Surge (non-linear) uptake of ammonium, measured by incorporation of 15N, was investigated in three species of macroalgae (Ulva lactuca Linnaeus (Chlorophyta), Soliera robusta (Greville) Kylin (Rhodophyta) and Dictyota dichotoma (Hudson) Lamouroux (Phaeophyta)) from Kavaratti atoll (Lakshadweep, India). Addition of ammonium (up to 20 μmol L− 1) led to pronounced uptake within 4-6 min, with the amount of ammonium taken up during surge phase (< 4 min) accounting for from about half to 10 times that taken up during the remaining period of incubation (5-30 min). Amount of ammonium taken up during surge related linearly to the concentration of ammonium given. Surge uptake in the dark was also substantial, averaging 80% of that in light. Capability for rapid uptake of pulses of ammonium released by heterotrophs during the day or night could thus be an important mechanism of survival and proliferation of macroalgae in the N-impoverished atoll waters. 相似文献

8-Chloroadenosine (8-Cl-Ado) is a ribosyl nucleoside analog currently in phase I testing for the treatment of chronic lymphocytic leukemia (CLL). 8-Cl-Ado activity is dependent on adenosine kinase and requires intracellular accumulation of 8-Cl-Ado as mono-, di-, and tri-phosphates. In the current study with four mantle cell lymphoma cell lines, we report a new major metabolic pathway for 8-Cl-Ado intracellular metabolism, the formation of succinyl-8-chloro-adenosine (S-8-Cl-Ado) and its monophosphate (S-8-Cl-AMP). 8-Cl-AMP levels were highly associated with S-8-Cl-AMP levels and reached a steady-state prior to the secondary metabolites, 8-Cl-ATP and S-8-Cl-Ado. Consistent with fumarate as a required substrate for formation of succinyl-8-Cl-adenylate metabolites, the S-8-Cl-adenylate concentrations in multiple cell lines were associated with fumarate loss. The distribution of metabolites was also altered using the energy metabolism modifiers, metformin and oligomycin. The rates of succinyl-8-Cl-adenylate metabolism were enhanced by increasing the intracellular fumarate concentrations after metformin co-treatment. In addition, the S-8-Cl-AMP concentrations were increased after acute inhibition of ATP synthase by oligomycin. We conclude that 8-Cl-Ado metabolism not only affects intracellular purine metabolism; 8-Cl-Ado conversion to succinyl analogs ties its metabolism to the citric acid cycle by reduction of the fumarate pool. 相似文献

The purpose of this study was to determine the relationship between hip and knee strength, and valgus knee motion during a single leg squat. Thirty healthy adults (15 men, 15 women) stood on their preferred foot, squatted to approximately 60 deg of knee flexion, and returned to the standing position. Frontal plane knee motion was evaluated using 3-D motion analysis. During Session 2, isokinetic (60 deg/sec) concentric and eccentric hip (abduction/adduction, flexion/extension, and internal/external rotation) and knee (flexion/extension) strength was evaluated. The results demonstrated that hip abduction (r2=0.13), knee flexion (r2=0.18), and knee extension (r2=0.14) peak torque were significant predictors of frontal plane knee motion. Significant negative correlations showed that individuals with greater hip abduction (r=-0.37), knee flexion (r=-0.43), and knee extension (r=-0.37) peak torque exhibited less motion toward the valgus direction. Men exhibited significantly greater absolute peak torque for all motions, excluding eccentric internal rotation. When normalized to body mass, men demonstrated significantly greater strength than women for concentric hip adduction and flexion, knee flexion and extension, and eccentric hip extension. We have previously shown that the extracellular nucleoside triphosphate-hydrolyzing enzyme NTPDase2 is highly expressed in situ by stem/progenitor cells of the two neurogenic regions of the adult murine brain: the subventricular zone (type B cells) and the dentate gyrus of the hippocampus (residual radial glia). We explored the possibility that adult multipotent neural stem cells express nucleotide receptors and investigated their functional properties in vitro. Neurospheres cultured from the adult mouse SVZ in the presence of epidermal growth factor and fibroblast growth factor 2 expressed the ecto-nucleotidases NTPDase2 and the tissue non-specific isoform of alkaline phosphatase, hydrolyzing extracellular ATP to adenosine. ATP, ADP and, to a lesser extent, UTP evoked rapid Ca(2+) transients in neurospheres that were exclusively mediated by the metabotropic P2Y(1) and P2Y(2) nucleotide receptors. In addition, agonists of these receptors and low concentrations of adenosine augmented cell proliferation in the presence of growth factors. Neurosphere cell proliferation was attenuated after application of the P2Y(1)-receptor antagonist MRS2179 and in neurospheres from P2Y(1)-receptor knockout mice. In situ hybridization identified P2Y(1)-receptor mRNA in clusters of SVZ cells. Our results infer nucleotide receptor-mediated synergism that augments growth factor-mediated cell proliferation. Together with the in situ data, this supports the notion that extracellular nucleotides contribute to the control of adult neurogenesis. 相似文献

The recruitment of FtsZ to the septum and its subsequent interaction with other cell division proteins in a spatially and temporally controlled manner are the keys to bacterial cell division. In the present study, we have tested the hypothesis that FtsZ and FtsW of Mycobacterium tuberculosis could be binding partners. Using gel renaturation, pull-down, and solid-phase assays, we confirm that FtsZ and FtsW interact through their C-terminal tails, which carry extensions absent in their Escherichia coli counterparts. Crucial to these interactions is the cluster of aspartate residues Asp(367) to Asp(370) of FtsZ, which most likely interact with a cluster of positively charged residues in the C-terminal tail of FtsW. Mutations of the aspartate residues 367-370 showed that changing three aspartate residues to alanine resulted in complete loss of interaction. This is the first demonstration of the direct interaction between FtsZ and FtsW. We speculate that this interaction between FtsZ and FtsW could serve to anchor FtsZ to the membrane and link septum formation to peptidoglycan synthesis in M. tuberculosis. The findings assume particular significance in view of the global efforts to explore new targets in M. tuberculosis for chemotherapeutic intervention. 相似文献

Hepatoprotective activity of the n-heptane extract of Cassia fistula leaves was investigated by inducing hepatotoxicity with paracetamol in rats. The extract at a dose of 400 mg/kg body wt. exhibited orally, significant protective effect by lowering the serum levels of transaminases (SGOT and SGPT), bilirubin and alkaline phosphatase (ALP).
