Summary An anaerobic fluidized bed reactor, using sepiolite as support, for the treatment of distillery wastewater was started-up using a stepped loading regime with addition of methanol. We develop an alternative input–output approach and apply it to the determination of key sectors in emissions. This methodology allows us to assess and classify the different productive sectors according to their greenhouse gas emissions and the role that they play in the productive structure, as well as the participation of their output in the total volume of production. In contrast with previous approaches, we do not focus on the responsibility of final demand, but on the responsibility of the total production of each sector. We apply our methodology to the 2014 input–output table for Spain provided by the World Input–Output Database (2016). The results show that the sectors that induce more emissions from other sectors are manufacture of food products, wholesale and retail trade, and construction. Those that are pulled to emit coincide with those that are relevant for their own final demand, being the most important electricity and gas provision, agriculture, and transportation. The classification obtained allows to orient the design of greenhouse gas emission mitigation policies for the different sectors.  相似文献

It has been suggested that L-DOPA-induced hyperhomocysteinemia can increase the risk of stroke, heart disease, and dementia and is an additional pathogenetic factor involved in the progression of Parkinson's disease. In Chinese hamster ovary (CHO) cells stably cotransfected with adenosine A(2A) and dopamine D2 receptors, homocysteine selectively decreased the ability of D2 receptor stimulation to internalize adenosine A(2A)-dopamine D2 receptor complexes. Radioligand-binding experiments in the same cell line demonstrated that homocysteine acts as an allosteric D2 receptor antagonist, by selectively reducing the affinity of D2 receptors for agonists but not for antagonists. Mass spectrometric analysis showed that, by means of an arginine (Arg)-thiol electrostatic interaction, homocysteine forms noncovalent complexes with the two Arg-rich epitopes of the third intracellular loop of the D2 receptor, one of them involved in A(2A)-D2 receptor heteromerization. However, homocysteine was unable to prevent or disrupt A(2A)-D2 receptor heteromerization, as demonstrated with Fluorescence Resonance Energy Transfer (FRET) experiments in stably cotransfected HEK cells. The present results could have implications for Parkinson's disease.  相似文献

Integrase (IN) is one of the three human immunodeficiency virus type 1 (HIV-1) enzymes essential for effective viral replication. S-1360 is a potent and selective inhibitor of HIV-1 IN. In this work, we have carried out molecular dynamics (MD) simulations using a hybrid Quantum Mechanics/Molecular Mechanics (QM/MM) approach, to determine the protein-ligand interaction energy for S-1360 and two analogues. Analysis of the MD trajectories reveals that the strongest protein-inhibitor interactions, observed in the three studied complexes, are established with Lys-159 residue and Mg(2+) cation. Calculations of binding energy using BLYP/MM level of theory reveal that there is a direct relationship between this theoretical computed property and the experimental determined anti-HIV activity.  相似文献

Identification of higher-order oligomers in the plasma membrane is essential to decode the properties of molecular networks controlling intercellular communication. We combined bioluminescence resonance energy transfer (BRET) and fluorescence resonance energy transfer (FRET) in a technique called sequential BRET-FRET (SRET) that permits identification of heteromers formed by three different proteins. In SRET, the oxidation of a Renilla luciferase (Rluc) substrate by an Rluc fusion protein triggers acceptor excitation of a second fusion protein by BRET and subsequent FRET to a third fusion protein. We describe two variations of SRET that use different Rluc substrates with appropriately paired acceptor fluorescent proteins. Using SRET, we identified complexes of cannabinoid CB(1), dopamine D(2) and adenosine A(2A) receptors in living cells. SRET is an invaluable technique to identify heteromeric complexes of more than two neurotransmitter receptors, which will allow us to better understand how signals are integrated at the molecular level.  相似文献

Changes on abscisic acid (ABA), jasmonic acid (JA) and indole-3-acetic acid (IAA) levels were investigated in papaya seedlings (Carica papaya L.) cv. “Baixinho de Santa Amalia” under progressive water stress and subsequent rehydration. Also, the behaviour of leaf gas exchange and leaf growth was determined under stress condition. The results indicated that ABA and JA differ in their pattern of change under water stress. ABA continuously increased in leaves and roots during the whole period of stress whereas JA showed a sharp increase and a later decrease in both organs. Re-watering reduced rapidly (24 h) leaf and root ABA to control levels whereas the influence on JA levels could not be assessed. Drought and recovery did not alter IAA levels in leaf and root tissues of papaya seedlings. In addition, water stress reduced stomatal conductance, photosynthetic rate, transpiration rate, the percentage of attached leaves and leaf growth. Rehydration reverted in few days the effects of stress on leaf growth and gas exchange parameters. Overall, the data suggest that ABA could be involved in the induction of several progressive responses such as the induction of stomatal closure and leaf abscission to reduce papaya water loss. In addition, the pattern of accumulation of JA is compatible with a triggering signal upstream ABA. The unaltered levels of IAA could suggest a certain adaptive ability of papaya to maintain active physiological processes under progressive drought stress.  相似文献

Metabolites of nandrolone were determined in the urine of several sportsmen, sedentary and post-menopausal women by capillary gas chromatography–mass spectrometry quadrupole (GC–MS) and capillary gas chromatography mass–mass spectrometry ion trap (GC–MS–MS) methods. The method employed was GC–EI-MS with 17α-methyltestosterone as internal standard with ethyl ether extraction prior to selected ion monitoring of the bis(trimethylsilyl) ethers at ion masses m/z 405 and 420 for the nandrolone metabolites, and 418 and 403 for nandrolone derivative. Recovery for nandrolone, 19-norandrosterone (19-NA) and 19-noretiocholanolone (19-NE) was 97.20, 94.17 and 95.54%, respectively. Detection limits for nandrolone, 19-NA and 19-NE were 0.03, 0.01 and 0.06 ng/ml. Metabolites of nandrolone (19-NA and 19-NE) were found in 12.5% (n=40) of sportsmen and 40% (n=10) of post-menopausal women.  相似文献
