Saltwater incursion carries high concentrations of sea salts, including sulfate, which can alter anaerobic microbial processes and plant community composition of coastal freshwater marshes. We studied these phenomena in a recently restored wetland on the coastal plain of North Carolina. We measured water inundation patterns, porewater chemistry, microbial process rates, plant tissue chemistry and iron plaque on plant roots, and quantified plant community composition across a hydrologic and salinity gradient to understand the potential interactions between saltwater incursion and changes in microbial processes and plant communities. Plant communities showed no obvious response to incursion, but were structured by inundation patterns and plant growth form (for example, graminoid versus forb). Saltwater incursion increased chloride and sulfate concentrations in surface and porewater, and drove resulting spatial patterns in anaerobic microbial metabolism rates. Plots experiencing saltwater incursion had higher sulfate reduction rates and were dominated by graminoid plant species (for example, sedges, rushes, and grasses). Graminoid plant species’ roots had greater iron plaque formation than forb and submerged species, indicative that graminoid plant species are supplying more oxygen to the rhizosphere, potentially influencing microbial metabolism. Future studies should focus on how plant and microbial communities may respond to saltwater incursion at different time scales, and on parsing out the influence that plants and microbes have on each other as freshwater wetlands experience sea level rise.  相似文献

In this single case study, visuospatial neglect patient P1 demonstrated a dissociation between an intact ability to make appropriate reflexive eye movements to targets in the neglected field with latencies of <400 ms, while failing to report targets presented at such durations in a separate verbal detection task. In contrast, there was a failure to evoke the usually robust Remote Distractor Effect in P1, even though distractors in the neglected field were presented at above threshold durations. Together those data indicate that the tight coupling that is normally shown between attention and eye movements appears to be disrupted for low-level orienting in P1. A comparable disruption was also found for high-level cognitive processing tasks, namely reading and scene scanning. The findings are discussed in relation to sampling, attention and awareness in neglect.  相似文献

Seed formation in flowering plants requires meiosis of the megaspore mother cell (MMC) inside the ovule, selection of a megaspore that undergoes mitosis to form an embryo sac, and double fertilization to initiate embryo and endosperm formation. During apomixis, or asexual seed formation, in Hieracium ovules, a somatic aposporous initial (AI) cell divides to form a structurally variable aposporous embryo sac and embryo. This entire process, including endosperm development, is fertilization independent. Introduction of reproductive tissue marker genes into sexual and apomictic Hieracium showed that AI cells do not express a MMC marker. Spatial and temporal gene expression patterns of other introduced genes were conserved commencing with the first nuclear division of the AI cell in apomicts and the mitotic initiation of embryo sac formation in sexual plants. Conservation in expression patterns also occurred during embryo and endosperm development, indicating that sexuality and apomixis are interrelated pathways that share regulatory components. Heat shock response is characterized by the induction of heat shock proteins (HSPs), which facilitate protein folding, and non-HSP proteins with diverse functions, including protein degradation, and is regulated by heat shock factors (HSFs). HSF1 is a master regulator of HSP expression during heat shock in mammals, as is HSF3 in avians. HSF2 plays roles in development of the brain and reproductive organs. However, the fundamental roles of HSF2 in vertebrate cells have not been identified. Here we find that vertebrate HSF2 is activated during heat shock in the physiological range. HSF2 deficiency reduces threshold for chicken HSF3 or mouse HSF1 activation, resulting in increased HSP expression during mild heat shock. HSF2-null cells are more sensitive to sustained mild heat shock than wild-type cells, associated with the accumulation of ubiquitylated misfolded proteins. Furthermore, loss of HSF2 function increases the accumulation of aggregated polyglutamine protein and shortens the lifespan of R6/2 Huntington's disease mice, partly through αB-crystallin expression. These results identify HSF2 as a major regulator of proteostasis capacity against febrile-range thermal stress and suggest that HSF2 could be a promising therapeutic target for protein-misfolding diseases.  相似文献

