We carried out a soil sampling survey in September 2008 in central Cayenne, French Guiana, using molecular methods to assess the presence of the dimorphic fungus Histoplasma capsulatum. Four of the 31 samples collected (12.9?%) tested positive by PCR, with confirmation of the result by DNA sequencing. H. capsulatum is therefore present in urban environments in French Guiana. These results provide additional support for the primary prophylaxis of AIDS-related histoplasmosis in French Guiana.  相似文献

Autologous disc cell implantation, growth factors and gene therapy appear to be promising therapies for disc regeneration. Unfortunately, the replicative lifespan and growth kinetics of human nucleus pulposus (NP) cells related to host age are unclear. We investigated the potential relations among age, replicative lifespan and growth rate of NP cells, and determined the age range that is suitable for cell-based biological therapies for degenerative disc diseases. We used NP tissues classified by decade into five age groups: 30s, 40s, 50s, 60s and 70s. The mean cumulative population doubling level (PDL) and population doubling rate (PDR) of NP cells were assessed by decade. We also investigated correlations between cumulative PDL and age, and between PDR and age. The mean cumulative PDL and PDR decreased significantly in patients in their 60s. The mean cumulative PDL and PDR in the younger groups (30s, 40s and 50s) were significantly higher than those in the older groups (60s and 70s). There also were significant negative correlations between cumulative PDL and age, and between PDR and age. We found that the replicative lifespan and growth rate of human NP cells decreased with age. The replicative potential of NP cells decreased significantly in patients 60 years old and older. Young individuals less than 60 years old may be suitable candidates for NP cell-based biological therapies for treating degenerative disc diseases.  相似文献

A cAMP-adenosine binding protein partially purified from exponentially growing Dictyostelium discoideum cells carries S-adenosyl-L-homocysteine (SAH) hydrolase activity. This protein is present throughout the developmental cycle and has many properties in common with a cAMP binding activity previously reported from this laboratory (Gunzburg and Véron, 1981). Direct binding measurements with radioactive ligands indicate a dissociation constant of 0.2 microM for adenosine and 9 nM for cAMP, a value in good agreement with measurements of the rate constants for cAMP binding (k+1 = 2.4 X 10(4) M-1 sec-1) and dissociation (k-1 = 1.1 X 10(-4) sec-1). The binding of cAMP is completely abolished in the presence of 1 microM adenosine; a maximum 60 per cent inhibition of adenosine binding can be achieved with cAMP concentrations as high as 0.1 microM, suggesting that at least some of the cAMP and adenosine binding sites are not identical. The protein has a sedimentation coefficient of 9.2S and a native molecular weight of 190,000, as judged by gel filtration. Labeling with the photoaffinity ligand 8-azido-[3H]-cAMP followed by SDS polyacrylamide gel electrophoresis results in a single band of 47,000 MW, suggesting that the protein may be a tetramer. The physiological importance of the protein and its association with SAH hydrolase activity is discussed in relation to a possible role in the regulation of protein and phospholipid methylation that occurs during chemotaxis.  相似文献

