The objective of this study was to characterize the histodifferentiation of somatic embryogenesis obtained from leaf explants of C. arabica. Therefore, we histologically analyzed the respective stages of the process: leaf segments at 0, 4, 7, 15 and 30 days of cultivation, Type 1 primary calli (primary calli with embryogenic competence) and 2 (primary calli with no embryogenic competence), embryogenic calli, globular, torpedo and cotyledonary embryos, and mature zygotic embryos. Callus formation occurred after seven days of culture, with successive divisions of procambium cell. In this cultivation phase, it was found that Type 1 primary calli are basically formed by parenchymal cells with reduced intercellular spacing, whereas Type 2 primary calli are predominantly composed of parenchymal cells with ample intercellular spaces and embryogenic calli composed entirely of meristematic cells. After 330 days, it was evident from the differentiation of somatic embryogenesis that there was formation of globular somatic embryos, consisting of a characteristic protoderm surrounding the fundamental meristem. With the maturation of these propagules after 360 days, torpedo-stage somatic embryos arose, in which tissue polarization and early differentiation of procambial strands were verified. After 390 days, cotyledonary somatic embryos were obtained, where the onset of vessel elements differentiation was verified, a characteristic also observed in mature zygotic embryos. We concluded that somatic embryogenesis obtained from C. arabica leaves initiates from procambium cell divisions that, in the course of cultivation, produce mature somatic embryos suitable for regenerating whole plants. 相似文献

Leptodactylus paraensis Heyer, is a Neotropical anuran species that inhabits Rainforest habitats in the eastern Amazon, but because it has only been recently separated from the Leptodactylus pentadactylus (Laurenti) species group, little is known about its helminth fauna. This study describes a new species of Oswaldocruzia Travassos, 1917 and records the first occurrence of this genus parasitising L. paraensis and the second species for the Caxiuanã National Forest in the eastern Amazon, Brazil. Oswaldocruzia lanfrediae n. sp. is characterised by having an anterior extremity with a smooth cephalic vesicle divided into two portions, a claviform oesophagus, well-developed cuticular longitudinal ridges and lateral alae. Females have a well-developed ovojector, with didelphic and amphidelphic uteri. Males show complex robust spicules divided into a slightly curved shoe, a bifurcated fork and a blade terminating in 2–3 processes. The new species differs from its congeners especially regarding the lateral alae and the morphology of the spicules, in addition to morphometric characters such as body size, oesophagus length, deirid position, nerve-ring position and relative position of the vulva in females. 相似文献

Flows of energy and matter across habitat boundaries can be major determinants of the functioning of recipient ecosystems. It is currently debated whether terrestrial dissolved organic matter (tDOM) is a resource subsidy or a resource subtraction in recipient lakes. We present data from a long-term field experiment in which pelagic phosphorus concentration and whole-ecosystem primary production increased with increasing tDOM input, suggesting that tDOM acted primarily as a direct nutrient subsidy. Piecewise structural equation modeling supports, however, a substantial contribution of a second mechanism: colored tDOM acted also as a resource subtraction by shading benthic algae, preventing them from intercepting nutrients released across the sediment–water interface. Inhibition of benthic algae by colored tDOM thus indirectly promoted pelagic algae and whole-ecosystem primary production. We conclude that cross-ecosystem terrestrial DOM inputs can modify light and nutrient flows between aquatic habitats and alter the relative contributions of benthic and pelagic habitats to total primary production. These results are particularly relevant for shallow northern lakes, which are projected to receive increased tDOM runoff. 相似文献

