Minimally invasive fetal interventions require accurate imaging from inside the uterine cavity. Twin‐to‐twin transfusion syndrome (TTTS), a condition considered in this study, occurs from abnormal vascular anastomoses in the placenta that allow blood to flow unevenly between the fetuses. Currently, TTTS is treated fetoscopically by identifying the anastomosing vessels, and then performing laser photocoagulation. However, white light fetoscopy provides limited visibility of placental vasculature, which can lead to missed anastomoses or incomplete photocoagulation. Photoacoustic (PA) imaging is an alternative imaging method that provides contrast for hemoglobin, and in this study, two PA systems were used to visualize chorionic (fetal) superficial and subsurface vasculature in human placentas. The first system comprised an optical parametric oscillator for PA excitation and a 2D Fabry‐Pérot cavity ultrasound sensor; the second, light emitting diode arrays and a 1D clinical linear‐array ultrasound imaging probe. Volumetric photoacoustic images were acquired from ex vivo normal term and TTTS‐treated placentas. It was shown that superficial and subsurface branching blood vessels could be visualized to depths of approximately 7 mm, and that ablated tissue yielded negative image contrast. This study demonstrated the strong potential of PA imaging to guide minimally invasive fetal therapies.   相似文献

Limited information is currently available on molecular events that underlie schizophrenia-like behaviors in animal models. Accordingly, we developed an organelle proteomic approach enabling the study of neurotransmission-related proteins in the prefrontal cortex (PFC) of postpubertal (postnatal day 60 (PD60)) neonatally ventral hippocampal (nVH) lesioned rats, an extensively used neurodevelopmental model of schizophrenia-like behaviors. The PFC was chosen because of its purported role in the etiology of the disease. Statistical analysis of 392 reproducible spots on 2-D organelle proteomic patterns revealed significant changes in intensity of 18 proteinous spots in plasma membrane-enriched fractions obtained from postpubertal nVH lesioned rats compared to controls. Mass spectrometric analysis and database searching allowed the identification of a single protein in each of the nine differential spots, including proteins of low abundance, such as neurocalcin delta. Most of the identified dysregulated proteins, including clathrin light chain B, syntaxin binding protein 1b and visinin-like protein 1 are known to be linked to various neurotransmitter systems and to play key roles in plasma membrane receptor expression and recycling as well as synaptic vesicle exocytosis/recycling. Organelle proteomic approaches have hence proved to be most useful to identify key proteins linked to a given behavior in animal models of brain diseases.  相似文献

Tolerance to high levels of ethanol is an ecologically and industrially relevant phenotype of microbes, but the molecular mechanisms underlying this complex trait remain largely unknown. Here, we use long-term experimental evolution of isogenic yeast populations of different initial ploidy to study adaptation to increasing levels of ethanol. Whole-genome sequencing of more than 30 evolved populations and over 100 adapted clones isolated throughout this two-year evolution experiment revealed how a complex interplay of de novo single nucleotide mutations, copy number variation, ploidy changes, mutator phenotypes, and clonal interference led to a significant increase in ethanol tolerance. Although the specific mutations differ between different evolved lineages, application of a novel computational pipeline, PheNetic, revealed that many mutations target functional modules involved in stress response, cell cycle regulation, DNA repair and respiration. Measuring the fitness effects of selected mutations introduced in non-evolved ethanol-sensitive cells revealed several adaptive mutations that had previously not been implicated in ethanol tolerance, including mutations in PRT1, VPS70 and MEX67. Interestingly, variation in VPS70 was recently identified as a QTL for ethanol tolerance in an industrial bio-ethanol strain. Taken together, our results show how, in contrast to adaptation to some other stresses, adaptation to a continuous complex and severe stress involves interplay of different evolutionary mechanisms. In addition, our study reveals functional modules involved in ethanol resistance and identifies several mutations that could help to improve the ethanol tolerance of industrial yeasts.  相似文献