HUPO initiated the Plasma Proteome Project (PPP) in 2002. Its pilot phase has (1) evaluated advantages and limitations of many depletion, fractionation, and MS technology platforms; (2) compared PPP reference specimens of human serum and EDTA, heparin, and citrate-anti-coagulated plasma; and (3) created a publicly-available knowledge base (www.bioinformatics.med.umich.edu/hupo/ppp; www.ebi.ac.uk/pride). Thirty-five participating laboratories in 13 countries submitted datasets. Working groups addressed (a) specimen stability and protein concentrations; (b) protein identifications from 18 MS/MS datasets; (c) independent analyses from raw MS-MS spectra; (d) search engine performance, subproteome analyses, and biological insights; (e) antibody arrays; and (f) direct MS/SELDI analyses. MS-MS datasets had 15 710 different International Protein Index (IPI) protein IDs; our integration algorithm applied to multiple matches of peptide sequences yielded 9504 IPI proteins identified with one or more peptides and 3020 proteins identified with two or more peptides (the Core Dataset). These proteins have been characterized with Gene Ontology, InterPro, Novartis Atlas, OMIM, and immunoassay-based concentration determinations. The database permits examination of many other subsets, such as 1274 proteins identified with three or more peptides. Reverse protein to DNA matching identified proteins for 118 previously unidentified ORFs. We recommend use of plasma instead of serum, with EDTA (or citrate) for anticoagulation. To improve resolution, sensitivity and reproducibility of peptide identifications and protein matches, we recommend combinations of depletion, fractionation, and MS/MS technologies, with explicit criteria for evaluation of spectra, use of search algorithms, and integration of homologous protein matches. This Special Issue of PROTEOMICS presents papers integral to the collaborative analysis plus many reports of supplementary work on various aspects of the PPP workplan. These PPP results on complexity, dynamic range, incomplete sampling, false-positive matches, and integration of diverse datasets for plasma and serum proteins lay a foundation for development and validation of circulating protein biomarkers in health and disease.  相似文献

Aim Few studies have attempted to assess the overall impact of fragmentation at the landscape scale. We quantify the impacts of fragmentation on plant diversity by assessing patterns of community composition in relation to a range of fragmentation measures. Location The investigation was undertaken in two regions of New Zealand – a relatively unfragmented area of lowland rain forest in south Westland and a highly fragmented montane forest on the eastern slopes of the Southern Alps. Methods We calculated an index of community similarity (Bray–Curtis) between forest plots we regarded as potentially affected by fragmentation and control forest plots located deep inside continuous forest areas. Using a multiple nonlinear regression technique that incorporates spatial autocorrelation effects, we analysed plant community composition in relation to measures of fragmentation at the patch and landscape levels. From the resulting regression equation, we predicted community composition for every forest pixel on land‐cover maps of the study areas and used these maps to calculate a landscape‐level estimate of compositional change, which we term ‘BioFrag’. BioFrag has a value of one if fragmentation has no detectable effect on communities within a landscape, and tends towards zero if fragmentation has a strong effect. Results We detected a weak, but significant, impact of fragmentation metrics operating at both the patch and landscape levels. Observed values of BioFrag ranged from 0.68 to 0.90, suggesting that patterns of fragmentation have medium to weak impacts on forest plant communities in New Zealand. BioFrag values varied in meaningful ways among landscapes and between the ground‐cover and tree and shrub communities. Main conclusions BioFrag advances methods that describe spatial patterns of forest cover by incorporating the exact spatial patterns of observed species responses to fragmentation operating at multiple spatial scales. BioFrag can be applied to any landscape and ecological community across the globe and represents a significant step towards developing a biologically relevant, landscape‐scale index of habitat fragmentation.  相似文献

The human enteric flora plays a significant role in intestinal health and disease. Certain enteric bacteria can inhibit the NF-kappaB pathway by blockade of IkappaB-alpha ubiquitination. IkappaB-alpha ubiquitination is catalyzed by the E3-SCF(betaTrCP) ubiquitin ligase, which is itself regulated via covalent modification of the cullin-1 subunit by the ubiquitin-like protein NEDD8. Neddylation is a biochemical event associated with diverse cellular processes related to cell signaling, however, physiological regulation of cullin neddylation has not been described in mammalian systems. We report that interaction of nonpathogenic bacteria with epithelial cells resulted in a rapid loss of neddylated Cul-1 and consequent repression of the NF-kappaB pathway. This observation may explain the ability of intestinal bacterial communities to influence diverse eukaryotic processes in general and inflammatory tolerance of the mammalian intestinal epithelia specifically.  相似文献
