Dormancy of hazel (Corylus avellana L.) seeds is alleviated by a chilling treatment during which cytological, hormonal, and biochemical changes occur. Phytic acid and phosphate mobilization have been examined during this treatment. Phytic acid accounted for 0.7% and up to 3.2% of dry weight in axiferous and cotyledonary tissue, respectively. Phytic acid levels in embryonic axes were reduced by 60% within the first 3 weeks of chilling, with little subsequent change, in contrast to warm-imbibed tissue where levels did not change significantly. In cotyledons, phytic acid was mobilized to a lesser extent. Phosphate levels expressed on a fresh weight basis remained almost unaltered suggesting either the operation of a homeostatic mechanism for intracellular concentration or rapid utilization due to active metabolism. Phytase activity increased during stratification in both axiferous and cotyledonary tissue. The initial rise observed was associated with dormancy alleviation, since it occurred before the realization of full germination potential by the seeds and not in warm-imbibed tissue. Protein bodies were isolated from hazel seeds by non-aqueous density gradients. Phytase activity was closely associated with the purified organelles, where phytic acid was located by light microscopy. Overall, these findings suggest that phytic acid mobilization by phytase and previously described processes associated with protein bodies, such as considerable proteolysis, are early participants in the plethora of events leading to seed dormancy relief and germination in hazel.  相似文献

INTRODUCTION: The repeatability of subjective and objective assessments of neck muscle fatigue is very important with regard to the clinical applicability of these methods. METHOD: To establish between-days reliability, 33 healthy volunteers performed a 60% maximum voluntary isometric contraction test from a standing position in all neck movements. Cervical muscle fatigue was assessed on three separate occasions from the spectral (median frequency, MF) and amplitude (root mean square, RMS) analysis of the electromyogram (EMG) signal recorded from the cervical paraspinal group, splenius capitis, levator scapulae and sternocleidomastoid. Subjective assessment of fatigue was rated by employing the Borg scale. Intraclass correlation coefficient ICC((1,1)), standard error of measurement (SEM), smallest detectable difference (SDD) indices and Pearson's correlation co-efficient were calculated for the analysis of the results. RESULTS: Normalised median frequency (MF) slope had low repeatability and large between-day error (ICC((1,1))=0.28-0.61; SEM=0.33-0.60%/s; SDD=132.7-703.2%) for the protagonist muscles of each movement. Initial median frequency (IMF) had moderate to good reliability and small error (ICC((1,1))=0.64-0.81; SEM=2.8-8.8Hz; SDD=19.9-38.5%). The RMS slope yielded also poor repeatability. The Borg assessment was more reliable than the EMG estimate though variability between sessions was still quite high (SDD=29.2-136.5%). No correlation was found between the EMG and Borg assessment of neck muscle fatigue (r=-0.01-0.39). CONCLUSION: The protocol used for assessing neck muscle fatigue proved to be reliable only for the IMF but the clinical usefulness of this measure remains questionable. The lack of correlation between objective and subjective estimation of neck muscle fatigue was possibly a consequence of the poor measurement repeatability. Further research is needed to identify the factors responsible for these results on neck area.  相似文献

The purpose of this study was to examine the relations between patellar tendon moment arm length and several relevant anthropometric characteristics of 22 healthy men. The patellar tendon moment arm length was measured using magnetic resonance imaging with two different methods: (1) measurement of patellar tendon moment arm length (d(PT)) with respect to the tibiofemoral contact point (d(PTCP)) and (2) measurement of d(PT) with respect to the intersection point of the anterior and posterior cruciate ligament (d(PTIP)). Pearson correlation coefficients and a stepwise linear regression analysis were used to examine the relationships between the d(PT) and anthropometric measurements taken. Furthermore, a Student's t-test was used to determine differences between the d(PTCP) and d(PTIP) values. Only knee circumference was a significant d(PTCP) predictor (P < 0.05) but with a very low R2 (0.139). None of the anthropometric parameters examined was found to be a significant d(PTIP) predictor. The correlation coefficients ranged from -0.04 to 0.42. The d(PTIP) values were significantly higher (by 0.84-1.89 cm) than the d(PTCP) values (P < 0.05). These results are in disagreement with previous in vitro findings that d(PT) variance may be explained by knee joint size differences. Hence, existing imaging-based methodologies remain necessary for accurate quantification of the patellar tendon moment arm.  相似文献

Toxorhynchites guadeloupensis (Dyar Knab), a poorly known mosquito species, was observed preying upon Aedes aegypti (L.) larvae, in an oviposition trap placed for routine dengue entomological surveillance, during 2003-2004 in the urban area of Boa Vista, Roraima, Brazil. This is the first report for Tx. guadeloupensis using Ae. aegypti oviposition traps as breeding places. This finding may have important consequences in the epidemiology and local dengue control since Ae. aegypti density is a basic variable in dengue prediction. Whether predation of Ae aegypti by Tx. guadeloupensis in the Amazon is of significance, is a question to be examined. Also, larval predation may be a cause for underestimation of the actual Ae aegypti numbers. Together these hypotheses need to be better investigated as they are directly related to dengue epidemiology, to the success of any outbreak prediction and surveillance program.  相似文献

