The C-11=C-12 double bond of the retinylidene chromophore of rhodopsin holds a central position in its light-induced photoisomerization and hence the photosensory function of this visual pigment. To probe the local environment of the HC-11=C-12H element we have prepared the 11-methyl and 12-methyl derivatives of 11-Z retinal and incorporated these into opsin to generate the rhodopsin analogs 11-methyl and 12-methyl rhodopsin. These analog pigments form with much slower kinetics and lower efficiency than the native pigment. The initial photochemistry and the signaling activity of the analog pigments were investigated by UV-vis and FTIR spectroscopy, and by a G protein activation assay. Our data indicate that the ultrafast formation of the first photointermediate is strongly perturbed by the presence of an 11-methyl substituent, but much less by a 12-methyl substituent. These results support the current concept of the mechanism of the primary photoisomerization event in rhodopsin. An important stronghold of this concept is an out-of-plane movement of the C-12H element, which is facilitated by torsion as well as extended positive charge delocalization into the C-10-C-13 segment of the chromophore. We argue that this mechanism is maintained principally with a methyl substituent at C-12. In addition, we show that both an 11-methyl and a 12-methyl substitutent perturb the photointermediate cascade and finally yield a low-activity state of the receptor. The 11-methyl pigment retains about 30% of the G protein activation rate of native rhodopsin, while the 12-methyl chromophore behaves like an inverse agonist up to at least 20 degrees C, trapping the protein in a perturbed Meta-I-like conformation. We conclude that the isomerization region of the chromophore and the spatial structure of the binding site are finely tuned, in order to achieve a high photosensory potential with an efficient pathway to a high-activity state.  相似文献

Background

The risk of premature cardiovascular disease in patients with familial hypercholesterolemia (FH) can be profoundly reduced by cholesterol-lowering therapy, and current guidelines for FH advocate ambitious low-density lipoprotein cholesterol (LDL-C) goals. In the present study, we determined whether these goals are reflected in current clinical practice once FH has been diagnosed.

Methodology/Principal Findings

In 2008, we sent questionnaires to all subjects (aged 18–65 years) who were molecularly diagnosed with FH in the year 2006 through the screening program in the Netherlands. Of these 1062 subjects, 781 completed the questionnaire (46% males; mean age: 42±12 years; mean LDL-C at molecular diagnosis (baseline): 4.1±1.3 mmol/L). The number of persons that used cholesterol-lowering therapy increased from 397 (51%) at baseline to 636 (81%) after diagnosis. Mean treated LDL-C levels decreased significantly to 3.2±1.1 mmol/L two years after diagnosis. Only 22% achieved the LDL-C target level of ≤2.5 mmol/L.

Conclusions/Significance

The proportion of patients using cholesterol-lowering medication was significantly increased after FH diagnosis through genetic cascade screening. The attained LDL-C levels were lower than those reported in previous surveys on medication use in FH, which could reflect the effect of more stringent lipid target levels. However, only a minority of the medication users reached the LDL-C target.  相似文献

AimAvailability of uniformly collected presence, absence, and abundance data remains a key challenge in species distribution modeling (SDM). For invasive species, abundance and impacts are highly variable across landscapes, and quality occurrence and abundance data are critical for predicting locations at high risk for invasion and impacts, respectively. We leverage a large aquatic vegetation dataset comprising point‐level survey data that includes information on the invasive plant Myriophyllum spicatum (Eurasian watermilfoil) to: (a) develop SDMs to predict invasion and impact from environmental variables based on presence–absence, presence‐only, and abundance data, and (b) compare evaluation metrics based on functional and discrimination accuracy for presence–absence and presence‐only SDMs.LocationMinnesota, USA.MethodsEurasian watermilfoil presence–absence and abundance information were gathered from 468 surveyed lakes, and 801 unsurveyed lakes were leveraged as pseudoabsences for presence‐only models. A Random Forest algorithm was used to model the distribution and abundance of Eurasian watermilfoil as a function of lake‐specific predictors, both with and without a spatial autocovariate. Occurrence‐based SDMs were evaluated using conventional discrimination accuracy metrics and functional accuracy metrics assessing correlation between predicted suitability and observed abundance.ResultsWater temperature degree days and maximum lake depth were two leading predictors influencing both invasion risk and abundance, but they were relatively less important for predicting abundance than other water quality measures. Road density was a strong predictor of Eurasian watermilfoil invasion risk but not abundance. Model evaluations highlighted significant differences: Presence–absence models had high functional accuracy despite low discrimination accuracy, whereas presence‐only models showed the opposite pattern.Main conclusionComplementing presence–absence data with abundance information offers a richer understanding of invasive Eurasian watermilfoil''s ecological niche and enables evaluation of the model''s functional accuracy. Conventional discrimination accuracy measures were misleading when models were developed using pseudoabsences. We thus caution against the overuse of presence‐only models and suggest directing more effort toward systematic monitoring programs that yield high‐quality data.  相似文献