We studied the habitat use and mobility of the Grayling butterfly (Hipparchia semele) and the Blue-Winged Grasshopper (Oedipoda caerulescens), two threatened insects within spatially structured blond and grey dunes in a nature reserve along the Belgian coast. Although both species occur in the same biotope, H. semele were more abundant in open, dynamic sites with a relatively high amount of bare sand, while O. caerulescens preferred sheltered, more stable environments with a lower amount of bare sand. Unlike H. semele, substrate use varied in accordance to body colouration in O. caerulescens, especially on cold days, with light-coloured animals being more abundant on sand and dark-coloured animals more abundant on moss. During a mark-recapture-study, we marked 493 Grayling butterflies and 1289 Blue-Winged Grasshoppers. On average, both sexes of H. semele were equally mobile (about 150 m/day; maximum recorded distance of about 1700 m) while male O. caerulescens were significantly more mobile than females (daily average 47 vs. 5 m; maximum distances observed for O. caerulescens were about 800 m). The importance of habitat heterogeneity (within and among patches) and the consequences of habitat use and mobility of both species for the conservation of typical coastal dune habitats are discussed. The complementary use of species-specific information to site-based management measures is advocated.  相似文献

Migration counts can offer a cost-effective method for monitoring the state of migrant raptor populations. However, differential migration strategies between inexperienced juveniles and experienced non-juveniles are rarely accounted for when inferring population trends from raptor migration counts. Since 2011, the Batumi Raptor Count (BRC) monitors the autumn migration of more than 1 million raptors along the eastern Black Sea coast in the Republic of Georgia. We also systematically sampled age information to assess differential migration timing between age groups and estimate age-specific linear trends in abundance between 2011 and 2018 for eight focal species. In so doing we aimed (1) to reassess the global relevance of BRC counts for each species and the potential for monitoring abundance of juveniles and non-juveniles, and (2) to identify demographic changes underlying recent trends in overall abundance. We found that the mean annual passage of non-juveniles at Batumi represents at least 1% of the estimated global breeding population of five study species. As expected, counts of juveniles were more variable than counts of non-juveniles. Yet despite our short monitoring period our models had sufficient statistical power to detect changes in abundance of 10%/year or less for at least one age group in all species except Pallid Harrier Circus macrourus. Our results indicate stable abundance and demography for half of the study species. We also found strong and significant increases in the abundance of Black Kites Milvis migrans and Short-toed Eagles Circaetus gallicus that were primarily due to increasing numbers of non-juveniles. By contrast, juvenile Montagu's Harriers Circus pygargus and Booted Eagles Hieraaetus pennatus significantly decreased in abundance. The first decade of BRC surveys offers an important benchmark for monitoring raptor populations using the eastern African–Palearctic flyway in the 21st century. We discuss possible causes of the observed trends and hope our work will stimulate demographic monitoring at migration count sites.  相似文献

Human activities impact upon natural habitats used by birds for breeding and foraging, and lead to changes in the composition and spatial distribution of predator communities, mainly through loss, fragmentation and disturbance of formerly pristine habitat. Yet possible fitness consequences of such changes through impacts on bird nest-site selection remain poorly known. Here we study nest-site selection and reproductive success of Placid Greenbuls Phyllastrephus placidus in the Taita Hills, southeast Kenya. We show that habitat features associated with nest-site selection by this insectivorous, open-cup-nesting bird species vary among forest fragments that are exposed to different levels of habitat disturbance. Such differences in sites selected for breeding result from a plastic response to fragment-specific conditions or may be driven by fragment-specific variation in the distribution and availability of certain habitat features. Given the overall high nest predation rates in our study area, we expected variation in nest-site selection to correlate with reproductive success and nestling condition, but detected no such relationship. Because predator density and nest predation rates may vary strongly in space and time, a better understanding of spatio-temporal variation in predator communities is needed to assess the possible adaptive value of nest-site selection strategies for reducing the high predation rates that are typical for this and many other open-cup-nesting tropical passerines.  相似文献
