1. Shallow lakes and ponds are a major component of the northern landscape and often contain a high zooplankton biomass despite clear waters that are poor in phytoplankton. 2. In this study we quantified zooplankton food sources and feeding rates in the shallow waters of two contrasting high‐latitude biomes: subarctic forest tundra (Kuujjuarapik, Quebec) and high arctic polar desert (Resolute, Nunavut). Five substrate types were tested (beads, bacteria, picophytoplankton, filamentous plankton and microbial mats). Special attention was given to the role of benthos, a component that is usually poorly integrated into models of aquatic foodwebs. 3. Consistent with observations elsewhere in the circumpolar region, high concentrations of adult macrozooplankton occurred in all sites (up to 17 100 crustaceans m?3) while phytoplankton concentrations and primary productivity were low. The communities were composed of multiple species, including Daphnia middendorfiana, Hesperodiaptomus arcticus, Leptodiaptomus minutus, Artemiopsis stefanssoni and Branchinecta paludosa. 4. Detritus made 89–98% of the planktonic resource pool and bacteria contributed the highest biomass (up to 29 mg C m?3) of the planktonic food particles available to zooplankton. Benthic resources were dominated by microbial mats that grew in nutrient‐rich conditions at the base of the ponds and which dominated overall ecosystem biomass and productivity. 5. All species were flexible in their feeding but there were large, order of magnitude differences in clearance rates among taxa. These differences likely resulted from different grazing strategies among cladocerans, copepods and fairy shrimps, and possibly also from adaptation to specific food types and size ranges that occur locally in these waters. 6. The subarctic cladocerans Ceriodaphnia quadrangula and D. middendorfiana, and the arctic fairy shrimp B. paludosa were observed to graze directly on the microbial mats and the feeding experiments confirmed their assimilation of benthic substrates. The other zooplankton species showed a more pelagic feeding mode but were capable of using microbial mat filaments, thus may be indirectly linked to benthic processes via resuspension. 7. Our study indicates that the classical aquatic food web in which phytoplankton provide the sole production base for grazers does not apply to northern shallow lakes and ponds. Instead, microbial mats increase the physical complexity of these high latitude ecosystems and likely play a role in sustaining their high zooplankton biomass.  相似文献

The African savannah elephant (Loxodonta africana) is a ‘keystone’ species that plays a vital role in regulating the dynamics of both plant and animal communities and yet it is endangered and its numbers have been reduced to approximately 500 000 across their entire continental range. Molecular genetic markers are important tools for providing genetic information useful in formulating effective management and conservation strategies for the surviving elephant populations. We describe the isolation and characterization of six new polymorphic microsatellite markers in the African savannah elephant and demonstrate that these loci can be PCR (polymerase chain reaction)‐multiplexed, a desirable attribute that saves costs in large‐scale microsatellite screening.  相似文献

1. Improving our understanding of dietary differences among omnivorous, benthic crustacea can help to define the scope of their trophic influence in benthic food webs. In this study, we examined the trophic ecology of two non‐native decapod crustaceans, the Chinese mitten crab (Eriocheir sinensis) (CMC) and the red swamp crayfish (Procambarus clarkii) (RSC), in the San Francisco Bay ecosystem to describe their food web impacts and explore whether these species are functionally equivalent in their impacts on aquatic benthic communities. 2. We used multiple methods to maximise resolution of the diet of these species, including N and C stable isotope analysis of field data, controlled feeding experiments to estimate isotopic fractionation, mesocosm experiments, and gut content analysis (GCA). 3. In experimental enclosures, both CMC and RSC caused significant declines in terrestrially derived plant detritus (P < 0.01) and algae (P < 0.02) relative to controls, and declines in densities of the caddisfly Gumaga nigricula by >50% relative to controls. 4. Plant material dominated gut contents of both species, but several sediment‐dwelling invertebrate taxa were also found. GCA and mesocosm results indicate that CMC feed predominantly on surface‐dwelling invertebrates, suggesting that trophic impacts of this species could include a shift in invertebrate community composition towards sediment‐dwelling taxa. 5. Stable isotope analysis supported a stronger relationship between CMC and both algae and algal‐associated invertebrates than with allochthonous plant materials, while RSC was more closely aligned with terrestrially derived detritus. 6. The trophic ecology and life histories of these two invasive species translate into important differences in potential impacts on aquatic food webs. Our results suggest that the CMC differs from the RSC in exerting new pressures on autochthonous food sources and shallow‐dwelling invertebrates. The crab's wide‐ranging foraging techniques, use of intertidal habitat, and migration out of freshwater at sexual maturity increases the distribution of the impacts of this important invasive species.  相似文献

