The genus Pseudoacanthocephalus Petrochenko, 1958 currently includes 14 species of acanthocephalans parasitic in amphibians and reptiles worldwide. This work describes two new species of Pseudoacanthocephalus from amphibians and reptiles collected in several localities on Luzon Island, Philippines. Pseudoacanthocephalus nickoli n. sp. was found in two species of frogs, Rana luzonensis Boulenger and Rana similis (Günther), and Pseudoacanthocephalus smalesi n. sp. was found in a scincid lizard, Sphenomorphus abdictus Brown & Alcala. Differential diagnoses of the two new species of Pseudoacanthocephalus from their congeners are provided. Comparative analysis of nuclear ribosomal rRNA sequences encompassing the 3′ end of 18S nuclear rDNA gene, internal transcribed spacer region (ITS1+5.8S+ITS2), and 5′ end of the 28S gene strongly corroborated the morphological evidence and demonstrated significant differences between the two new species as well as between these species and closely related species from continental China and Vietnam. No intraspecific sequence variability was detected among different individuals representing each of the examined species. This is the first report of Pseudoacanthocephalus in the Philippines. A key to known species of Pseudoacanthocephalus is provided.  相似文献

Neosychnocotyle maggiae, n. gen., n. sp., (Aspidogastrea) is described from the pig-nosed turtle, Carettochelys insculpta, and reported from the Victoria River red-faced turtle, Emydura victoriae, all from the Daly River, Northern Territory, Australia. Neosychnocotyle n. gen. is differentiated from all aspidogastrean genera but one by the absence of a cirrus sac. The similar Sychnocotyle also lacks a cirrus sac, but Neosychnocotyle n. gen. differs from the former genus by possessing a narrow, tapered anterior end, a ventral disk that covers the posterior end of the body, a genital pore that is displaced anteriorly, and a vas deferens that is less convoluted and less robust. Analysis of ribosomal DNA sequences demonstrates substantial sequence variability between representatives of Sychnocotyle and Neosychnocotyle maggiae, n. gen., n. sp. It is possible that this new parasite may reach sexual maturity only in C. insculpta. This is only the second aspidogastrean species reported from Australian freshwater turtles.  相似文献

Dieldrin is an organochlorine pesticide that is toxic for monoaminergic neurons. This study was designed to test the hypothesis that a weak DNA repair response to dieldrin by nigrostriatal dopaminergic (DA) neurons results in depletion of striatal DA. The activity of the mammalian base excision repair enzyme oxyguanosine glycosylase was utilized as the index of DNA repair. Other measures of oxidative stress were also studied, including the regional distribution of lipid peroxidation and superoxide dismutase (SOD) activity. The effects of acute and slow infusion of dieldrin on striatal DA levels were biphasic with a transient initial depression followed by increases beyond normal steady-:state levels. Dieldrin administration caused a global oxidative stress evidenced by increased levels of lipid peroxidation in all brain regions, an effect consistent with its capacity to affect mitochondrial bioenergetics. Dieldrin also elicited strong antioxidative and DNA repair responses across the entire mouse brain. Although mitochondrial SOD was not as increased in midbrain as it was in other regions following a cumulative dose of 24 mg/kg, this response, along with the robust DNA repair response, appeared to be sufficient to protect potentially vulnerable DA neurons from cytotoxicity. However, the long-:term consequences of chronic low-:dose dieldrin exposure remain to be studied, especially in light of the concept of "slow excitotoxicity,' which postulates that even a mild bioenergetic compromise can over time result in the demise of neurons.  相似文献

To monitor the lateral segregation of lipids into liquid-ordered (Lo) and -disordered (Ld) phases in lipid membranes, environment-sensitive dyes that partition in both phases but stain them differently have been developed. Of particular interest is the dual-color F2N12S probe, which can discriminate the two phases through the ratio of its two emission bands. These bands are associated with the normal (N) and tautomer (T) excited-state species that result from an excited-state intramolecular proton transfer. In this work, we investigated the potency of the time-resolved fluorescence parameters of F2N12S to discriminate lipid phases in model and cell membranes. Both the long and mean lifetime values of the T form of F2N12S were found to differ by twofold between Ld and Lo phases as a result of the restriction in the relative motions of the two aromatic moieties of F2N12S imposed by the highly packed Lo phase. This differed from the changes in the ratio of the two emission bands between the two phases, which mainly resulted from the decreased hydration of the N form in the Lo phase. Importantly, the strong difference in lifetimes between the two phases was preserved when cholesterol was added to the Ld phase. The two phases could be imaged with high contrast by fluorescence lifetime imaging microscopy (FLIM) on giant unilamellar vesicles. FLIM images of F2N12S-labeled live HeLa cells confirmed that the plasma membrane was mainly in the Lo-like phase. Furthermore, the two phases were found to be homogeneously distributed all over the plasma membrane, indicating that they are highly mixed at the spatiotemporal resolution of the FLIM setup. Finally, FLIM could also be used to sensitively monitor the change in lipid phase upon cholesterol depletion and apoptosis.  相似文献

Kurilonema browni n. sp. is described on the basis of specimens found in the lungs of the scincid lizard Sphenomorphus abdictus aquilonius from Aurora Province, Luzon Island, Philippines. The new species differs from Kurilonema markovi, the only previously known species in the genus, by the presence of 2 prominent lateral pseudolabia, larger body size, shorter tail length relative to total body length, and more numerous eggs in the uteri, containing fully developed larvae. The inner surface of the buccal capsule in the new species is densely covered with rounded scales that have not been observed in K. markovi or any other rhabdiasid species. This is the first record of the genus from the Philippines.  相似文献

