By the example of 2,4-dichlorophenoxyacetic acid (2,4-D) assay by the inhibition of latex agglutination, new synthetic polymer
microspheres for the conjugation with antibodies to 2,4-D and agglutinators based on ovalbumin and polyacrylamide were developed
and characterized. The effect of various parameters (the concentration of reagents, the type and the degree of modification
of the microsphere surface, and the nature of the carrier in the composition of the agglutinator) on the rate of agglutination
and the maximal optical absorption observed during the reaction were studied by turbidimetry. The optimal parameters were

Activation of phospholipase C (PLC)-mediated signaling pathways in non-excitable cells causes the release of calcium (Ca2+) from inositol 1,4,5-trisphosphate (InsP3)-sensitive intracellular Ca2+ stores and activation of Ca2+ influx via plasma membrane Ca2+ channels. The properties and molecular identity of plasma membrane Ca2+ influx channels in non-excitable cells is a focus of intense investigation. In the previous studies we used patch clamp electrophysiology to describe the properties of Ca2+ influx channels in human carcinoma A431 cell lines. Now we extend our studies to human embryonic kidney HEK293 cells. By using a combination of Ca2+ imaging and whole cell and single channel patch clamp recordings we discovered that: 1) HEK293 cells contain four types of plasma membrane Ca2+ influx channels: I(CRAC), Imin, Imax, and I(NS); 2) I(CRAC) channels are highly Ca2+-selective (P(Ca/Cs)>1000) and I(CRAC) single channel conductance is too small for single channel analysis; 3) Imin channels in HEK293 cells display functional properties identical to Imin channels in A431 cells, with single channel conductance of 1.2 pS for divalent cations, 10 pS for monovalent cations, and divalent cation selectivity P(Ba/K)=20; 4) Imin channels in HEK293 cells are activated by InsP3 and inhibited by phosphatidylinositol 4,5-bisphosphate, but store-independent; 5) when compared with Imin, Imax channels have higher conductance for divalent (17 pS) and monovalent (33 pS) cations, but less selective for divalent cations (P(Ba/K)=4), 6) Imax channels in HEK293 cells can be activated by InsP3 or by Ca2+ store depletion; 7) I(NS) channels are non-selective (P(Ba/K)=0.4) and display a single channel conductance of 5 pS; and 8) I(NS) channels are not gated by InsP3 but activated by depletion of intracellular Ca2+ stores. Our findings provide novel information about endogenous Ca2+ channels supporting receptor-operated and store-operated Ca2+ influx pathways in HEK293 cells. 相似文献

The source of activity of every individual or group is to a great extent determined by the necessity of interaction with other individuals or groups (Afanasiev, 1990), i.e. with the diversity of anthropological surroundings. This manifests itself in the participation of individuals in the organization of their ecosystems, including the ethnocultural and physical conditions of life (Sukharev, 1998). Therefore it is possible to single out certain ecological aspects of perception of anthropological specificity of the surrounding population by an individual (group), leaning upon the definition of anthropoecology as a discipline, studying the laws of interaction between human communities and the system of natural, social and other factors, (Anthropological dictionary, 2004) as well as the coevolution of humans with their environment, in the process of adaptation (Lisseyev, 2001). At the same time it is usually emphasized that the ecological approach is best of all realized on the base of the principle of reciprocity of development of an individual and the surrounding reality, on the perception of the objects of this reality (including other people) with due regard to the reasons for the preference and corresponding values orientation. This represents the subject of ecological psychology (Sergeyenko, 2002; Pavlenko, 2002). In this context the relations between the individual and the environment (autoecology) as well as between the individual and a group (sinecology) are considered (Lisseyev, 2001).The study of the aesthetic perception of anthropological types by individuals belonging to different ethnoterritorial and age groups is the subject matter of a special branch of physical anthropology-anthropoaesthetics, which studies the peculiarities of the aesthetic preference of human facial features in modern populations, analyzing the dependence of human perception on the anthropological environment (Haldeyeva, 2004). 相似文献

Inheritance of the two main types of the plant resistance to insects was investigated in the sorghum-greenbug (Schizaphis graminum Rond.) and wheat-bird cherry-oat aphid (Rhopalosiphon padi L.) interaction systems. The data obtained support the hypothesis that antixenosis (avoiding of the plant by the insect, given a choice) and antibiosis (adverse effect of the plant on the insect feeding on it) are pleiotropic manifestations of the same genes. This is confirmed by the following facts. (1) Identical patterns of segregation for antixenosis and antibiosis in different cases of sorghum resistance to the greenbug: monogenic control (gene Sgr4), digenic control (Sgr1, Sgr2 and Sgr7, Sgr8), and complementary action of the genes (Sgr9 and Sgr10). (2) Correlated changes in the levels of antibiosis and antixenosis during long-term reproduction of a greenbug clone on the resistant sorghum variety k-1206 (resistance controlled by one gene). (3) Simultaneous expression of antixenosis and antibiosis in F3 wheat hybrid families to the bird cherry-oat aphid. 相似文献

The scarcity of water in arid and semiarid regions of the world is a problem that every day increases by climate change. The subsurface drip irrigation (SDI) and changes in population density of plants are alternatives that can be used to make a sustainable use of water. Therefore, the objectives of this study were to determine the combination that allows for an increased corn performance and efficient use of water without losing the quality of forage. Three different irrigation levels were applied through a system of a SDI at three different densities of forage maize plants in an arid region. The results demonstrated that by applying different levels of water, either enough or lack of soil moisture is created, which is directly reflected in crop yield, and its determining variables such as green forage and dry matter yield, and nutritional quality. The irrigation level to a 100% of potential evapotranspiration (PET), at a density of 80000 plants/ha, increased yield of green forage to 57664 kg/ha; crude protein was 8.59%, while the rest of the quality parameters decreased. This study allowed to conclude that the irrigation level was the major factor in the response of the crop. 相似文献

Non-growing free cells as well as immobilized ones produced higher levels of lysine than growing free cells, when cultured for 24 h in a medium containing 80 g/L glucose as a carbon source. Blood serum gamma-globulin and albumin were tested for their effect on the sodium currents of voltage clamped internally perfused serum deprived neuroblastoma cells. Albumin in the concentration corresponding to its content in 5% blood serum (22 microM) produced an increase in sodium channel currents and shifts in activation and inactivation curves along the voltage axis towards more negative or positive potentials, respectively. These results both qualitatively and quantitatively reproduce the effects of 5% blood serum observed previously (Zubov, Sal'nikov, 1984, 1986). The addition of gamma-globulin failed to change the parameters registered. The data obtained led us to an assumption of the role of albumin as an active substance responsible for the effect of blood serum on the potential-dependent sodium-transporting system of neuroblastoma cell membrane. 相似文献

The ultramicroscopic composition and supramolecular structure of wood matrix were investigated by methods of electron and atomic force microscopy (AFM). New data on specific features of the composition and cell wall structure of Juniper wood (Juniperus communis L.) were obtained. Native lignin was found to be water soluble.
