In this study, we have used a single link system with a pair of muscles that are excited with alpha and gamma signals to achieve both point to point and oscillatory movements with variable amplitude and frequency.The system is highly nonlinear in all its physical and physiological attributes. The major physiological characteristics of this system are simultaneous activation of a pair of nonlinear muscle-like-actuators for control purposes, existence of nonlinear spindle-like sensors and Golgi tendon organ-like sensor, actions of gravity and external loading. Transmission delays are included in the afferent and efferent neural paths to account for a more accurate representation of the reflex loops.A reinforcement learning method with an actor-critic (AC) architecture instead of middle and low level of central nervous system (CNS), is used to track a desired trajectory. The actor in this structure is a two layer feedforward neural network and the critic is a model of the cerebellum. The critic is trained by state-action-reward-state-action (SARSA) method. The critic will train the actor by supervisory learning based on the prior experiences. Simulation studies of oscillatory movements based on the proposed algorithm demonstrate excellent tracking capability and after 280 epochs the RMS error for position and velocity profiles were 0.02, 0.04 rad and rad/s, respectively.  相似文献

A nonsense mutation at codon 95 (R95X) in the C9 gene is responsible for most Japanese C9 deficiency (C9D) cases, with a carrier frequency of 6.7%. Upon analysis of microsatellite markers and newly identified dinucleotide repeat number polymorphisms in the 3' flanking region of the C9 gene, a founder effect was demonstrated for the R95X mutation of the C9 gene in Japanese. Screening for the R95X mutation in Korean and Chinese individuals showed that the R95X carrier frequencies in Koreans and Chinese were 2.0% and 1.0%, respectively. Although homozygotes for the R95X mutation were not found in Korea or China, the shared haplotype of the dinucleotide repeat number polymorphisms appeared to be associated with the R95X mutation in the heterozygotes in Korea and China. The founder effect found in East Asians (Japanese, Koreans and Chinese) but not in Caucasians, as well as the haplotype sharing in only a small chromosomal interval, suggested that the R95X mutation of C9 gene was ancient and had occurred after the divergence of East Asians and Caucasians, and before migration of the Yayoi people to Japan. Since the mortality of meningococcal infections in complement-deficient patients is lower than that in normal individuals, a founder effect and a selective advantage in isolation might be the main reasons for the high frequency of the R95X mutation in Japan.  相似文献

Synthetic and botanical insecticides can have a profound effect on the developmental period, growth, adult emergence, fecundity, fertility, and egg hatch, resulting in effective control at sub-lethal concentrations. This paper investigated sub-lethal concentrations of fenitrothion, lambda-cyhalothrin, and Callitris glaucophylla Joy Thomps. & L.P. Johnson (Cupressaceae) extract to characterize their effects on the development of Aedes aegypti L. (Diptera: Culicidae) mosquito larvae. The LC25, LC50, and LC75 (four replicates) were used for each synthetic insecticide and the LC25 and LC75 (four replicates) were used for C. glaucophylla. Observations of larval mortality, duration of larval stage, pupal mortality, duration of pupal stage, adult emergence, sex ratio, and malformations were recorded over 14 days. A dose-response effect was observed for all insecticides. Although C. glaucophylla extract doses were higher than synthetic insecticide doses, the LC75 treatment outperformed synthetics by completely prohibiting adult emergence. Consequently, this botanical is recommended for field application either in combination with synthetic or natural insecticides or alone.  相似文献

Life table, reproduction, and population growth parameters of the beet armyworm, Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae), were determined on nine soybean (Glycine max L.) cultivars ('032', '033', 'Hill', 'M4', 'M7', 'M9', 'M11', 'TMS', and 'Zane') at 25°C. The age-specific survival rate of eggs and immature stages was higher on 033 in comparison with other cultivars. The gross fecundity rate ranged from 911 on M9 to 1,296 on Zane, but the differences were not statistically significant. The net fecundity rate was significantly higher on 033 (1,082 eggs per female) than on other cultivars. The gross fertility rate significantly differed on tested cultivars, and was the highest on Zane (1,257 eggs per female) and lowest on Hill (813 eggs per female). The net fertility rate varied from 532 on M11 to 1,082 eggs per female on 033. Both daily number of eggs and daily number of fertile eggs laid per female varied significantly among the different cultivars, and were the highest on TMS (90 for both parameter) and lowest on M7 (47 and 43), respectively. The values of net reproductive rate, intrinsic rate of increase, and finite rate of increase were significantly higher on 033. The shortest generation time (23 d), shortest doubling time (1.66 d), and highest percentage of female offspring (56%) also were obtained on 033. According to population growth parameters, 033 and Hill partially were susceptible and resistant, respectively, to S. exigua. The results of this study provide direction to design a more comprehensive integrated pest management program for this pest.  相似文献