Long‐distance migratory flights are predicted to be associated with higher mortality rates when individuals encounter adverse weather conditions. However, directly connecting environmental conditions experienced in‐flight with the survival of migrants has proven difficult. We studied how the in‐flight mortality of 53 satellite‐tagged Black‐tailed Godwits (Limosa limosa limosa) during 132 crossings of the Sahara Desert, a major geographical barrier along their migration route between The Netherlands and sub‐Saharan Africa, is correlated with the experienced wind conditions and departure date during both southward and northward migration. We show that godwits experienced higher wind assistance during southward crossings, which seems to reflect local prevailing trade winds. Critically, we found that fatal northward crossings (15 deaths during 61 crossings) were associated with adverse wind conditions. Wind conditions during migration can thus directly influence vital rates. Changing wind conditions associated with global change may thus profoundly influence the costs of long‐distance migration in the future.  相似文献

Restoration and conservation of species-rich nature reserves requires inclusion of landscape-scale connections and transport processes such as hydrologic flows and species dispersal. These are important because they provide suitable habitat conditions and an adequate species pool. This study aimed at identifying the key hydrologic flows and plant dispersal processes affecting a landscape with species-rich fen reserves where restoration measures are carried out to set back succession. It also intended to use this information for delineating the area relevant for conservation planning on an Operational Landscape Unit map. The study was carried out for complexes of fen ponds in former turbaries in the Vechtplassen area, The Netherlands. A number of recent insights on plant dispersal were integrated with knowledge on hydrologic flows in the present approach. The results showed that groundwater discharge to ensure mesotrophic, base-rich conditions, should be enhanced by restoring the groundwater recharge areas NE of the reserves. A nearby lake with suitable water chemistry was also identified as a key source of surface water to feed the fens in dry periods. Water dispersal was identified as important within the fen reserves, whereas dispersal by daily migrating dabbling ducks, typically occurring over 2–3 km, was the most important route connecting the reserves with the surrounding landscape. The delineation of the Operational Landscape Unit for this region provides a basis for conservation and restoration that take fundamental landscape connections and transport processes into account. This unique approach simultaneously considers hydrological transport processes as well as species dispersal in the larger landscape beyond the reserves themselves and therefore leads to greater success of restoration and conservation.  相似文献

Sexual size dimorphism (SSD) is common in birds and has been linked to various selective forces. Nevertheless, the question of how and when the sexes start to differentiate from each other is poorly studied. This is a critical knowledge gap, as sex differences in growth may cause different responses to similar ecological conditions. In this study, we describe the sex‐specific growth – based on body mass and five morphometric measurements – of 56 captive Black‐tailed Godwit Limosa limosa limosa chicks raised under ad libitum food conditions, and conclude that all six growth curves are sex‐specific. Females are the larger sex in terms of body mass and skeletal body size. To test whether sex‐specific growth leads to sex‐specific susceptibility to environmental conditions, we compared the age‐specific sizes of male and female chicks in the wild with those of Black‐tailed Godwits reared in captivity. We then tested for a relationship between residual growth and relative hatching date, age, sex and habitat type in which the wild chicks were born. Early‐hatched chicks were relatively bigger and in better condition than late‐hatched chicks, but body condition and size were not affected by natal habitat type. Female chicks deviated more negatively from the sex‐specific growth curves than male chicks for body mass and total‐head length. This suggests that the growth of the larger females is more susceptible to limiting environmental conditions. On average, the deviations of wild chicks from the predicted growth curves were negative for all measurements, which suggests that conditions are limiting in the current agricultural landscape. We argue that in estimating growth curves for sexually dimorphic species, it is critical first to make accurate sex and age determinations.  相似文献
