The costimulatory molecule CD28 has a restricted tissue distribution and is expressed on T cells and some plasmacytoma cells. Although CD28 is constitutively expressed, its expression is transiently down-regulated following T cell activation and declines progressively with in vitro senescence. In vivo, CD8+ T cells and, less frequently, CD4+ T cells may completely lose CD28 surface expression during chronic infections and with aging. This correlates with changes of nuclear protein-binding activities to two motifs, site alpha and beta, within the CD28 minimal promoter. Both alpha- and beta-bound complexes are found only in lymphoid tissues, in CD28+ T cells, and in some transformed B cells. These complexes are coordinately expressed except during replicative senescence, which is characterized by the down-modulation of site beta- but not site alpha-binding activities. In contrast, T cell activation induces a parallel decline in both site alpha- and beta-binding activities. CD4+ and CD8+ T cells differ in their beta-binding profiles, which may explain the more pronounced down-regulation of CD28 in senescent CD8+ T cells. In vivo expanded CD4+CD28null and CD8+CD28null T cells uniformly lack alpha- and beta-bound complexes, resembling the pattern seen in chronically activated cells and not of senescent cells.  相似文献

Domiciliated Rhodnius prolixus and sylvatic R. colombiensis were analyzed in order to confirm their genetic divergence and verify the risk that the latter represents in the domiciliation process, and to provide tools for identifying the sources of possible reinfestation by triatomines in human dwellings allowing control programs to be undertaken. Comparison of random amplified polymorphic DNA amplification patterns and cluster analysis suggests reproductive discontinuity between the two species. The calculated statistical F value of 0.24 and effective migration rate of 0.6 individuals per generation are insufficient to maintain genetic homogeneity between them and confirm the absence of present genetic flow. R. colombiensis presents higher intrapopulation variability. Polymerase chain reaction of ribosomal DNA supports these findings. The low genetic flow between the two species implies that R. colombiensis do not represent an epidemiological risk for the domiciliary transmission of Trypanosoma cruzi in the Tolima Department. The lower variability of the domiciliated R. prolixus could result in greater susceptibility to the use of pesticides in control programs.  相似文献

Bacillus thuringiensis subsp. medellin was grown until sporulation in a 1.1 l fermenter in batch and intermittent fed-batch culture. At optimum conditions 25 g dry cells l–1 and 9×108 spores ml–1 were produced. Question: Is post‐fire, medium‐term vegetation dynamics determined by land‐use or fire history prior to fire? Location: South‐facing slope in the Gallinera valley, Alicante province, eastern Spain. Methods: After mapping the land‐use and fire history of the study site using photo‐interpretation, we sampled vegetation structure on a set of plots representing the most frequent land‐use and fire history combinations on an area burned six years before sampling. We studied the effects of land‐use history, comparing the one‐fire land‐use trajectories. We analysed the effects of fire history; comparing one‐ and two‐fire plots for both previously cropped and uncropped areas. Results: Most variables were not significantly different between the earliest abandoned plots (abandoned at least 38 years before the fire) and the uncropped plots. On the most recently abandoned plots (abandoned between one and four years before the fire), the therophyte richness and the ratio of seeder: resprouter richness were significantly greatest. Different fire recurrences did not determine different post‐fire vegetation on either the uncropped or the early abandoned plots (all dominated by fire‐recruited seeder shrubs). The most recently abandoned plots had a lower resilience to fire. Conclusions: Land‐use history and recent pre‐fire land use, in particular, determined the post‐fire vegetation in the medium term. The vegetation composition converged during secondary succession among land‐use histories. Increasing fire recurrence had a small effect on mature plant communities, due to the combination of life‐history traits determining the response to fire of the dominant species.  相似文献

Differences in morphology, biomass allocations and physiological responses were investigated in seedlings of Mastic tree (Pistacia lentiscus L.) and Cork oak (Quercus suber L.) submitted to contrasting fertilization and light regimes during early growth. These species are two evergreen sclerophyllous Mediterranean species frequently used in Mediterranean reforestation programmes. Fertilization was the treatment that affected most of the morphological and physiological variables evaluated in P. lentiscus and Q. suber seedlings. Leaf area and specific leaf area (SLA) were affected by shading treatment in both species, showing higher values in seedlings grown under shade. P. lentiscus seedlings showed a high capacity to modify root morphological variables and root hydraulic conductance (KR) with the fertilization treatment. In contrast, Q. suber showed low to moderate root system changes with the treatments applied, although the fertilization level affected biomass allocation (i.e., root to shoot ratio) in both species. Under high water demand, P. lentiscus seedlings with high KR allowed transpiration (E) to increase without increasing the water potential gradient between soil and leaves. In Q. suber, high fertilization induced significant increases in photosynthesis (A), as well as a tendency to increase E with significantly lower leaf water potential (ψL).  相似文献

In the Mediterranean Basin, most cultivated areas were abandoned in the last century and are now in various stages of old-field succession. The aim of this work was to analyse the successional trajectories of these ecosystems, and to assess possible deviations in these pathways due to fire occurrence at high or low recurrence levels. Old-fields abandoned either about 50 or about 100 years ago were selected in SE Spain. Within the 50-year-old abandoned fields, plots were established which had been burned by 1, 2 and 3 fires in the last 25 years. Cover values of vascular species were sampled and then analysed by means of multivariate analysis. Euclidean distances between resulting communities were used as an indicator of the possible deviation from the unburned successional pathway. Our results pointed to the possibility that different successional pathways may exist depending on fire occurrence and recurrence. In the absence of fire, the vegetation is dominated by pioneer species, mainly Pinus. With the passage of time this vegetation will become dominated by later successional tree species (Quercus). However, when early-successional communities are affected by fire, the succession can be diverted. A single fire is enough to change Pinus forests into alternative stable states dominated by Rosmarinus officinalis shrub communities, where the colonisation of species in later successional stages is arrested. This deviation increases in high fire recurrence regimes where the vegetation changes to dwarf shrubs and herbs.  相似文献
