A new genus and species of centrohelid heliozoan Pinjata ruminata from the Tuzlukkol’ River (Orenburg Region of Russia) and Gor?koe Lake (Chelyabinsk Region of Russia) is studied with light‐ and electron microscopy. Pinjata ruminata has two types of plate scales, partially running up the sides of the axopodia. Inner plate scales (3.2–4.9 × 1.5–2.6 μm) are flat, ovate‐oblong and have a broad axial thickening and a thin electron‐dense border. Outer plate scales (4.2–6.7 × 1.5–3.0 μm) are concave, elongated, of irregular shape, often curved, and broadened towards one end. Roundish depressions are forming two rows on both sides of the narrow axial thickening. The cells are attached to the substratum. Molecular phylogenetic analysis based on the SSU rDNA robustly placed P. ruminata in the family Yogsothothidae. This position is confirmed with the presence of five panacanthocystid increase regions. The morphology of the new genus is in a good accordance with diagnosis of the family. The status of a genus “Heteroraphidiophrys” is discussed. Other potential findings of Pinjata from literature are analyzed. Pinjata represents the third lineage of centrohelids, characterized with the presence of only tangentially oriented plate scales. The halophilic nature of Yogsothothidae is suggested.  相似文献

Here, we report from the first direct observation of a red fox (Vulpes vulpes) intrusion on an arctic fox (Vulpes lagopus) breeding den from the southern Arctic tundra of Yamal Peninsula, Russia in 2007. At the same time, as a current range retraction of the original inhabitant of the circumpolar tundra zone the arctic fox is going on, the red fox is expanding their range from the south into arctic habitats. Thus, within large parts of the northern tundra areas the two species are sympatric which gives opportunities for direct interactions including interference competition. However, direct first-hand observations of such interactions are rare, especially in the Russian Arctic. In the present study, we observed one red fox taking over an arctic fox breeding den which resulted in den abandonment by the arctic fox. On July 19, eight arctic fox pups were observed on the den before the red fox was observed on the same den July 22. The pups were never seen at the den or elsewhere after the red fox was observed on the den for as long as we stayed in the area (until August 10). Our observation supports the view that direct interference with red fox on breeding dens may contribute to the range retraction of arctic foxes from the southern limits of the Arctic tundra in Russia.  相似文献

Epidermis contains a compartment of stem cells but currently there is no common criterion to recognize individual stem cells with any confidence. Epidermis appears to contain stem cells of different levels of maturity and it is very likely that the main repository of epidermal stem cells is located in the hair follicle from which cells can emigrate into epidermis and also give rise to follicular and sebaceous keratinocytes. Epidermis consists of proliferative units containing stem and transit-amplifying cells, but the exact size of a proliferative unit cannot be measured accurately. The available data suggest that populations of stem and transit-amplifying cells are not discrete but represent a continuum from cells with a high self-renewal capacity and a low probability of differentiation to those with low self-renewal capacities and high commitments to differentiation. Stem cells occupy a special niche that provides a microenvironment, including an adhesion of stem cells to the basal membrane and their paracrine interactions with neighbor epidermal and mesenchymal cells. The fate of an epidermal stem cell depends on its prehistory and microenvironment.  相似文献

The effect of light on succinate dehydrogenase (SDH) activity and mRNA content was studied in Arabidopsis thaliana plants. The transition from darkness to light caused a short transient increase in the SDH activity followed by a decrease to a half of the original activity. The white or red light were found to be down-regulating factors for the mRNA content of the sdh1-2 and sdh2-3 genes and SDH catalytic activity both in A. thaliana wild-type plants and in the mutant deficient in the phytochrome B gene, but not in the mutant deficient in the phytochrome A gene, while the far-red light of 730 nm reversed the red light effect. It is concluded that phytochrome A participates in the regulation of mitochondrial respiration through effect on SDH expression.  相似文献

Abstract

Experimentally observed sequence-selective binding of metal ion to DNA oligonucleotides have been compared with variations of electrostatic potential (EP) along the helix. Calculations of EP have been performed for three atomic models of the oligonucleotide duplex [d(CGCGAATTCGCG)2] using several variants of EP calculations, including a solution of non-linear Poisson-Boltzmann equation (NPBE). N7 atom of guanine adjacent to adenine base was identified as a region with the most negative electrostatic potential in the major groove. The EP value for the Me ion binding site surpasses the value for N7 of other guanines by 10–26% depending on particular duplex conformation. Qualitatively, the sequence dependent variations of EP near guanine N7 atoms are in agreement with the sequence-selective behavior of Mn(II) and Zn(II) ions as revealed by NMR experiments. But the difference in EP between the two most negative regions near guanine N7 atoms does not exceed 1.25 kT/e. Simple model suggests that metal ions are capable to form ion-hydrate complexes with G-Pu steps of DNA duplex. These complexes are formed via one Me…G and five Me…water coordination bonds with water molecules hydrogen bonded to two adjacent purine bases in the same chain. We suppose that such a stereospecific structural possibility is the main factor which control the sequence-selectivity in the metal ion binding. A combination of both mechanisms allows to explain sequence specific Mn(II) and Zn(II) binding to a set of oligonucleotides.  相似文献