Coral reef geomorphology and community composition were investigated in the tropical northeastern Pacific during April 1994. Three areas were surveyed in the Revillagigedo Islands (Mexico), and an intensive study was conducted on Clipperton Atoll (1,300 km SW of Acapulco), including macro-scale surface circulation, sea surface temperature (SST) climatology, geomorphology, coral community structure, zonation, and biogeography. Satellite-tracked drifter buoys from 1979–1993 demonstrated complex patterns of surface circulation with dominantly easterly flow (North Equatorial Counter Current, NECC), but also westerly currents (South Equatorial Current, SEC) that could transport propagules to Clipperton from both central and eastern Pacific regions. The northernmost latitude reached by the NECC is not influenced by El Niño-Southern Oscillation (ENSO) events, but easterly flow velocity evidently is accelerated at such times. Maximum NECC flow rates indicate that the eastern Pacific barrier can be bridged in 60 to 120 days. SST anomalies at Clipperton occur during ENSO events and were greater at Clipperton in 1987 than during 1982–1983. Shallow (15–18 m)and deep (50–58 m) terraces are present around most of Clipperton, probably representing Modern and late Pleistocene sea level stands. Although Clipperton is a well developed atoll with high coral cover, the reef-building fauna is depauperate, consisting of only 7 species of scleractinian corals belonging to the generaPocillopora, Porites, Pavona andLeptoseris, and 1 species of hydrocoral in the genusMillepora. The identities of the one Pocilpopora species and one of the twoPorites species are still unknown. Two of the remaining scleractinians (Pavona minuta, Leptoseris scabra) and the hydrocoral (Millepora exaesa), all formerly known from central and western Pacific localities, represent new eastern Pacific records. Scleractinian corals predominate (10–100% cover) over insular shelf depths of 8 to 60m, and crustose coralline algae are dominant (5–40% cover) from 0.5 to 7m. Spur and groove features, constructed of alternating frameworks ofPocillopora andPorites, and veneered with crustose coralline algae, are generally well developed around most atoll exposures. Although crustose coralline algae predominate in the breaker zone (with up to 100% cover), a prominent algal ridge is absent with only a slight buildup (ca. 10 cm) to seaward. Frequent grazing by the pufferfishArothron meleagris results in the removal of large amounts of live tissue and skeleton fromPorites lobata. Acanthaster planci is present, but rare. The grazing of large diadematid sea urchins, (2 species each ofDiadema andEchinothrix) on dead corals cause extensive erosion in some areas. Large numbers of corals on the 15–18 m terrace had recently suffered partial (P. lobata, 60–70% maximum of all colonies sampled) or total (Pocillopora sp., 80% maximum) mortality. The lengths of regenerating knobs and the rates of linear skeletal growth inP. lobata, determined by sclerochronologic analysis, indicated a period of stress during 1987. Massive skeletal growth is significantly higher at intermediate (16–17 m) than shallow (6–8 m) depths with mean extension rates of 1.5 mm yr–1 inP. lobata and 1.4 mm yr–1 inP. minuta at intermediate depths. Skeletal growth inP. lobata was depressed during the 1987 El Nifio event at Clipperton. The branching coralPocillopora sp. demonstrated high and similar skeletal growth rates at both shallow (25.4 mm yr–1) and intermediate (26.5 mm yr–1) depths. The presence of widely distributed Indo-Pacific zooxanthellate corals at Clipperton and the Revillagigedo Islands indicates that these NE Pacific Islands probably serve as a stepping stone for dispersal into the far eastern Pacific region.  相似文献

Metal concentrations in the gill, muscle and liver tissues of Labeo rosae from two impoundments, Loskop and Flag Boshielo dams on the Olifants River, were evaluated in 2011 to detect patterns in metal associations between tissues and impoundments. Elevated concentrations of Ba, Zn, B, Al, Si and Fe, relative to a pristine site in the catchment, were found in the muscle, liver and gill tissues at both impoundments. Molybdenum concentrations were exceptionally high in all tissues at Loskop Dam and in liver at Flag Boshielo Dam. No definite pattern in the ratio metal concentrations within, or between, fish tissues was identified. The expected trend, liver > gills > muscle, was found at both impoundments, but was less prominent at Loskop Dam. Metal concentrations in muscle of Loskop Dam fish were significantly higher than in those at Flag Boshielo Dam. The inverse was true for liver. The long-term impact of elevated metal concentrations on fish health at both impoundments raises concern.  相似文献

The effect of silver ionic exchange on the glass structure in a molten bath at 350 °C was presented in a previous paper Catan et al. [1] (J NonCryst Solids 354:1026–1031, 2008). In this paper, the experiment is driven for a temperature near 310 °C, the eutectic of a 10% AgNO3–NaNO3 molten salt. The various exchanged silicate glasses are further annealed to obtain silver particles in the matrix. Infrared spectroscopy combined with UV/Visible spectroscopy and scanning electron microscopy analysis allowed to correlate the silver-ion penetration and particle formation with the degree of polymerisation of the silicate network. The previous results have demonstrated an insertion of silver ions in the glass structure that is about 10% higher than the departure of sodium ions. Infrared results obtained after ion exchange have proved that local alterations lead to a higher degree of depolymerisation of the silicate network. Here, the annealing of the samples promotes the formation of silver nanoparticles, the infrared measurements prove that the aggregation is correlated to a repolymerisation of the silicate network. Transmission electron microscopy (TEM) is used to evaluate the distribution size of the silver particles after annealing and to correlate it to the evolution of the absorption curves. The TEM observations prove that the particle are below the incident wavelength but shape factor could lead to scattering contribution when particle growths and to absorption spectra evolution.  相似文献
