We have isolated a gene, AAT1, encoding an aspartate aminotransferase (AspAT) from a Saccharomyces cerevisiae genomic library. AAT1 encodes a 451 amino acid protein with a predicted molecular weight of 51,687, which is likely to be the yeast mitochondrial AspAT. Sequence comparison of this yeast AspAT with AspATs from other organisms shows a high degree of homology in regions previously shown to be important for catalysis. However, the yeast mitochondrial AspAT contains four obvious insertions with respect to all other known AspATs, suggesting that the AAT1-encoded protein represents a distinct AspAT. 相似文献

Insulin binding to the plasma membrane is known to be altered by modifying the membrane composition through dietary treatment. As insulin binding receptors are also present on nuclear membrane, this study was undertaken to investigate if specific binding of insulin to the liver nuclei is altered by diet. 8-wk-old female C57 B 6J lean and ob/ob mice were fed semipurified diets containing 20% (w/w) fat of either high or low polyunsaturated-to-saturated (P/S) fatty acid ratio for 4 wk. Liver nuclei were prepared, insulin binding was measured and nuclear phospholipids were isolated for lipid analysis. Insulin binding was highest in nuclei prepared from lean mice fed a high P/S diet. Specific binding of insulin to nuclei prepared from obese mice was also increased by the high P/S diet, but to a lesser extent compared to lean mice. Feeding a high P/S diet increased polyunsaturated fatty acid content of membrane phospholipids from both lean and ob/ob mice. Obese mice were characterized by higher levels of arachidonic acid and lower levels of linoleic acid in phosphatidylcholine. The present study establishes that insulin binding to liver nuclei is increased by feeding a high P/S diet, and that insulin binding to liver nuclei from obese mice is lower than from lean mice. 相似文献

N-1-Naphthylphthalmic acid (NPA)-binding protein is a plasmalemma (PM) protein involved in the control of cellular auxin efflux. We re-evaluated the spatial relationship of this protein with the PM of zucchini (Cucurbita pepo L.) hypocotyls. First, Triton X-114 partitioning indicated that the NPA-binding protein was more hydrophobic than most PM proteins. Second, the NPA-binding activity was found to be resistant to proteolytic digestion in membranes. Maximum concentrations of binding sites for NPA were virtually identical in untreated and proteinase K-treated PMs: 19.2 and 20.6 pmol [3H]NPA bound/mg protein, respectively. The insensitivity of the NPA-binding protein was not due to its presence inside tightly sealed vesicles or due to lack of protease activity in the conditions tested. This protein could be made sensitive to proteolytic degradation upon solubilization by 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate in the presence of sodium molybdate. Proteinase K treatment decreased the concentration of binding sites to 0.84 pmol [3H]NPA bound/mg protein from 9.2 for untreated, solubilized PM. Third, this activity could not be solubilized by chaotropic agents or sodium carbonate treatment of intact PM. This study indicates that the NPA-binding protein may be an integral membrane protein and contradicts previously reported findings that suggested that this protein was peripheral to the PM. 相似文献

The site of action of hydantocidin was probed using Arabidopsis thaliana plants growing on agar plates. Herbicidal effects were reversed when the agar medium was supplemented with AMP, but not IMP or GMP, suggesting that hydantocidin blocked the two-step conversion of IMP to AMP in the de novo purine biosynthesis pathway. Hydantocidin itself did not inhibit adenylosuccinate synthetase or adenylosuccinate lyase isolated from Zea mays. However, a phosphorylated derivative of hydantocidin, N-acetyl-5'-phosphohydantocidin, was a potent inhibitor of the synthetase but not of the lyase. These results identify the site of action of hydantocidin and establish adenylosuccinate synthetase as an herbicide target of commercial potential. 相似文献

