Small mammals use plant species for gathering food resources and for shelter. Preferences for certain plant species are related to nutritional restrictions and behavioural patterns, which could be altered in the presence of an infectious disease. Several native small mammals are part of the wild cycle of the protozoan Trypanosoma cruzi, responsible for Chagas disease in humans. This is a vector‐borne disease transmitted by insects of the subfamily Triatominae. We examined the effect of T. cruzi infection status on the use and preference patterns of shrub species by two native rodent species: Octodon degus and Phyllotis darwini. This study was conducted during four sampling years (2010–2013) in a hyper‐endemic zone of Chagas disease located in a semiarid Mediterranean ecosystem. We captured individuals of 599 O. degus and 575 P. darwini (89% of the total captures), which were related to nine shrub species and examined for T. cruzi infection. In a community‐level analysis, infected and non‐infected O. degus used individual shrub species within the shrub community significantly non‐randomly relative to their availability; the same pattern was detected for non‐infected P. darwini individuals, whereas infected individuals used the shrub community according to the abundance of each shrub species. Examining individual preferences, both rodents showed a strong preference for Flourensia thurifera and Colliguaja odorifera regardless of their infection status. Preferences for specific shrub species were variable among years, showing a ‘core’ of preferred shrub species and variable levels of use of the remaining ones. Our results show that T. cruzi infection in wild small mammals can modify habitat use patterns and preferences for certain shrub species, probably affecting processes acting at community level. 相似文献

Expression of fibroblast growth factor (FGF)-inducible 14 (Fn14), a member of the tumor necrosis factor receptor superfamily, is typically low in healthy adult organisms, but strong Fn14 expression is induced in tissue injury and tissue remodeling. High Fn14 expression is also observed in solid tumors, which is why this receptor is under consideration as a therapeutic target in oncology. Here, we describe various novel mouse-human cross-reactive llama-derived recombinant Fn14-specific antibodies (5B6, 18D1, 4G5) harboring the human IgG1 Fc domain. In contrast to recombinant variants of the established Fn14-specific antibodies PDL192 and P4A8, all three llama-derived antibodies efficiently bound to the W42A and R56P mutants of human Fn14. 18D1 and 4G5, but not 5B6, efficiently blocked TNF-like weak inducer of apoptosis (TWEAK) binding at low concentrations (0.2–2 µg/ml). Oligomerization and Fcγ receptor (FcγR) binding converted all antibodies into strong Fn14 agonists. Variants of 18D1 with enhanced and reduced antibody-dependent cell-mediated cytotoxicity (ADCC) activity were further analyzed in vivo with respect to their effect on metastasis. In a xenogeneic model using human colon carcinoma cancer cells, both antibody variants were effective in reducing metastasis to the liver. In contrast, only the 18D1 variant with enhanced ADCC activity, but not its ADCC-defective counterpart, suppressed lung metastasis in the RENCA model. In sum, this suggests that Fn14 targeting might primarily act by triggering of antibody effector functions, but also by blockade of TWEAK-Fn14 interaction in some cases. 相似文献

Cowdriosis, caused by Cowdria ruminantium, is transmitted by Amblyomma ticks, which are widely distributed in Zimbabwe. To assess the distribution of this disease in Zimbabwe, cattle either exposed to Amblyomma ticks or maintained in areas free from these ticks were tested for antibodies to Cowdria. A total of 324 sera were tested using competitive ELISA and the indirect fluorescent antibody test (IFAT). At diptanks in Amblyomma-infested areas 52% (n=95) and 26% (n=47) of sera were positive by cELISA and IFAT, respectively. At diptanks in Amblyomma-free areas 11% (n=125) and 10% (n=134) of sera were positive by cELISA and IFAT, respectively. The results were significantly different between Amblyomma-infested and tick-free areas (2=24.73, P0.005 for IFAT and 2=57.53, P0.005 for cELISA). High background readings in field sera, possibly due to cross-reactive antibodies to Ehrlichia spp., complicated the determination of a realistic cut-off point, especially in cELISA. On the basis of the distribution of Amblyomma ticks, currently a large part of Zimbabwe can be considered endemic for the disease. 相似文献

