weather mainly increased leaf senescence and decreased plant vegetative growth. Leaf litter decomposition was slowed, but only for species characterized by the fastest rates of litter decomposition. Mowing increased plant diversity by restricting the dominant grass species, thereby favoring subordinates. In the short term, this subalpine grassland was rather resistant to extreme summer weather, whereas mowing cessation remained the main factor affecting its biodiversity. 相似文献
