MyD88 is an adapter protein that links toll-like receptors (TLRs) and Interleukin-1 receptors (IL-1Rs) with downstream signaling molecules. The MyD88 has been found to be an essential mediator in the development of osteoarthritis in articular cartilage. However, the role of the MyD88 pathway has yet to be elucidated in the intervertebral disk (IVD). Using in vitro techniques, we analyzed the effect of MyD88 pathway-specific inhibition on the potent inflammatory and catabolic mediator LPS and IL-1 in bovine and human nucleus pulposus (NP) cells by assessing matrix-degrading enzyme expression, including matrix metalloproteases (MMPs) and a disintegrin-like and metalloprotease with thrombospondin motifs (ADAMTS family). We also analyzed inhibition of MyD88 in the regulation of inducible nitric oxide synthase and TLR-2. Finally, we used an ex vivo organ culture model to assess the effects of MyD88 inhibitor (MyD88i) on catabolic factor-induced disk degeneration in mice lumbar disks. In bovine NP cells, MyD88i potently antagonizes LPS- or IL-1-mediated induction of cartilage-degrading enzyme production, including MMP-1, MMP-13, ADAMTS-4, and ADAMTS-5. MyD88i also attenuates the LPS- or IL-1-mediated induction of iNOS and TLR-2 gene expression. Our ex vivo findings reveal inhibition of MyD88 via counteraction of IL-1-mediated proteoglycan depletion. The findings from this study demonstrate the potent anti-inflammatory and anti-catabolic effects of inhibition of MyD88 pathway inhibition on IVD homeostasis, suggesting a potential therapeutic benefit of a MyD88i in degenerative disk disease in the future. 相似文献

Background
Much progress has been made in understanding the 3D structure of proteins using methods such as NMR and X-ray crystallography. The resulting 3D structures are extremely informative, but do not always reveal which sites and residues within the structure are of special importance. Recently, there are indications that multiple-residue, sub-domain structural relationships within the larger 3D consensus structure of a protein can be inferred from the analysis of the multiple sequence alignment data of a protein family. These intra-dependent clusters of associated sites are used to indicate hierarchical inter-residue relationships within the 3D structure. To reveal the patterns of associations among individual amino acids or sub-domain components within the structure, we apply a k-modes attribute (aligned site) clustering algorithm to the ubiquitin and transthyretin families in order to discover associations among groups of sites within the multiple sequence alignment. We then observe what these associations imply within the 3D structure of these two protein families.Results
The k-modes site clustering algorithm we developed maximizes the intra-group interdependencies based on a normalized mutual information measure. The clusters formed correspond to sub-structural components or binding and interface locations. Applying this data-directed method to the ubiquitin and transthyretin protein family multiple sequence alignments as a test bed, we located numerous interesting associations of interdependent sites. These clusters were then arranged into cluster tree diagrams which revealed four structural sub-domains within the single domain structure of ubiquitin and a single large sub-domain within transthyretin associated with the interface among transthyretin monomers. In addition, several clusters of mutually interdependent sites were discovered for each protein family, each of which appear to play an important role in the molecular structure and/or function.Conclusions
Our results demonstrate that the method we present here using a k- modes site clustering algorithm based on interdependency evaluation among sites obtained from a sequence alignment of homologous proteins can provide significant insights into the complex, hierarchical inter-residue structural relationships within the 3D structure of a protein family.104.

There is a clear need to develop conservation agriculture technologies appropriate for surface irrigated conditions, and the adoption by small-scale farmers. The objective of this paper is to evaluate the effect on soil quality of five different tillage-crop residue management systems (conventionally tilled raised beds [CTB] with residue incorporated and permanent raised beds [PB] with residue burned, removed, partly retained or fully retained). Data were collected in a long-term trial established in 1992 with a wheat-maize rotation under irrigated, arid conditions in north-western Mexico. Three groups of tillage-straw systems with different characteristics in relation to the soil environment were distinguished: PB-straw burned, CTB-straw incorporated, and PB-straw not burned. The PB-straw burned had high electrical conductivity, Na concentration and penetration resistance and low soil resilience and aggregation, showing that the combination of PB with the burning of residues is not a sustainable management option. The CTB-straw incorporated was distinguished from the PB practices by the soil physical variables, especially the low direct infiltration and aggregate stability, indicating degradation of physical soil quality in this system. The practice of PB, where all or part of the residue is retained in the field, seems to be the most sustainable option for this cropping system. 相似文献