Defining correlates of immunity by comprehensively interrogating the extensive biological diversity in naturally or experimentally protected subjects may provide insights critical for guiding the development of effective vaccines and antibody‐based therapies. We report advances in a humoral immunoprofiling approach and its application to elucidate hallmarks of effective HIV‐1 viral control. Systematic serological analysis for a cohort of HIV‐infected subjects with varying viral control was conducted using both a high‐resolution, high‐throughput biophysical antibody profiling approach, providing unbiased dissection of the humoral response, along with functional antibody assays, characterizing antibody‐directed effector functions such as complement fixation and phagocytosis that are central to protective immunity. Profiles of subjects with varying viral control were computationally analyzed and modeled in order to deconvolute relationships among IgG Fab properties, Fc characteristics, and effector functions and to identify humoral correlates of potent antiviral antibody‐directed effector activity and effective viral suppression. The resulting models reveal multifaceted and coordinated contributions of polyclonal antibodies to diverse antiviral responses, and suggest key biophysical features predictive of viral control. 相似文献

Several wild animal species have been implicated as hosts of Ehrlichia ruminantium (formerly Cowdria ruminantium), the rickettsial agent causing heartwater, a fatal disease of domestic ruminants in sub-Saharan Africa and eastern Caribbean. However, evidence for infection in most wild species is inconclusive because of inadequate diagnostic techniques. Infection has been proven only in 12 African ruminants, three non-African ruminants and two African rodents. A subclinical carrier state occurs in eight of the African ruminant species. Further studies on E. ruminantium infection in wild animal species are needed in order to determine the host range of this pathogen accurately. The host range of Ehrlichia ruminantium in wildlife is reviewed here and the role played by these species in the epidemiology and spread of heartwater is discussed. 相似文献

Suman Mahan and co-authors review the strategies applied to develop improved vaccines for Cowdria ruminantium infections (heartwater). Inactivated vaccines using cell-cultured C. ruminantium organisms combined with an adjuvant are capable of protecting goats, sheep and cattle against lethal C. ruminantium challenge. Immune responses induced with this vaccine, or after recovery from infection, target outer membrane proteins of C. ruminantium, in particular the major antigenic protein 1 (MAP-1). Genetic immunizations with the gene encoding MAP-1 induce protective T helper cell type 1 responses against lethal challenge in a mouse model. Similarly, homologues of MAP-1 in other phylogenetically and antigenically related ehrlichial agents such as Anaplasma marginale and Ehrlichia chaffeensis are also targets of protective responses. Given the antigenic similarities between the related ehrlichial agents, common strategies of vaccine development could be applied against these agents that cause infections of importance in animals and humans. 相似文献

The effect of a single exhaustive swimming exercise bout on immune competence of untrained or exercise-trained female Wistar rats was compared with the competence of control sedentary rats. After the exhaustive exercise bout, the blastogenic response to concanavalin A by spleen cells of untrained rats was extensively suppressed, whereas the response of the trained rats was only marginally suppressed. The suppressed immune competence of the untrained rats after the exhaustive exercise was associated with an increase in immune-suppressive activity of splenic lymphocytes. The macrophages of the untrained rats and of the control sedentary rats were slightly immune suppressive to normal spleen cells through a prostaglandin-dependent mechanism. The addition of prostaglandin E2 (PGE2) to the blastogenesis cultures revealed that the spleen cells of untrained rats were unusually sensitive to the suppressive effects of PGE2. In contrast to the untrained rats, the marginal level of immune suppression in trained rats after the exhaustive exercise was associated with a lesser degree of lymphocyte-suppressive activity, an immune stimulatory activity by the splenic macrophages, and an insensitivity of the splenic lymphocytes to the suppressive effects of PGE2. 相似文献

Precipitation throughfall (TF) plays an important role in the water balance of tropical forests. This study used 164 gauges to quantify precipitation and TF variability in a tropical pre‐montane transitional cloud forest on the Caribbean slope of the Cordillera Tilarán, Costa Rica, to identify the ecological and meteorological drivers of this variability. Daily TF measurements were taken from 28 June to 17 July 2012 and 12 June to 16 July 2013, for a total of 39 precipitation events. The total mean TF was 87.9 percent and TF at individual gauges ranged from 22.7 percent to 245.7 percent. Leaf area index (LAI) was calculated above each gauge using hemispheric photography for a mean study‐site LAI of 7.7. There was no statistically significant relationship between LAI and TF. However, the amount of TF was positively correlated with precipitation intensity, while the variability of TF was negatively correlated with precipitation intensity. Our calculations indicate that at least 61 gauges are required to obtain mean TF estimates with less than 5 percent error. This study demonstrates that TF is highly spatially heterogeneous due to multiple compounding effects. 相似文献
