Piroxicam is used in the treatment of rheumatoid arthritis, osteoarthritis, and other inflammatory diseases. Upon oral administration, it is reported to cause ulcerative colitis, gastrointestinal irritation, edema and peptic ulcer. Hence, an alternative delivery system has been designed in the form of transethosome. The present study describes the preparation, optimization, characterization, and ex vivo study of piroxicam-loaded transethosomal gel using the central composite design. On the basis of the prescreening study, the concentration of lipids and ethanol was kept in the range of 2–4% w/v and 0–40% v/v, respectively. Formulation was optimized by measuring drug retention in the skin, drug permeation, entrapment efficiency, and vesicle size. Optimized formulation was incorporated in hydrogel and compared with other analogous vesicular (liposomes, ethosomes, and transfersomes) gels for the aforementioned responses. Among the various lipids used, soya phosphatidylcholine (SPL 70) and ethanol in various percentages were found to affect drug retention in the skin, drug permeation, vesicle size, and entrapment efficiency. The optimized batch of transethosome has shown 392.730 μg cm?2 drug retention in the skin, 44.312 μg cm?2 h?1 drug permeation, 68.434% entrapment efficiency, and 655.369 nm vesicle size, respectively. It was observed that the developed transethosomes were found superior in all the responses as compared to other vesicular formulations with improved stability and highest elasticity. In 1980 the State of Connecticut began a tidal marsh restoration program targeting systems degraded by tidal restrictions and impoundments. Such marshes become dominated by common reed grass (Phragmites australis) and cattail (Typha angustifolia and T. latifolia), with little ecological connection to Long Island Sound. The management and scientific hypothesis was that returning tidal action, reconnecting marshes to Long Island Sound, would set these systems on a recovery trajectory. Specific restoration targets (i.e., pre‐disturbance conditions or particular reference marshes) were considered unrealistic. However, it was expected that with time restored tides would return ecological functions and attributes characteristic of fully functioning tidal salt marshes. Here we report results of this program at nine separate sites within six marsh systems along 110 km of Long Island Sound shoreline, with restoration times of 5 to 21 years. Biotic parameters assessed include vegetation, macroinvertebrates, and use by fish and birds. Abiotic factors studied were soil salinity, elevation and tidal flooding, and soil water table depth. Sites fell into two categories of vegetation recovery: slow, ca. 0.5%, or fast, more than 5% of total area per year. Although total cover and frequency of salt marsh angiosperms was positively related to soil salinity, and reed grass stand parameters negatively so, fast versus slow recovery rates could not be attributed to salinity. Instead, rates appear to reflect differences in tidal flooding. Rapid recovery was characterized by lower elevations, greater hydroperiods, and higher soil water tables. Recovery of other biotic attributes and functions does not necessarily parallel those for vegetation. At the longest studied system (rapid vegetation recovery) the high marsh snail Melampus bidentatus took two decades to reach densities comparable with a nearby reference marsh, whereas the amphipod Orchestia grillus was well established on a slow‐recovery marsh, reed grass dominated after 9 years. Typical fish species assemblages were found in restoration site creeks and ditches within 5 years. Gut contents of fish in ditches and on the high marsh suggest that use of restored marsh as foraging areas may require up to 15 years to reach equivalence with reference sites. Bird species that specialize in salt marshes require appropriate vegetation; on the oldest restoration site, breeding populations comparable with reference marshland had become established after 15 years. Use of restoration sites by birds considered marsh generalists was initially high and was still nearly twice that of reference areas even after 20 years. Herons, egrets, and migratory shorebirds used restoration areas extensively. These results support our prediction that returning tides will set degraded marshes on trajectories that can bring essentially full restoration of ecological functions. This can occur within two decades, although reduced tidal action can delay restoration of some functions. With this success, Connecticut's Department of Environmental Protection established a dedicated Wetland Restoration Unit. As of 1999 tides have been restored at 57 separate sites along the Connecticut coast. 相似文献