Living and fossil megaspores produced by Selaginella (Lycopsida) and its extinct ancestors form distinctive (and occasionally iridescent) exines. Ultrastructural studies of these spores have provided data that demonstrate a colloidal mode of development which in turn implies a degree of self-assembly in the construction of these exines. We present here experimental evidence in support of the theory of selaginellalean megaspore exine construction by depletion flocculation. Iridescent colloidal flocculations of polystyrene latex particles demonstrate an ultrastructural organization virtually indistinguishable from that of the biological system, and clearly demonstrate that self-assembly of complex Selaginella exines by a relatively simple construction process is plausible.  相似文献

Formalin fixation has enjoyed widespread use in the preparation of antibacterial and other vaccines, but rather less use in antitumor vaccines. Previous studies from our laboratories have demonstrated the efficacy of antimelanoma vaccines in mice, produced from formalinized antigens shed by cultured melanoma cells. In this study, we provide evidence that the immunodominant component of that vaccine is the well-characterized B700 melanoma antigen.  相似文献

Sequence analyses of the plastid genes atpB and rbcL support an expanded order Malvales. Within this alliance, core Malvales are clearly supported and comprise most genera that have previously been included in Sterculiaceae, Tiliaceae, Bombacaceae, and Malvaceae. Additional well supported malvalean alliances include the bixalean clade (Bixaceae, Diego-dendraceae, and Cochlospermaceae), the cistalean clade (Cistaceae, Dipterocarpaceae, and Sarcolaenaceae) and Thymelaeaceae (including Gonystyloideae and Aquilarioideae). Our results indicate sister-group relationships between (1) Neuradaceae and the cistalean clade; (2) Sphaerosepalaceae and Thymelaeaceae; (3) these two clades (1 and 2); and (4) all these and an alliance comprising the bixalean clade and core Malvales, but this pattern is weakly supported by the bootstrap. The affinities of Muntingiaceae and Petenaea are especially ambiguous, although almost certainly they are Malvales s.l. The traditional delimitation of families within core Malvales is untenable. Instead, we propose to merge Sterculiaceae, Tiliaceae and Bombacaceae with Malvaceae and subdivide this enlarged family Malvaceae into nine subfamilies based on molecular, morphological, and biogeographical data: (1) Byttnerioideae, including tribes Byttnerieae, Lasiopetaleae and Theobromeae (all of which have cucullate petals) and Hermannieae; (2) Grewioideae, including most genera of former Tiliaceae; (3) Tilioideae, monogeneric in our analysis; (4) Helicteroideae, comprising most of the taxa previously included in Helictereae, plus Mansonia, Triplochiton (indicating that apocarpy evolved at least twice within Malvaceae) and possibly Durioneae; (5) Sterculioideae, defined by apetalous, apocarpous, usually unisexual flowers with androgynophores; (6) Brownlowioideae, circumscribed as in previous classifications; (7) Dombeyoideae, expanded to include Burretiodendron, Eriolaena, Pterospermum, and Schoutmia; (8) Bombacoideae, corresponding to former Bombacaceae (without Durioneae) but including Fremontodendreae  相似文献

Although mat-forming cyanobacteria dominate many freshwater ecosystems in the Arctic and Antarctic, their optimal temperature for growth (Topt) is usually much higher than the temperature range of their native habitat. The present study compared the temperature dependence of growth, pigment composition and absorbance, photosynthesis and photosynthate partitioning for two strains of cyanobacteria with contrasting Topt values; Phormidium subfuscum , isolated from McMurdo Ice Shelf, Antarctica, and Phormidium tenue , collected from the Kuparuk River in the tundra region of northern Alaska. Phormidium subfuscum grew between 5 and 20°C with a Topt of 15°C whereas P. tenue showed detectable growth from 10 to 40°C and a Topt of 30°C. Light utilization efficiency, photosynthetic capacity and the irradiance at the onset of light saturation increased with increasing temperature up to Topt in both strains. The cellular concentrations of chlorophyll a (Chl a) and carotenoid (CAR) and the in vivo absorbance maxima for Chl a, CAR, C-phycocyanin and allophycocyanin changed little for P. subfuscum but all these variables increased across the temperature range up to Topt for P. tenue . Neither P. subfuscum nor P. tenue showed changes in relative carbon allocation with varying temperature, suggesting that gross biochemical alterations are not a characteristic of temperature acclimation in these cyanobacteria. We conclude that the eurythermal cyanobacterium P. tenue optimizes growth over a wide range of temperatures by adjusting its light-capturing as well as carbon fixation characteristics, whereas stenothermal P. subfuscum relies on changes in carbon fixation without concomitant shifts in pigment content.  相似文献