To model the developmental pattern of human prothymocytes and thymopoiesis, we used NOD-scid/γc(-/-) mice grafted with human umbilical cord blood CD34(+) hematopoietic progenitor cells (HPCs). Human prothymocytes developed in the murine bone marrow (BM) from multipotent CD34(++)CD38(lo)lineage(-) HPCs to CD34(++)CD7(+)CD2(-) pro-T1 cells that progressed in a Notch-dependent manner to CD34(+)CD7(++)CD2(+) pro-T2 cells, which migrated to the thymus. BM prothymocyte numbers peaked 1 mo after graft, dropped at mo 2, and persisted at low levels thereafter, with only a few CD34(+)CD7(lo) prothymocytes with limited T potential being detected by mo 5. As a consequence, thymopoiesis in this xenogeneic setting began by weeks 4-6, peaked at mo 3, and decreased thenceforth. Analyzing mice grafted at 2, 4 or 8, mo of age showed that in an "older" BM, prothymocyte differentiation was perturbed and resulted in CD34(+)CD7(lo) prothymocytes with limited T potential. Whereas the early drop in BM thymopoietic activity was related to a Notch-independent loss of T potential by CD34(++)CD38(lo)lineage(-) HPCs, the later age-dependent production decline of prothymocytes was linked to a more complex mix of cell-intrinsic and microenvironmental defects. Accordingly, and contrasting with what was observed with umbilical cord blood HPCs, CD34(+) HPCs from human adult BM displayed only marginal thymopoietic activity when grafted into young 2-mo-old NOD-scid/γc(-/-) mice. These data demonstrate that the developmental pattern of BM prothymocytes during human late fetal and early postnatal life can be reproduced in humanized mice, and they suggest that onset of human thymus involution relates to decreased colonization by prothymocytes.  相似文献

Effect of long-term orthodontic treatment on salivary nickel and chromium has not been quite assessed except in few retrospective studies with controversial results. The aim of this prospective study was to measure salivary levels of these ions during 1?year of orthodontic treatment. Saliva samples were collected from 20 orthodontic patients, before treatment (control) and 6 and 12?months later. Nickel and chromium concentrations were determined using atomic absorption spectrophotometry. Data were analyzed using one- and two-way repeated-measures ANOVA, Bonferroni, Friedman (???=?0.05), and Wilcoxon signed-ranks tests (???=?0.016). Average nickel level changed from 9.75?±?5.02 to 10.37?±?6.94 and then to 8.32?±?4.36???g/L in 1?year. Average chromium concentration changed from 3.86?±?1.34 to 4.6?±?6.11 and then to 2.04?±?1.66???g/L. Alterations in nickel values were not statistically significant [P?=?0.468 (ANOVA)], but fluctuations in chromium levels were [P?=?0.021 (Friedman)]. The decrease in chromium concentration after 12?months was significant compared to the control [P?=?0.004 (Wilcoxon)]. Although slightly increased after 6?months, the concentration of both ions dropped to levels slightly lower than the control groups after 12?months.  相似文献

A set of unique sequences in bacterial genomes, responsible for protecting bacteria against bacteriophages, has recently been used for the genetic manipulation of specific points in the genome. These systems consist of one RNA component and one enzyme component, known as CRISPR (“clustered regularly interspaced short palindromic repeats”) and Cas9, respectively. The present review focuses on the applications of CRISPR/Cas9 technology in the development of cellular and animal models of human disease. Making a desired genetic alteration depends on the design of RNA molecules that guide endonucleases to a favorable genomic location. With the discovery of CRISPR/Cas9 technology, researchers are able to achieve higher levels of accuracy because of its advantages over alternative methods for editing genome, including a simple design, a high targeting efficiency and the ability to create simultaneous alterations in multiple sequences. These factors allow the researchers to apply this technology to creating cellular and animal models of human diseases by knock‐in, knock‐out and Indel mutation strategies, such as for Huntington's disease, cardiovascular disorders and cancers. Optimized CRISPR/Cas9 technology will facilitate access to valuable novel cellular and animal genetic models with respect to the development of innovative drug discovery and gene therapy.  相似文献

Isozyme electrophoresis data of seed extracts from 11 populations of diploid wheat species (Triticum boeoticum Bioss. and Triticum urartu Thumanian ex Gandilyan), distributed mainly in the western and west-northern Iran, were investigated. The five enzyme systems used were peroxidase, polyphenol oxidase, superoxide dismutase, malate dehydrogenase and catalase. The first three were found to be useful as molecular marker for characterization of diploid wheat populations. A total of 13 bands from three enzyme systems were recorded. The value of a ‘Jaccard's’ similarity coefficient ranges from 0.333 to 1.000. Data analysis was done using clustering method UPGMA. On the basis of Jaccard's coefficient, the obtained dendrogram supports previous relationship between T. boeoticum and T. urartu as separate species as well as reflecting their distinct gene pools and substantiating their specific recognition despite the overall morphological similarity.  相似文献
