XRD studies of structure of N-acetoxy-N-methoxyurea and N,N-bis(methoxycarbonyl)-N-methoxyimide have revealed that in N-methoxy-N-X-ureas (X = OAc, Cl, OMe, N(+)C(5)H(5)) the additional shortening of N-OMe bond took place, which arising from an n(O(Me))-sigma*(N-X) anomeric orbital interaction. XRD studies of N-chloro-N-ethoxyurea crystal have revealed the presence of two kinds of anomeric nitrogen configuration in the O-N-Cl group in the form of a pyramidal configuration and a planar configuration for same 1-N nitrogen atom. XRD studies of N-4-chlorobenzoyloxy-N-ethoxyurea have revealed that the degree of pyramidality of the 1-N nitrogen in N-aroyloxy-N-alkoxyureas is tuned by orientation of benzoyl group with respect to the N-O bond, which in turn depends of size of N-alkoxy group.  相似文献

Peculiarities of the course of metabolic processes in tissues of the bivalve mollusc Anadara inaequivalvis Br. were studied under conditions of experimental anoxia. In the absence of oxygen, in gill and foot the protein catabolism processes were found to be enhanced; this led to a decrease of the protein content and to an increase of the free amino acid and urea levels. Predominantly hydrolyzed were low molecular peptides, which was indicated by a decrease of the cathepsin D activity on the background of a rise of the γ-glutamyltranspeptidase activity. Anoxia was accompanied by enhancement of the succinate thiokinase and fumarate reductase reactions controlled by alanine and aspartate aminotransferases. This prevented accumulation of toxic lactate in tissues and allowed obtaining an additional macroerg resource. Metabolic processes in the mollusc hepatopancreas were oriented to production of amino acids.  相似文献

In Escherichia coli phage T4 and many of its phylogenetic relatives, gene 43 consists of a single cistron that encodes a PolB family (PolB-type) DNA polymerase. We describe the divergence of this phage gene and its protein product (gp43) (gene product 43) among 26 phylogenetic relatives of T4 and discuss our observations in the context of diversity among the widely distributed PolB enzymes in nature. In two T4 relatives that grow in Aeromonas salmonicida phages 44RR and 25, gene 43 is fragmented by different combinations of three distinct types of DNA insertion elements: (a) a short intercistronic untranslated sequence (IC-UTS) that splits the polymerase gene into two cistrons, 43A and 43B, corresponding to N-terminal (gp43A) and C-terminal (gp43B) protein products; (b) a freestanding homing endonuclease gene (HEG) inserted between the IC-UTS and the 43B cistron; and (c) a group I intron in the 43B cistron. Phage 25 has all three elements, whereas phage 44RR has only the IC-UTS. We present evidence that (a) the split gene of phage 44RR encodes a split DNA polymerase consisting of a complex between gp43A and gp43B subunits; (b) the putative HEG encodes a double-stranded DNA endonuclease that specifically cleaves intron-free homologues of the intron-bearing 43B site; and (c) the group I intron is a self-splicing RNA. Our results suggest that some freestanding HEGs can mediate the homing of introns that do not encode their own homing enzymes. The results also suggest that different insertion elements can converge on a polB gene and evolve into a single integrated system for lateral transfer of polB genetic material. We discuss the possible pathways for the importation of such insertion elements into the genomes of T4-related phages.  相似文献

Studies are beginning to emerge that demonstrate intriguing differences between human‐induced pluripotent stem cells (hiPSCs) and human embryonic stem cells (hESCs). Here, we investigated the expression of key members of the Nodal embryonic signaling pathway, critical to the maintenance of pluripotency in hESCs. Western blot and real‐time RT‐PCR analyses reveal slightly lower levels of Nodal (a TGF‐β family member) and Cripto‐1 (Nodal's co‐receptor) and a dramatic decrease in Lefty (Nodal's inhibitor and TGF‐β family member) in hiPSCs compared with hESCs. The noteworthy drop in hiPSC's Lefty expression correlated with an increase in the methylation of Lefty B CpG island. Based on these findings, we addressed a more fundamental question related to the consequences of epigenetically reprogramming hiPSCs, especially with respect to maintaining a stable ESC phenotype. A global comparative analysis of 365 microRNAs (miRs) in two hiPSC versus four hESC lines ultimately identified 10 highly expressed miRs in hiPCSs with >10‐fold difference, which have been shown to be cancer related. These data demonstrate cancer hallmarks expressed by hiPSCs, which will require further assessment for their impact on future therapies. J. Cell. Selective H2-histamine agonist dimaprit was shown to produce relaxation of the isolated frog subclavian vein, with it persisting under the effect of selective H2-histamine antagonist cimetidine. Possible nonspecific mechanisms of relaxation produced by histamine are discussed. The data presented do not exclude that there are atypical H2-histamine receptors in subclavian vein of frogs, the activation of which initiates the attenuation of the active tension.  相似文献