Surface sediments were collected from the coastal zone of Drapetsona–Keratsini (Saronikos Gulf, Greece) in December 2012 for determining the local benthic foraminiferal community, identifying their spatial distribution patterns, and evaluating the response of foraminiferal species to geochemical composition through the hierarchical cluster analysis, principal component analysis and Spearman's rho correlation. Foraminifera can be classified into three distinct assemblages associated with the granulometry, elemental geochemistry, particulate organic carbon content and degree of sediment contamination. A relatively low-diversity assemblage, dominated by stress-tolerant taxa with Ammonia tepida Bolivina spathulata and Bulimina elongata being the prevailing species, is characteristic of the silty seabed of the main part of Drapetsona coastal zone and the Keratsini Port central basin, where organic carbon content, aliphatic and polycyclic aromatic hydrocarbons concentrations and trace metal loads are greatly elevated. On the sandy bottom of the investigated area, relatively high frequencies of miliolids prevail. An epiphytic rotaliid-dominated assemblage is recorded in the slightly-polluted sedimentary bottom of the inner and western part of the Keratsini Port.  相似文献

Chemotaxis, together with motility, helps bacteria foraging in their habitat. Motile bacteria exhibit a variety of motility patterns, often controlled by chemotaxis, to promote dispersal. Motility in many bacteria is powered by a bidirectional flagellar motor. The flagellar motor has been known to briefly pause during rotation because of incomplete reversals or stator detachment. Transient pauses were previously observed in bacterial strains lacking CheY, and these events could not be explained by incomplete motor reversals or stator detachment. Here, we systematically analyzed swimming trajectories of various chemotaxis mutants of the monotrichous soil bacterium, Azospirillum brasilense. Like other polar flagellated bacterium, the main swimming pattern in A. brasilense is run and reverse. A. brasilense also uses run-pauses and putative run-reverse-flick-like swimming patterns, although these are rare events. A. brasilense mutant derivatives lacking the chemotaxis master histidine kinase, CheA4, or the central response regulator, CheY7, also showed transient pauses. Strikingly, the frequency of transient pauses increased dramatically in the absence of CheY4. Our findings collectively suggest that reversals and pauses are controlled through signaling by distinct CheY homologs, and thus are likely to be functionally important in the lifestyle of this soil organism.  相似文献

Understanding the different background landscapes in which malaria transmission occurs is fundamental to understanding malaria epidemiology and to designing effective local malaria control programs. Geology, geomorphology, vegetation, climate, land use, and anopheline distribution were used as a basis for an ecological classification of the state of Roraima, Brazil, in the northern Amazon Basin, focused on the natural history of malaria and transmission. We used unsupervised maximum likelihood classification, principal components analysis, and weighted overlay with equal contribution analyses to fine-scale thematic maps that resulted in clustered regions. We used ecological niche modeling techniques to develop a fine-scale picture of malaria vector distributions in the state. Eight ecoregions were identified and malaria-related aspects are discussed based on this classification, including 5 types of dense tropical rain forest and 3 types of savannah. Ecoregions formed by dense tropical rain forest were named as montane (ecoregion I), submontane (II), plateau (III), lowland (IV), and alluvial (V). Ecoregions formed by savannah were divided into steppe (VI, campos de Roraima), savannah (VII, cerrado), and wetland (VIII, campinarana). Such ecoregional mappings are important tools in integrated malaria control programs that aim to identify specific characteristics of malaria transmission, classify transmission risk, and define priority areas and appropriate interventions. For some areas, extension of these approaches to still-finer resolutions will provide an improved picture of malaria transmission patterns.  相似文献

In this work, we describe an NMR-based method that utilizes an orientation media composed of the chiral polypeptide liquid crystal poly-gamma-benzyl-L-glutamate (PBLG) dissolved in CDCl(3), to measure the (1)H-(1)H, (1)H-(13)C and (13)C-(13)C residual dipolar couplings (RDCs) of (R) and (S)-ibuprofen. Calculated RDCs, obtained from the lowest energy conformers, are then compared with the experimentally measured RDCs to predict the stereochemistry of each enantiomer. Excellent agreement between calculated and experimental RDCs was found when the lowest energy structure of each enantiomer, obtained in a simulated PBLG/CDCl(3) environment, was used to back-calculate the RDCs. This method is generally useful for small molecular weight molecules that possess either one or two chiral centers, are soluble in low viscosity organic solvents, and will not crystallize (Clegg, Crystal Structure Analysis. Principles and Practice. New York: Oxford University Press; 2002) or cannot be derivatized with a Mosher's reagent (Dale and Mosher, J Am Chem Soc 1973;95:512-519).  相似文献

Knowledge of the dietary habits of an endangered species is essential to its management and conservation. The brown bear (Ursus arctos) exists in two isolated population in Greece, but only initial management actions have been taken to improve the habitat of this species. To improve our understanding of the ecology and habitat requirements of this species, we examined the dietary habits of the brown bear in the Panagia-Grevena region, of Macedonia, Greece. In total, 360 scats of brown bear were collected between 2002 and 2004 during spring, summer and autumn months. Scats were analyzed by their frequency of occurrence, volumetric and dry weights, and their importance values. Microhistological analysis was applied to estimate the proportion of wild and cultivated plants in the diet. The most important type of food in the spring was green vegetation, while, in the summer, fruits of Pyrus sp., Morus sp., Prunus sp. and Rubus sp. were important food items. In autumn, hard masts, mainly oak (Quercus sp.), were essential foods for the brown bear. The frequency of vertebrates in the diet was higher in the summer and autumn while that of invertebrates was higher in the spring. In summary, the brown bear is an omnivorous species that lives in Greece and adapts its diet according to food availability and human activities in its habitat. For this reason, human activities in the study area must take the needs and requirement of brown bears into consideration.  相似文献