The crystal structure of a dipeptide L -leucyl–L -leucine (C12H24N2O3) has been determined. The crystals are monoclinic, space group P21, with a = 5.434(4) Å, b = 15.712(7) Å, c = 11.275(2) Å, β = 100.41(1)°, and Z = 2. The crystals contain one molecule of dimethyl sulfoxide (DMSO) as solvent of crystallization for each dipeptide molecule. The structure has been solved by direct methods and refined to a final R index of 0.059 for 920 reflections (sinθ/λ ? 0.60 Å?1) with I ? 2σ (I). The trans peptide unit shows substantial degree of non-planarity (Δω = 14°). The peptide backbone adopts an extended conformation with torsion angles of ψ1 = 138(1)°, ω1 = 166(1)°, ?2 = ? 149.3(7)°, ψ21 = 164.2(7)°, and ψ22 = ? 15(1)°. For the first leucyl residue, the side-chain conformation is specified by the torsion angles 1χ1 = 176.7(7)°, 1χ21 = 62(1)°, 1χ22 = ? 177.4(8)°; the second leucyl residue adopts a Sterically unfavorable conformation with 2χ1 = 61(1)°, 2χ21 = 97(1)°, and 2χ22 = ?151(1)°. The packing involves head-to-tail interaction of peptide molecules and segregation of polar and nonpolar regions. Single 2h administration of diazepam (benzodiazepine) in 3.5% ethanol solution was found to evoke advance and delay phase
shifts in the locomotor activity rhythm in the field mouseMus booduga. Through such pulsed administration of diazepam at various phases of circadian rhythm a phase response curve could be constructed.
Phase advance occurred during early subjective day (CT 2) and phase delays were observed in the remaining phases. The shape
of the diazepam phase response curve is similar to the general shape of the phase response curves generated by intraperitoneal
injections of other benzodiazepines in hamsters. The phase shifting action of diazepam may be explained by its agonistic action

Adult male rodents have a pulsatile profile of growth hormone (GH) release, whereas female rodents have a relatively steady-state pattern with uniform, albeit lower levels of GH. The expression of a number of sexually differentiated hepatic proteins is primarily determined by these plasma GH profiles and only secondarily regulated by gonadal hormones. An important subset of these sexually dimorphic proteins is cytochrome P450s. CYP3A10/6 beta-hydroxylase is a cytochrome P450 that catalyzes the 6 beta-hydroxylation of lithocholic acid. CYP3A10/6 beta-hydroxylase is expressed only in male hamsters; however, mimicking the male GH secretion pattern in females induces expression of the gene to male levels. Using chimeric CYP3A10/6 beta-hydroxylase promoter/luciferase reporter genes transfected into hamster primary hepatocytes, we have shown a GH-mediated induction of promoter activity. A combination of 5'-deletion constructs, heterologous promoter constructs, and specific mutagenesis was used to localize the DNA element involved in the GH-mediated regulation of CYP3A10/6 beta-hydroxylase promoter activity, which resembles a STAT binding site. Footprint and gel shift analyses confirmed that the expression of the protein binding to this site is regulated by GH and that the DNA-protein complex can be partially supershifted by anti-STAT-5 antibodies. This protein is 50% more abundant in male than in female hamster livers, is absent in hypophysectomized female livers, and is restored when hypophysectomized females are injected with GH in a manner that masculinizes female hamsters in terms of CYP3A10/6 beta-hydroxylase expression. The system characterized and described here is ideally suited for dissecting the molecular details governing the sexually dimorphic expression of liver-specific genes. 相似文献

The effects of root colonization by the arbuscular mycorrhizal (AM) fungus Glomus intraradices Schenck & Smith on nutritional, growth, and reproductive attributes of two tropical maize cultivars with different sensitivities to drought were studied. Freshly regenerated seeds of selection cycles 0 (cv. C0, drought-sensitive) and 8 (cv. C8, drought-resistant) of the lowland tropical maize population "Tuxpeño sequía" were used in this greenhouse experiment. Maize plants were subjected to drought stress for 3 weeks following tasselling (75–95 days after sowing) and rewatered for the subsequent 5 weeks until harvest. Mycorrhizal (M+) plants had significantly higher uptake of N, P, K, Mg, Mn, and Zn into grain than non-mycorrhizal (M–) plants under drought conditions. AM inoculation also produced significantly greater shoot masses in C0 and C8 regardless of the drought-stress treatment. In the sensitive cultivar C0, drought stress reduced the shoot mass and grain yield by 23% and 55%, respectively, when roots were not colonized, while the reductions were only 12% and 31%, respectively, with mycorrhizal association. In addition, the emergence of tassels and silks was earlier in M+ plants than in M– plants under drought conditions. Mycorrhizal response was more pronounced under both well-watered and drought conditions in C0 than in the C8 cultivar. The overall results suggest that AM inoculation affects host plant nutritional status and growth and thereby alters the reproductive behaviour of maize under drought conditions. 相似文献