The horse fly fauna (Diptera: Tabanidae) of Jordan is, after Israel, the richest in the Levant, with 24 known species. During the 20-year project "The Ecology and Zoogeography of the Lepidoptera of the Near East," we regularly collected blood-feeding flies, resulting in 11 additional species of Tabanidae for Jordan. The new records are: Atylotus quadrifarius (Loew, 1874), Chrysops caecutiens (Linnaeus, 1758), Dasyrhamphis nigritus (Fabricius, 1794), Haematopota pallens Loew, 1871, Nemorius irritans (Ricardo, 1901), Philipomyia graeca (Fabricius, 1794), Tabanus cordiger Meigen, 1820, Tabanus taeniola Palisot de Beauvois, 1806, Tabanus quatuornotatus Meigen, 1820, Tabanus separatus Effllatoun, 1930, and Tabanus spectabilis Loew, 1858. Most of the new records (10/11) are of Palearctic origin; of these, six are of a Mediterranean and one each of West Palearctic, Euroasiatic, Irano-Turanian, and Eremic providence. Only one species, T. taeniola, is an Afrotropical-Eremic element.  相似文献

Although airway epithelial cells provide important barrier and host defense functions, a crucial role for these cells in development of acute lung inflammation and injury has not been elucidated. We investigated whether NF-kappaB pathway signaling in airway epithelium could decisively impact inflammatory phenotypes in the lungs by using a tetracycline-inducible system to achieve selective NF-kappaB activation or inhibition in vivo. In transgenic mice that express a constitutively active form of IkappaB kinase 2 under control of the epithelial-specific CC10 promoter, treatment with doxycycline induced NF-kappaB activation with consequent production of a variety of proinflammatory cytokines, high-protein pulmonary edema, and neutrophilic lung inflammation. Continued treatment with doxycycline caused progressive lung injury and hypoxemia with a high mortality rate. In contrast, inducible expression of a dominant inhibitor of NF-kappaB in airway epithelium prevented lung inflammation and injury resulting from expression of constitutively active form of IkappaB kinase 2 or Escherichia coli LPS delivered directly to the airways or systemically via an osmotic pump implanted in the peritoneal cavity. Our findings indicate that the NF-kappaB pathway in airway epithelial cells is critical for generation of lung inflammation and injury in response to local and systemic stimuli; therefore, targeting inflammatory pathways in airway epithelium could prove to be an effective therapeutic strategy for inflammatory lung diseases.  相似文献

Both diverse assemblages of small skeletal fossils and a representative chemostratigraphical record make the Siberian Platform widely regarded as one of the key regions for the reconstruction of global biotic and abiotic events in the late Ediacaran and early Cambrian. However, the wide distribution of intertidal–subtidal facies in the Ediacaran–Cambrian transitional strata of the central and southwestern Siberian Platform (Turukhansk–Irkutsk–Olekma facies region) produces a dramatic depletion of the palaeontological record and considerably limits their age‐calibration and long‐distance correlation. We report new lithological, palaeontological and carbonate carbon‐isotope data for the Ediacaran–Cambrian sections of the Turukhansk Uplift (northwestern Siberian Platform, western facies region). These data provide a robust framework for the chemostratigraphical correlation of the western facies region with sections of the transitional and eastern regions of the Siberian Platform and further confirms a depositional hiatus at the base of the Tommotian Stage in the stratotype section (Aldan River, SE Siberia). The carbon‐isotope curve from the Turukhansk Uplift sections correlates positively with the most chemostratigraphically representative Ediacaran–Cambrian sections (Siberia, Morocco, South China). It records major carbon‐isotope oscillations globally recognized in the lower Cambrian, enabling localization of the Fortunian and Cambrian Stage 2 boundaries in the Platonovskaya Formation. Although there is extreme paucity and poor preservation of the small skeletal fossils in the western facies region, we report individual Barskovia, Blastulospongia and chancelloriid sclerites from the Platonovskaya Formation. A combination of palaeontological and chemostratigraphical data suggests the base of P. antiqua Assemblage Zone is located in the middle Platonovskaya Formation. The earliest spiral gastropods probably occurred at ~541 Ma, as demonstrated by the discovery of a specimen of Barskovia near the base of the large negative excursion in the lower Platonovskaya Formation, correlated with the BACE negative carbon‐isotope peak in the sections of the Yangtze Platform.  相似文献

Indirect effects of climate change are often mediated by trophic interactions and consequences for individual species depend on how they are tied into the local food web. Here we show how the response of demographic rates of an arctic bird of prey to fluctuations in small rodent abundance changed when small rodent community composition and dynamics changed, possibly under the effect of climate warming. We observed the breeding biology of rough‐legged buzzards (Buteo lagopus) at the Erkuta Tundra Monitoring Site in southern Yamal, low arctic Russia, for 19 years (1999–2017). At the same time, data on small rodent abundance were collected and information on buzzard diet was obtained from pellet dissection. The small rodent community experienced a shift from high‐amplitude cycles to dampened fluctuations paralleled with a change in species composition toward less lemmings and more voles. Buzzards clearly preferred lemmings as prey. Breeding density of buzzards was positively related to small rodent abundance, but the shift in small rodent community lead to lower numbers relative to small rodent abundance. At the same time, after the change in small rodent community, the average number of fledglings was higher relative to small rodent abundance than earlier. These results suggest that the buzzard population adapted to a certain degree to the changes in the major resource, although at the same time density declined. The documented flexibility in the short‐term response of demographic rates to changes in structure and dynamics of key food web components make it difficult to predict how complex food webs will be transformed in a warmer Arctic. The degree of plasticity of functional responses is indeed likely to vary between species and between regions, depending also on the local food web context.  相似文献
