Acute kidney injury (AKI) is an underestimated, yet important risk factor for development of chronic kidney disease (CKD). Even after initial total recovery of renal function, some patients develop progressive and persistent deterioration of renal function and these patients are more likely to progress to end-stage renal disease (ESRD). Animal models are indispensable for unravelling the mechanisms underlying this progression towards CKD and ESRD and for the development of new therapeutic strategies in its prevention or treatment. Ischemia (i.e. hypoperfusion after surgery, bleeding, dehydration, shock, or sepsis) is a major aetiology in human AKI, yet unilateral ischemia-reperfusion is a rarely used animal model for research on CKD and fibrosis. Here, we demonstrate in C57Bl/6J mice, by both histology and gene expression, that unilateral ischemia-reperfusion without contralateral nephrectomy is a very robust model to study the progression from acute renal injury to long-term tubulo-interstitial fibrosis, i.e. the histopathological hallmark of CKD. Furthermore, we report that the extent of renal fibrosis, in terms of Col I, TGFβ, CCN2 and CCN3 expression and collagen I immunostaining, increases with increasing body temperature during ischemia and ischemia-time. Thus, varying these two main determinants of ischemic injury allows tuning the extent of the long-term fibrotic outcome in this model. Finally, in order to cover the whole practical finesse of ischemia-reperfusion and allow model and data transfer, we provide a referenced overview on crucial technical issues (incl. anaesthesia, analgesia, and pre- and post-operative care) with the specific aim of putting starters in the right direction of implementing ischemia in their research and stimulate them, as well as the community, to have a critical view on ischemic literature data.  相似文献

The relationship between body size and basal metabolic rate (BMR) in homeotherms has been treated in the literature primarily by comparison between species of mammals or birds. This paper focuses on the intraindividual changes in BMR when body mass (W) varies with different maintenance regimens. BMR varied in individual kestrels in proportion to W1.67, which is considerably steeper than the mass exponents for homomorphic change (0.667; Heusner, 1984) for interspecific comparison among all birds (0.677) or raptors (0.678), for interindividual comparison of kestrels on ad libitum maintenance regimens (0.786), and for mass proportionality (1.00). The circadian range of telemetered core temperature also varied more strongly with intraindividual than with interspecific (Aschoff, 1981a) variation in mass. This was due to reduced nocturnal core temperature at low-maintenance regimens, which was, however, insufficient to account for the excessive reduction in BMR. kidney lean mass at Carcass analysis of eight birds sacrificed revealed a disproportionate reduction in heart and kidney lean mass at low-maintenance regimens. We surmise that variation in BMR primarily reflects variation in these metabolically highly active tissues. This may account for positive correlations found between heart, kidney, and BMR residuals relative to interspecific allometric prediction, and between alpha and rho residuals, as expected on the basis of the constant excess of BMR during alpha above BMR during rho (Aschoff & Pohl, 1970a).  相似文献

Eubacterium lentum (33 strains) isomerized the 12-cis double bond of C18 fatty acids with cis double bonds at C-9 and C-12 into an 11-trans double bond before reduction of the 9-cis double bond. The 14-cis double bond of homo-γ-linolenic acid was isomerized by 29 strains into a 13-trans double bond. The same strains isomerized the 14-cis double bond of arachidonic acid into a 13-trans double bond and then isomerized the 8-cis double bond into a 7-trans double bond; the 13-cis double bond of 10-cis, 13-cis-nonadecadienoic acid was isomerized into a 12-trans double bond. None of these isomerization products was further reduced. Studies with resting cells showed optimal isomerization velocity at a linoleic acid concentration of 37.5 μM; higher concentrations were inhibitory. The pH optimum for isomerization was 7.5 to 8.5. The isomerase was inhibited by the sulfhydryl reagents iodoacetamide, bromoacetate, and N-ethylmaleimide and by the chelators EDTA and 1,10-phenanthroline.  相似文献

Background and aims

Conservation agriculture, the combination of minimal soil movement (zero or reduced tillage), crop residue retention and crop rotation, might have the potential to increase soil organic C content and reduce emissions of CO2.

Methods

Three management factors were analyzed: (1) tillage (zero tillage (ZT) or conventional tillage (CT)), (2) crop rotation (wheat monoculture (W), maize monoculture (M) and maize-wheat rotation (R)), and (3) residue management (with (+r), or without (?r) crop residues). Samples were taken from the 0–5 and 5–10?cm soil layers and separated in micro-aggregates (< 0.25?mm), small macro-aggregates (0.25 to 1?mm) and large macro-aggregates (1 to 8?mm). The carbon content of each aggregate fraction was determined.

Results

Zero tillage combined with crop rotation and crop residues retention resulted in a higher proportion of macro-aggregates. In the 0–5?cm layer, plots with a crop rotation and monoculture of maize and wheat in ZT+r had the greatest proportion of large stable macro-aggregates (40%) and highest mean weighted diameter (MWD) (1.7?mm). The plots with CT had the largest proportion of micro-aggregates (27%). In the 5–10?cm layer, plots with residue retention in both CT and ZT (maize 1?mm and wheat 1.5?mm) or with monoculture of wheat in plots under ZT without residues (1.4?mm) had the greatest MWD. The 0–10?cm soil layer had a greater proportion of small macroaggregates compared to large macro-aggregates and micro-aggregates. In the 0–10?cm layer of soil with residues retention and maize or wheat, the greatest C content was found in the small and large macro-aggregates. The small macro-aggregates contributed most C to the organic C of the sample. For soil cultivated with maize, the CT treatments had significantly higher CO2 emissions than the ZT treatments. For soil cultivated with wheat, CTR-r had significantly higher CO2 emissions than all other treatments.

Conclusion

Reduction in soil disturbance combined with residue retention increased the C retained in the small and large macro-aggregates of the top soil due to greater aggregate stability and reduced the emissions of CO2 compared with conventional tillage without residues retention and maize monoculture (a cultivation system normally used in the central highlands of Mexico).  相似文献

The total size of the world population is likely to increase from its current 7 billion to 8–10 billion by 2050. This uncertainty is because of unknown future fertility and mortality trends in different parts of the world. But the young age structure of the population and the fact that in much of Africa and Western Asia, fertility is still very high makes an increase by at least one more billion almost certain. Virtually, all the increase will happen in the developing world. For the second half of the century, population stabilization and the onset of a decline are likely. In addition to the future size of the population, its distribution by age, sex, level of educational attainment and place of residence are of specific importance for studying future food security. The paper provides a detailed discussion of different relevant dimensions in population projections and an evaluation of the methods and assumptions used in current global population projections and in particular those produced by the United Nations and by IIASA.  相似文献