Enhancers are regulatory DNA sequences that can work over a large distance. Efficient enhancer action over a distance clearly requires special mechanisms for facilitating communication between the enhancer and its target. While the chromatin looping model can explain the majority of the observations, some recent experimental findings suggest that a chromatin scanning mechanism is used to establish the loop. These new findings help to understand the mechanism of action of the elements that can prevent enhancer-promoter communication (insulators).  相似文献

Human embryo fibroblasts were cultured within three-dimensional collagen gel in the Eagle medium supplemented with 10% calf serum. Addition of fibronectin (Fn) to the nutrient medium neither stimulated nor inhibited gel contraction by fibroblasts. Gel contraction also proceeded in the medium containing ultroser G or Fn exhausted calf serum. Addition of 10 or 50 micrograms/ml of Fn to the medium with ultroser G exerted no effect on the process of gel contraction. Addition of antibodies prepared against Fn (70 micrograms/ml) and tetrapeptide RGDS (100 and 500 micrograms/ml) to the medium supplemented with Fn exhausted calf serum did not change the rate of collagen gel contraction. The results obtained suggest that the rate of collagen gel contraction by fibroblasts is not dependent on the presence of exogenous fibronectin.  相似文献

The study of the effects of actinomycin D on stationary cultures of Chinese hamster cells and those stimulated by medium changing has revealed that RNA synthesis is more sensitive in the latter, the difference in the first place being attributed to the rate of labeled uridine incorporation into cell nucleoli. There is no significant difference in H3-actinomycin D uptake between stationary and stimulated cells, but the latter incorporate more labeled actinomycin D into their nuclei. A pronounced variation in sensitivity to actinomycin D is observed during the prereplicative period of stimulated cells. The first part of this period is more sensitive, which may be due to the necessity for stimulated cells to synthesize a great number of ribosomes to enter the mitotic cycle and to proceed through it.  相似文献

Summary Autoradiographic investigation of the cell cycle of 12 diploid and 12 abnormal human fibroblast strains was carried out. Two cell strains (trisomy 7 and monosomy 21) derived from spontaneous abortuses showed prolongation of the G2 period accompanied by the shortening of the S period. Other cytogenetically abnormal embryonic strains (trisomy 9, 14 and triploidy) did not deviate from the diploid pattern. Three cell strains (LHC-1-70, LHC-6-70, LHC-411) derived from the patients with karyotypes 47,XXX, 47,XY,+18 and 46,XX,5p—respectivly had embryonic types of proliferation with a short G2 period. In two other strains from the patients with Down's syndrom the G2 period was prolonged. There was no statistically significant difference in the parameters of the cell cycle between the control and the strains derived from a patient with Klinefelter syndrom and from a male patient with karyotype 46,XX. The modificatory effect of the chromosomal abnormalities on the parameters of cell cycle is discussed.
Zusammenfassung Der Zellcyclus von 12 normal diploiden und 12 abnormen menschlichen Fibroblasten-Zellinien wurde untersucht. Zwei Zellinien (Trisomie 7 und Monosomie 21) zeigten eine Verlängerung der G2-Phase und eine Verkürzung der S-Phase. Zellinien (Trisomie 9, 14 und Triploidie) wichen nicht von der Norm ab. Drei Zellinien (47,XXX; 47,XY,+18; 46,XX,5p-) zeigten eine Verkürzung von G2. G2 war dagegen verlängert bei zwei Zellinien von Down-Syndrom. Keine Abweichung fand sich bei einem Klinefelter-Patienten und einem männlichen Probanden mit 46,XX.

Analysis of behavior of isolated epidermal keratinocytes demonstrated two dominant processes within the first two days of cultivation: formation of cell aggregates and their adhesion to the substrate. Keratinocyte spreading increased in the attached aggregates, where densely packed and well spread cells could be recognized in the aggregate center and periphery, respectively. Cell spreading and proliferation was observed in the following days. Randomly distributed p63-positive cells amounted to 35% in the studied 2–3-day keratinocyte cultures. The process of epidermal cell aggregation was reproduced in the population of basal keratinocytes, where Ki-67-expressing cells amounted to about 20%. Single cells expressing keratin 19 were observed during cell aggregation in the secondary culture. We believe that the aggregation of cultured keratinocytes reflects the formation of structural-functional units (SFUs) in vivo. Disaggregated epidermal keratinocytes maintained the information relevant for self-assembly of three-dimensional structures capable of SFU formation after transplantation into the body.  相似文献