Connectivity plays a crucial role in determining the spread, viability, and persistence of populations across space. Dispersal across landscapes, or the movement of individuals or genes among resource patches, is critical for functional connectivity. Yet current connectivity modelling typically uses information on species location or habitat preference rather than movement, which unfortunately may not capture key dispersal limitations. We argue that recent developments in species distribution modelling provide insightful lessons for addressing this gap and advancing our understanding of connectivity. We suggest shifting the focus of connectivity modelling from locating where animals potentially disperse to a process‐based approach directed towards understanding and mapping factors that limit successful dispersal. To do so, we propose defining species dispersal requirements through identifying spatial, environmental and intrinsic constraints to successful dispersal, analogous to identifying environmental dimensions that define niches. We discuss the benefits of this constraint‐based framework for understanding the distribution of species, predicting species responses to climate change, and connectivity conservation practice. We illustrate how the framework can aid in identifying potential detrimental effects of human activities on connectivity and species persistence, and can spur the implementation of innovative conservation strategies. The proposed framework clarifies the validity and contextual utility of objectives and measures in existing connectivity models, and identifies gaps that may impede our understanding of connectivity and its integration into successful conservation strategies. We expect that this framework will facilitate a mechanistic approach to understanding and conserving connectivity, which will aid in effectively predicting and mitigating effects of ongoing environmental change. 相似文献

BackgroundDengue virus (DENV) infection is prevalent across tropical regions and may cause severe disease. Early diagnosis may improve supportive care. We prospectively assessed the Standard Diagnostics (Korea) BIOLINE Dengue Duo DENV rapid diagnostic test (RDT) to NS1 antigen and anti-DENV IgM (NS1 and IgM) in children in Cambodia, with the aim of improving the diagnosis of DENV infection.ConclusionsThe DENV RDT had low sensitivity for the diagnosis of DENV infection. The high co-prevalence of infections in our cohort indicates the need for a broad microbiological assessment of non-localised febrile illness in these children. 相似文献

BACKGROUND: Juvenile hyaline fibromatosis (YHF) is a rare inherited disorder characterized by tumorous growth of hyalinized fibrous tissue. No report on cytomorphology of this condition is available in English on MEDLINE. CASE REPORT: A 6-year-old girl had multiple nontender nodules on both ear lobes, nose and scalp. Fine needle aspiration of the nodule on the left ear revealed benign, spindle-shaped cells with an eosinophilic ground substance in the background. The diagnosis of JHF was made following cytologic and histopathologic studies. CONCLUSION: Fine needle aspiration cytology is reliable for the diagnosis of JHF. 相似文献

Actomyosin contractility is a major engine of preimplantation morphogenesis, which starts at the 8-cell stage during mouse embryonic development. Contractility becomes first visible with the appearance of periodic cortical waves of contraction (PeCoWaCo), which travel around blastomeres in an oscillatory fashion. How contractility of the mouse embryo becomes active remains unknown. We have taken advantage of PeCoWaCo to study the awakening of contractility during preimplantation development. We find that PeCoWaCo become detectable in most embryos only after the second cleavage and gradually increase their oscillation frequency with each successive cleavage. To test the influence of cell size reduction during cleavage divisions, we use cell fusion and fragmentation to manipulate cell size across a 20- to 60-μm range. We find that the stepwise reduction in cell size caused by cleavage divisions does not explain the presence of PeCoWaCo or their accelerating rhythm. Instead, we discover that blastomeres gradually decrease their surface tensions until the 8-cell stage and that artificially softening cells enhances PeCoWaCo prematurely. We further identify the programmed down-regulation of the formin Fmnl3 as a required event to soften the cortex and expose PeCoWaCo. Therefore, during cleavage stages, cortical softening, mediated by Fmnl3 down-regulation, awakens zygotic contractility before preimplantation morphogenesis.During preimplantation morphogenesis, the mouse embryo relies on forces generated by the actomyosin cytoskeleton. This study uncovers how periodic actomyosin contractions increase in frequency during cleavage stages as blastomeres soften with each cleavage division. 相似文献

In the present study, the phytoremedation potential along with growth, physiological and biochemical response of tomato (Solanum lycopersicum) was assessed under heavy metal(loid) (HM) and arbuscular mycorrhizal fungus (AMF) amendment. Effect of AMF on uptake and accumulation of metal(loid)s was assessed and accumulation characteristics were expressed in terms of bioabsorption coefficient (BAC), bioconcentration factor (BCF), translocation factor (TLF) and transfer factor (TF). Results showed that AMF-inoculated plants showed not only a better growth, chlorophyll content, strengthened non-enzymatic and enzymatic defense mechanism, but also accumulated higher concentration of metal(loid)s. The correlation between biochemical and physiological parameters was significant at 0.01 level. A significant difference (p ≤ 0.001) in antioxidant enzyme activity was found on increasing metal(loid) dose and application of AMF. The accumulation of Cd and Pb in edible part exceeded the chronic reference dose stated by USEPA. The target hazard quotient (THQ) was >1 for Cd and Pb, whereas <1 for As. The study shows that tomato has good potential as Cd and Pb phytoremediator, hence must not be consumed when grown on Cd or Pb contaminated sites. 相似文献
