Dioecious alpine juniper has been influenced by human impacts, management and climate changes; hence, its present populations are remnant fragments of its former distribution in central Europe. Complex environmental shifts have had fatal consequences for growth, reproduction and health of juniper and hence for its population structure. We asked the questions: 1) what was the population size structure, the sex ratio and the health status of individuals? 2) How were the population parameters linked with the environmental conditions and surrounding vegetation? The study area was close and above the upper forest limit in central Europe, the Hrubý Jeseník Mts. The parameters of each juniper individual and its environmental conditions were obtained, Ellenberg's indicator values and habitat categories were also determined. Proportions of sex categories, shrub size and environmental conditions were investigated to assess relationships between the population categories and environmental factors. We revealed linkage among the shrub size, health vigor, vegetation cover, a habitat and environmental factors. While there was equal ratio of females and males (1:1), small individuals of undetermined sex predominated that reducing the effective population size. Juniper health vigor was associated with surrounding vegetation cover. Individuals of undetermined sex were smaller than those of remaining sex categories and prefer specific habitats at higher elevation, underlining the effect of abiotic conditions on sex performance and ratio. The observed pattern, which was attributed to shift in land-use practices in the middle of 20th century and climate changes, could be improved by management. We recommended experimental local grazing and mowing accompanying by profound and continuing assessment of interaction among environmental factors and juniper performance.  相似文献

Species inhabiting mountain ecosystems are expected to be particularly vulnerable to environmental change, yet information on their basic ecology is often lacking. Knowledge from field-based empirical studies remains essential to refine our understanding of the impact of current habitat alterations and for the consequential development of meaningful conservation management strategies. This study focuses on a poorly investigated and vulnerable mountain bird species in Europe, the Ring Ouzel Turdus torquatus. Our aim was to identify the species’ key ecological requirements during the crucial period of nestling provisioning in the context of environmental change. We radiotracked and observed Alpine Ring Ouzels in a high-density population, investigating their pattern of foraging habitat selection in 2015 and 2017, and evaluated the transferability of these results over a wider geographical range across the SW Swiss Alps. Foraging birds selected, consistently in space and time, short grass swards (< 10 cm) with interspersed patches of accessible and penetrable soils, at intermediate moisture levels (around 40–65% volumetric water content). In Alpine ecosystems, this microhabitat configuration is typically widespread during the spring snowmelt, but extremely seasonal, with a rapid decrease in its availability over the course of the breeding season. This underlines the high vulnerability of the Ring Ouzel to environmental change: an earlier snowmelt could generate a temporal mismatch between the peak of the breeding effort and optimal foraging conditions; however, abandoning grazing activities on semi-wooded Alpine pastures may further decrease foraging habitat suitability through taller and denser grass swards, and subsequent woody vegetation encroachment. This study provides a mechanistic appraisal of the challenges Ring Ouzels will face in the future, as well as initial guidelines for targeted habitat management within timberline ecotones.  相似文献

NADH cytochrome b5 oxidoreductase (Ncb5or) protects β-cells against oxidative stress and lipotoxicity. The predominant phenotype of lean Ncb5or-null mouse is insulin-dependent diabetes due to β-cell death. This suggests the putative role of NCB5OR polymorphism in human diabetes. Therefore, we aimed to investigate the effect of natural missense mutations on the expression of human NCB5OR. Protein and mRNA levels of five non-synonymous coding variants were analyzed in transfected HEK293 and HepG2 cells. Although the mRNA levels were only slightly affected by the mutations, the amount of Ncb5or protein was largely reduced upon two Glu to Gly replacements in the third exon (p.E87G, p.E93G). These two mutations remarkably and synergistically shortened the half-life of Ncb5or and their effect could be attenuated by proteasome inhibitors. Our results strongly indicate that p.E87G, p.E93G mutations lead to enhanced proteasomal degradation due to manifest conformational alterations in the b5 domain. These data provide first evidence for natural mutations in NCB5OR gene resulting in decreased protein levels and hence having potential implications in human pathology.  相似文献

DNA repair events have functional significance especially for genome stability. Although the DNA damage response within the whole genome has been extensively studied, the region-specific characteristics of nuclear sub-compartments such as the nucleolus or fragile sites have not been fully elucidated. Here, we show that the heterochromatin protein HP1 and PML protein recognize spontaneously occurring 53BP1- or γ-H2AX-positive DNA lesions throughout the genome. Moreover, 53BP1 nuclear bodies, which co-localize with PML bodies, also occur within the nucleoli compartments. Irradiation of the human osteosarcoma cell line U2OS with γ-rays increases the degree of co-localization between 53BP1 and PML bodies throughout the genome; however, the 53BP1 protein is less abundant in chromatin of ribosomal genes and fragile sites (FRA3B and FRA16D) in γ-irradiated cells. Most epigenomic marks on ribosomal genes and fragile sites are relatively stable in both non-irradiated and γ-irradiated cells. However, H3K4me2, H3K9me3, H3K27me3 and H3K79me1 were significantly changed in promoter and coding regions of ribosomal genes after exposure of cells to γ-rays. In fragile sites, γ-irradiation induces a decrease in H3K4me3, changes the levels of HP1β, and modifies the levels of H3K9 acetylation, while the level of H3K9me3 was relatively stable. In these studies, we confirm a specific DNA-damage response that differs between the ribosomal genes and fragile sites, which indicates the region-specificity of DNA repair.  相似文献