A Flow Injection Analysis (FIA) for sucrose using invertase (E.C. 3.2.1.26), mutarotase (E.C.5.1.3.3) and glucose oxidase (E.C.1.1.3.4) was developed. The enzymes were immobilised on glass beads using glutaraldehyde. The sucrose concentration was related to oxygen saturation. Fall in O2 concentration, as a result of sucrose oxidation, was detected by a low cost, home-made O2 electrode. The system was able to measure sucrose from 0.025 to 100mM with a response time of 6min using 200 l of sample, with an apparent Km of 42mM of sucrose. The feeding ecology of the lesser weever, Echiichthys vipera, from the adjacent coastal areas of the Douro and Tagus estuaries (Portugal) was studied between October 2000 and July 2002. The stomach contents of 246 individuals were analysed and diet was characterized by the numerical, gravimetric, occurrence and vacuity indices. Variation of feeding habits with fish length (<95 and >95 mm) and geographical area was considered. Diet of the lesser weever comprised a large variety of prey (28 species), the most important of which were crustaceans (numerical index, NI = 93.5%; occurrence index, OI = 75.6%), namely Mysidacea (especially Schistomysis sp.), Amphipoda (mainly Gammarus subtypicus) and Isopoda (Idotea spp.), and also Teleostei (mostly larval stages that posted a gravimetric index, GI = 53.0%). Diet varied with fish length, with large individuals showing a larger diversity of prey items. Furthermore, specimens from Douro also showed a higher diversity of prey items than those from Tagus. More than 50% of the stomachs were empty, being the highest vacuity values relative to smaller fishes as well as to individuals from the Tagus estuary adjacent coastal area. 相似文献

Neonatal N‐methyl‐D‐aspartate (NMDA) receptor blockade in rodents triggers schizophrenia (SCZ)‐like alterations during adult life. SCZ is influenced by gender in age of onset, premorbid functioning, and course. Estrogen, the hormone potentially driving the gender differences in SCZ, is known to present neuroprotective effects such as regulate oxidative pathways and the expression of brain‐derived neurotrophic factor (BDNF). Thus, the aim of this study was to verify if differences in gender and/or estrous cycle phase during adulthood would influence the development of behavioral and neurochemical alterations in animals neonatally administered ketamine. The results showed that ketamine‐treated male (KT‐male) and female‐in‐diestrus (KTF‐diestrus, the low estrogen phase) presented significant deficits in prepulse inhibition of the startle reflex and spatial working memory, two behavioral SCZ endophenotypes. On the contrary, female ketamine‐treated rats during proestrus (KTF‐proestrus, the high estradiol phase) had no behavioral alterations. This correlated with an oxidative imbalance in the hippocampus (HC) of both male and KTF‐diestrus female rats, that is, decreased levels of GSH and increased levels of lipid peroxidation and nitrite. Similarly, BDNF was decreased in the KTF‐diestrus rats while no alterations were observed in KTF‐proestrus and male animals. The changes in the HC were in contrast to those in the prefrontal cortex in which only increased levels of nitrite in all groups studied were observed. Thus, there is a gender difference in the adult rat HC in response to ketamine neonatal administration, which is based on the estrous cycle. The ability of oysters to purge themselves of microbial contaminants was investigated by identifying the microorganisms retained by oysters after they have been subjected to ultraviolet (UV) light-treated seawater. A UV intensity of 960 muw per min per cm(2) reduced the microbial count of seawater from 263 to 13 per ml. The coliform multitube test (MPN) was reduced from a high of 17 to <0.18 per 100 ml. Over 75% of the microorganisms found in treated seawater were Acinetobacter/Moraxella, Vibrio/Pseudomonas type II, and Flavobacterium/Cytophaga. With the exception of coliforms, the microbial composition of oysters subjected to UV-treated seawater remained at levels comparable to the control oysters held in untreated seawater. Total counts ranged between 10(3) and 10(5)/g. The microorganism most frequently encountered were Flavobacterium/Cytophaga, Vibrio/Pseudomonas type II, Pseudomonas type III or IV, Acinetobacter/Moraxella, gram-positive cocci and Bacillus. Together they comprised over 90% of the flora. Coagulase-positive, deoxyribonuclease-positive, and beta-hemolytic cocci were found in some samples, as were V. parahaemolyticus, V. aliginolyticus, and Aeromonas species. 相似文献