Cytoplasmic tyrosyl-tRNA synthetase (TyrRS) is one of the key enzymes of protein biosynthesis. TyrRSs of pathogenic organisms have gained attention as potential targets for drug development. Identifying structural differences between various TyrRSs will facilitate the development of specific inhibitors for the TyrRSs of pathogenic organisms. However, there is a deficiency in structural data for mammalian cytoplasmic TyrRS in complexes with substrates. In this work, we constructed spatial structure of full-length Bos taurus TyrRS (BtTyrRS) and its complexes with substrates using the set of computational modeling techniques. Special attention was paid to BtTyrRS complexes with substrates [L-tyrosine, K+ and ATP:Mg2+] and intermediate products [tyrosyl-adenylate (Tyr-AMP), K+ and PPi:Mg2+] with the different catalytic loop conformations. In order to analyze their dynamical properties, we performed 100 ns of molecular dynamics (MD) simulations. MD simulations revealed new structural data concerning the tyrosine activation reaction in mammalian TyrRS. Formation of strong interaction between Lys154 and γ-phosphate suggests the additional role of CP1 insertion as an important factor for ATP binding. The presence of a potassium-binding pocket within the active site of mammalian TyrRS compensates the absence of the second lysine in the KMSKS motif. Our data provide new details concerning a role of K+ ions at different stages of the first step of the tyrosylation reaction, including the coordination of substrates and involvement in the PPi releasing. The results of this work suggest that differences between ATP-binding sites of mammalian and bacterial TyrRSs are meaningful and could be exploited in the drug design.  相似文献

Phylogenetic interrelationships of 32 species belonging to 18 genera and four families of the superfamily Microphalloidea were studied using partial sequences of nuclear lsrDNA analysed by Bayesian inference and maximum parsimony. The resulting trees were well resolved at most nodes and demonstrated that the Microphalloidea, as represented by the present data-set, consists of three main clades corresponding to the families Lecithodendriidae, Microphallidae and Pleurogenidae + Prosthogonimidae. Interrelationships of taxa within each clade are considered; as a result of analysis of molecular and morphological data, Floridatrema Kinsella & Deblock, 1994 is synonymised with Maritrema Nicoll, 1907, Candidotrema Dollfus, 1951 with Pleurogenes Looss, 1896, and Schistogonimus Lühe, 1909 with Prosthogonimus Lühe, 1899. The taxonomic value of some morphological features, used traditionally for the differentiation of genera within the Lecithodendriidae and Prosthogonimidae, is reconsidered. Previous systematic schemes are discussed from the viewpoint of present results, and perspectives of future studies are outlined.  相似文献

The ultrastructure of the mature spermatozoon of the type genus of the Plagiorchiidae Plagiorchis elegans (Rudolphi, 1802), a parasite of the Golden hamster, Mesocricetus auratus is described. This study is the first ultrastructural study of the spermatozoon of a Plagiorchis, the second of a plagiorchiid species and only the third in the Plagiorchioidea. Previously data on spermatozoon ultrastructure existed only for the plagiorchiid Enodiotrema reductum and the omphalometrid Rubenstrema exasperatum. The mature spermatozoon of P. elegans exhibited the general pattern described in most digenean species, namely two axonemes of the 9 + “1” Trepaxonemata pattern, nucleus, mitochondria, external ornamentation of the plasma membrane, spine‐like bodies, and glycogen granules. However, the rather typical expansion of the plasma membrane is not found in P. elegans. Another peculiarity of the spermatozoon of P. elegans is the presence of a structure called thin cytoplasm termination. Spermatozoon ultrastructure of P. elegans is compared with that of E. reductum and R. exasperatum. Spermatozoon of P. elegans conforms to the general pattern described in E. reductum. Thus, this study further expands our knowledge on the spermatozoon ultrastructure among the members of the Plagiorchioidea, one of the most phylogenetically derived groups of the digenea. J. A new species of Urotrema inhabits Norops oxylophus and Norops cupreus from the Area de Conservación Guanacaste, Costa Rica. The new species is most similar to Urotrema scabridum but differs by having a relatively longer esophagus, at least 10% of the total body length versus 4-8% in U. scabridum; an oral sucker width: ventral sucker width ratio less than 1 : 1 (averaging 1 : 0.76) versus 1 : 1 or greater; an oral sucker width : pharyngeal width ratio less than 1 : 0.5 (averaging 1 : 0.42) versus greater than 1 :0.5; and by having many transversely oriented posttesticular uterine loops versus a few vertically oriented posttesticular uterine loops.  相似文献

1,3- bis-[4-(N-aza-15-crown-5)-benzylidene]cyclopentanone-2 ( I), a ketocyanine dye, was synthesized. The electronic absorption and emission spectra, protolytic interactions in aqueous alcohol media and complex formation with alkali earth metals in acetonitrile were studied for the dye ( I) and for several model compounds. It was shown that protonation of both dialkylamino groups of all compounds studied takes place at closely similar pH values. The complexation of alkali earth metals with azacrown derivative ( I) takes place at azacrown moieties and the carbonyl group. The sequence of binding to these sites is different for Mg2+ and Ba2+ ions. The effective ejection of Ba2+ ion out of the azacrown cycle was observed in the excited state, though, as in the case of Mg2+ ion, such process occurred only partially. The results obtained suggest that this azacrown derivative of dibenzylidene cyclopentanone is sensitive to alkali earth ions and has prospects for different biologically oriented applications.  相似文献
