Background
Inflammatory stimuli induce the hepatic iron regulatory hormone hepcidin, which contributes to anaemia of inflammation (AI). Hepcidin expression is regulated by the bone morphogenetic protein (BMP) and the interleukin-6 (IL-6) signalling pathways. Prior results indicate that the BMP type I receptor ALK3 is mainly involved in the acute inflammatory hepcidin induction four and 72 h after IL-6 administration. In this study, the role of ALK3 in a chronic model of inflammation was investigated. The intact, heat-killed bacterium Brucella abortus (BA) was used to analyse its effect on the development of inflammation and hypoferremia in mice with hepatocyte-specific Alk3-deficiency (Alk3fl/fl; Alb-Cre) compared to control (Alk3fl/fl) mice.Results
An iron restricted diet prevented development of the iron overload phenotype in mice with hepatocyte-specific Alk3 deficiency. Regular diet leads to iron overload and increased haemoglobin levels in these mice, which protects from the development of AI per se. Fourteen days after BA injection Alk3fl/fl; Alb-Cre mice presented milder anaemia (Hb 16.7 g/dl to 11.6 g/dl) compared to Alk3fl/fl control mice (Hb 14.9 g/dl to 8.6 g/dl). BA injection led to an intact inflammatory response in all groups of mice. In Alk3fl/fl; Alb-Cre mice, SMAD1/5/8 phosphorylation was reduced after BA as well as after infection with Staphylococcus aureus. The reduction of the SMAD1/5/8 signalling pathway due to hepatocyte-specific Alk3 deficiency partly suppressed the induction of STAT3 signalling.Conclusion
The results reveal in vivo, that 1) hepatocyte-specific Alk3 deficiency partly protects from AI, 2) the development of hypoferremia is partly dependent on ALK3, and 3) the ALK3/BMP/hepcidin axis may serve as a possible therapeutic target to attenuate AI.962.

Single particle levitation (SPL) is used to measure the sorption and desorption of water vapour from microparticles comprising of Bacillus spores. Water gain is determined from increases in the weight-balancing levitation voltages. Spore water isotherms are compared to values previously reported using bulk gravimetric methods. Two salient differences are found between the SPL and bulk data: (1) greater osmotic swelling is observed in the SPL data; and (2) the SPL data exhibit open loop hysteresis while bulk data exhibit closed loop hysteresis. 相似文献

The concentrations of Ca, Sr, Ba and Mn were determined in up to five successive needle age classes from 54 individual Norway spruce trees [Picea abies (L.) Karst] from nine different sites. The accumulation behaviour was found to be very nonuniform, going from an increase with needle age to a decrease; irregular patterns were also found. The type of accumulation is largely site specific. The increasing behaviour can in most cases be approximated by a simple arithmetic function. All four elements usually show the same accumulation pattern, the similarities being closest between Ca and Mn and least between Ca and Ba. It is postulated that the similarity between the four elements is due to their precipitation and storage as oxalates. The similarity between Ca, Sr and Ba is observed at all concentrations, that with Mn only at concentrations larger than 300 g/g. Mn at small concentrations (< 50 g/g) shows a decreasing pattern and no similarity at all with Ca, Sr and Ba, but behaves similar to mobile elements. 相似文献

Fatty acid methyl ester analysis (FAME) by gas chromatography coupled to mass spectrometry (GC‐MS) is a widely used technique in biodiesel/bioproduct (e.g. poly‐unsaturated fatty acids, PUFA) research but typically does not allow distinguishing between bound and free fatty acids. To understand and optimize biosynthetic pathways, however, the origin of the fatty acid is an important information. Furthermore the annotation of PUFAs is compromised in classical GC‐EI‐MS because the precursor molecular ion is missing. In the present protocol an alkaline methyl esterification step with TMS derivatization enabling the simultaneous analysis of bound and free fatty acids but also further lipids such as sterols in one GC‐MS chromatogram is combined. This protocol is applied to different lipid extracts from single cell algae to higher plants: Chlorella vulgaris, Chlamydomonas reinhardtii, Coffea arabica, Pisum sativum and Cuscuta japonica. Further, field ionization (GC‐FI‐MS) is introduced for a better annotation of fatty acids and exact determination of the number of double bonds in PUFAs. The proposed workflow provides a convenient strategy to analyze algae and other plant crop systems with respect to their capacity for third generation biodiesel and high‐quality bioproducts for nutrition such as PUFAs. 相似文献

The aim of the experiment on 100 cross-bred barrows was to compare commercial diets for fattening pigs based on either soya bean meal (SBM) imported from non-European countries with diets based on a mixture of locally produced rape seed meal, distillers’ dried grains with solubles and soya beans as main protein sources. In addition, these both types of diets were processed by two different technical feed treatments, i.e. coarse grinding without hydrothermal treatment or fine grinding and pelleting. With only few exceptions, nutrients of the diet without SBM were more digestible (p < 0.05) resulting in a higher metabolisable energy (ME) content. Fine grinding and pelleting increased also the ME content and the nutrient digestibility with the exception of crude fibre. Higher feed intake of animals that fed diets without SBM (p < 0.01) resulted in higher average daily gain (p < 0.01). However feeding this diet, the higher digestibility was not reflected in a decreased feed-to-gain ratio (FGR), but fine grinding and pelleting reduced FGR (p < 0.001). A higher pH value and a lower DM content of caecal chymus were detected in animals that received coarsely ground feed (p < 0.05). Animals that fed finely ground and pelleted feed had higher slaughter and relative liver weights and higher blood cholesterol concentrations (p = 0.040). The urea concentrations of blood were lower (p = 0.019) after feeding diets without SBM. In conclusion, SBM imported from non-European countries can be replaced by alternative local protein sources without compromising digestibility or performances of animals. Although fine grinding and thermal treatment particularly seemed to be advantageous for digestibility and performance, the possible risk of development of stomach lesions should be considered. 相似文献