Protein aggregation is related to a series of pathological disorders the main cause of which are the fibrillar species generated during the process. Human serum albumin (HSA) undergoes rapid fibrillation in the presence of Cu(II) at pH 7.4 in 60% ethanol after 6-h incubation (~65?°C) followed by room temperature incubation. Here, we have investigated the effect of a stoichiometric variation of Cu(II) on the self-assembly of HSA using Congo red and thioflavin T dye-binding studies, circular dichroism spectroscopy, Fourier transform infrared spectroscopy, electron paramagnetic resonance spectroscopy, fluorescence microscopy and transmission electron microscopy. The simulation of EPR spectra suggests that with the increment in Cu(II) ion concentration, there is a change in ligand field coordination. Kinetic parameters indicate reduced cooperativity that may be related to the nonspecific coordination on increment of Cu(II) concentration. Cu(II) is also able to direct the accumulation of a large number of fibers along with a formation of dense fibrillar network which is evident from microscopic images.  相似文献

Developmental plasticity is often correlated with diversity and has been proposed as a facilitator of phenotypic novelty. Yet how a dimorphism arises or how additional morphs are added is not understood, and few systems provide experimental insight into the evolution of polyphenisms. Because plasticity correlates with structural diversity in Pristionchus nematodes, studies in this group can test the role of plasticity in facilitating novelty. Here, we describe three new species, Pristionchus fukushimae sp. nov. , Pristionchus hoplostomus sp. nov. , and the hermaphroditic Pristionchus triformis sp. nov. , which are characterized by a novel polymorphism in their mouthparts. In addition to showing the canonical mouth dimorphism of diplogastrid nematodes, comprising a stenostomatous (‘narrow‐mouthed’) and a eurystomatous (‘wide‐mouthed’) form, the new species exhibit forms with six, 12, or intermediate numbers of cheilostomatal plates. Correlated with this polymorphism is another trait that varies among species: whereas divisions between plates are complete in P. triformis sp. nov. , which is biased towards a novel ‘megastomatous’ form comprising 12 complete plates, the homologous divisions in the other new species are partial and of variable length. In a reconstruction of character evolution, a phylogeny inferred from 26 ribosomal protein genes and a partial small subunit rRNA gene supported the megastomatous form of P. triformis sp. nov. as the derived end of a series of split‐plate forms. Although split‐plate forms were normally only observed in eurystomatous nematodes, a single 12‐plated stenostomatous individual of P. hoplostomus sp. nov. was also observed, suggesting independence of the two types of mouth plasticity. By introducing these new species to the Pristionchus model system, this study provides further insight into the evolution of polymorphisms and their evolutionary intermediates. © 2013 The Linnean Society of London  相似文献

The ATPase-driven dimeric molecular Hsp90 (heat shock protein 90) and its cofactor Cdc37 (cell division cycle 37 protein) are crucial to prevent the cellular depletion of many protein kinases. In complex with Hsp90, Cdc37 is thought to bind an important lid structure in the ATPase domain of Hsp90 and inhibit ATP turnover by Hsp90. As different interaction modes have been reported, we were interested in the interaction mechanism of Hsp90 and Cdc37. We find that Cdc37 can bind to one subunit of the Hsp90 dimer. The inhibition of the ATPase activity is caused by a reduction in the closing rate of Hsp90 without obviously bridging the two subunits or affecting nucleotide accessibility to the binding site. Although human Cdc37 binds to the N-terminal domain of Hsp90, nematodal Cdc37 preferentially interacts with the middle domain of CeHsp90 and hHsp90, exposing two Cdc37 interaction sites. A previously unreported site in CeCdc37 is utilized for the middle domain interaction. Dephosphorylation of CeCdc37 by the Hsp90-associated phosphatase PPH-5, a step required during the kinase activation process, proceeds normally, even if only the new interaction site is used. This shows that the second interaction site is also functionally relevant and highlights that Cdc37, similar to the Hsp90 cofactors Sti1 and Aha1, may utilize two different attachment sites to restrict the conformational freedom and the ATP turnover of Hsp90.  相似文献

A method is described to assess the reproductive status of male Hippocampus abdominalis on the basis of behavioural traits. The non‐invasive nature of this technique minimizes handling stress and reduces sampling requirements for experimental work. It represents a useful tool to assist researchers in sample collection for studies of reproduction and development in viviparous syngnathids, which are emerging as important model species.  相似文献