A field experiment was conducted under furrow irrigation on a Vertisol in arid northwestern Mexico, to evaluate sustainable production alternatives for irrigated wheat systems. Treatments included: tillage (conventionally tilled raised beds where new beds are formed after disc ploughing before planting [CTB] and permanent raised beds [PB]) and irrigation regimes (full and reduced). Physical and chemical soil quality was compared among treatments. PB improved soil structure and direct infiltration, increased topsoil K concentrations (0–5 cm; 1.6 cmol kg?1 in PB vs. 1.0–1.1 cmol kg?1 in CTB) and reduced Na concentrations (0–5 cm; 1.3–1.4 cmol kg?1 in PB vs. 1.9–2.2 cmol kg?1 in CTB) compared to CTB. Crop growth dynamics were studied throughout the season with an optical handheld NDVI sensor. Crop growth was initially slower in PB compared to CTB, but this was compensated by increased crop growth in the later stages of the crop cycle which influenced final yield, especially under reduced irrigation. These results were reflected in the final grain yield: in the third year after conversion to PB, no difference in grain yield was found between tillage systems under full irrigation. However, under reduced irrigation the improved soil quality with PB resulted in a 19% and 26% increment in bread and durum wheat grain yields, respectively. As projected climatic scenarios forecast higher evapotranspiration, less reliable rainfall and increased drought, our results indicate that PB could contribute to maintaining and increasing wheat yields in a sustainable way. 相似文献

The African malaria mosquito Anopheles gambiae sensu stricto continues to play an important role in malaria transmission, which is aggravated by its high degree of anthropophily, making it among the foremost vectors of this disease. In the current study we set out to unravel the strong association between this mosquito species and human beings, as it is determined by odorant cues derived from the human skin. Microbial communities on the skin play key roles in the production of human body odour. We demonstrate that the composition of the skin microbiota affects the degree of attractiveness of human beings to this mosquito species. Bacterial plate counts and 16S rRNA sequencing revealed that individuals that are highly attractive to An. gambiae s.s. have a significantly higher abundance, but lower diversity of bacteria on their skin than individuals that are poorly attractive. Bacterial genera that are correlated with the relative degree of attractiveness to mosquitoes were identified. The discovery of the connection between skin microbial populations and attractiveness to mosquitoes may lead to the development of new mosquito attractants and personalized methods for protection against vectors of malaria and other infectious diseases. 相似文献

More than 30% of all pregnancies in the UK require some form of assistance at delivery, with one of the more severe forms of assistance being an emergency Caesarean section (ECS). Previously it has been shown that the likelihood of a delivery via ECS is positively associated with the birth weight and size of the newborn and negatively with maternal height. Paternal height affects skeletal growth and mass of the fetus, and thus might also affect pregnancy outcomes. We hypothesized that the effect of newborn birth weight on the risk of ECS would decrease with increasing maternal height. Similarly, we predicted that there would be an increase in ECS risk as a function of paternal height, but that this effect would be relative to maternal height (i.e., parental height differences). We used data from the Millennium Cohort Study: a large-scale survey (N = 18,819 births) with data on babies born and their parents from the United Kingdom surveyed 9 to 12-months after birth. We found that in primiparous women, both maternal height and parental height differences interacted with birth weight and predicted the likelihood of an ECS. When carrying a heavy newborn, the risk of ECS was more than doubled for short women (46.3%) compared to tall women (21.7%), in agreement with earlier findings. For women of average height carrying a heavy newborn while having a relatively short compared to tall partner reduced the risk by 6.7%. In conclusion, the size of the baby, the height of the mother and parental height differences affect the likelihood of an ECS in primiparous women. 相似文献
