The climate warming of the postglacial has strongly reduced the distribution of cold‐adapted species over most of Central Europe. Such taxa have therefore become extinct over most of the lowlands and shifted to higher altitudes where they have survived to the present day. The lycaenid butterfly Lycaena helle follows this pattern of former widespread distribution and later restriction to mountain areas such as the European middle mountains. We sampled 203 individuals from 10 populations representing six mountain ranges (Pyrenees, Jura, Massif Central, Morvan, Vosges and Ardennes) over the species' western distribution. Allozyme and microsatellite polymorphisms were analysed to study the genetic status of these highly fragmented populations. Both molecular marker systems revealed a strong genetic differentiation among the analysed populations, coinciding with the orographic structure and highly restricted gene flow among them. The large‐scale genetic differentiation is more pronounced in allozymes (FCT: 0.326) than in microsatellites (RCT: 0.113), but microsatellites show a higher resolution on the regional scale (RSC: 0.082) compared with allozymes (FSC: n.s.). For both analytical tools, we found private alleles occurring exclusively in a single mountain area. The highly fragmented and isolated occurrence of populations is supported by the distribution pattern of potentially suitable climate suggested by species distribution models. Model projections under two climate warming scenarios predict a decline of climatically suitable areas, which will result in the extinction of most of the populations showing unique genetic characteristics.  相似文献

Extracellular phenoloxidase enzymes play an important role in the stability of soil carbon storage by contributing to the cycling of complex recalcitrant phenolic compounds. Climate warming could affect peatland functioning through an alteration of polyphenol/phenoloxidase interplay, which could lead them to becoming weaker sinks of carbon. Here, we assessed the seasonal variability of total phenolics and phenoloxidases subjected to 2–3 °C increase in air temperature using open‐top chambers. The measurements were performed along a narrow fen–bog ecological gradient over one growing season. Climate warming had a weak effect on phenoloxidases, but reduced phenolics in both fen and bog areas. Multivariate analyses revealed a split between the areas and also showed that climate warming exacerbated the seasonal variability of polyphenols, culminating in a destabilization of the carbon cycle. A negative relationship between polyphenols and phenoloxidases was recorded in controls and climate treatments suggesting an inhibitory effect of phenolics on phenoloxidases. Any significant decrease of phenolics through repeatedly elevated temperature would greatly impact the ecosystem functioning and carbon cycle through an alteration of the interaction of polyphenols with microbial communities and the production of extracellular enzymes. Our climate treatments did not have the same impact along the fen–bog gradient and suggested that not all the peatland habitats would respond similarly to climate forcing.  相似文献

Using an intensity rating with no external calibration, experiments were designed to measure the sensory oil-water partition coefficients of four aroma molecules (benzaldehyde, ethyl butyrate, linalool and acetophenone) as the ratios of concentration producing stimuli of equivalent intensities. Flavored water and oil phases were successively assessed for odor intensity by 24 panelists who were given total freedom regarding scaling strategy. Each session combined five concentration levels of three out of the four studied aromas in a solvent (water or oil). A predominant concentration effect was found for each aroma in both solvents and concentration dependencies of perceived intensity above water and oil were similar. Experimental data were modeled with Fechner, Stevens and Hill equations. Combining results above water and oil solutions to feed a common model led to the evaluation of an overall sensory oil-water partition coefficient for each aroma compound. All three models produced similar partition coefficient values for each aroma that were lower than the related instrumental partition coefficients. Biases previously detected when external calibration had been used were reduced in a large proportion while suggested enhancement of odor intensity by water vapor could not be excluded.  相似文献

Acclimation of foliage to growth temperature involves both structural and physiological modifications, but the relative importance of these two mechanisms of acclimation is poorly known, especially for isoprene emission responses. We grew hybrid aspen (Populus tremula x P. tremuloides) under control (day/night temperature of 25/20 °C) and high temperature conditions (35/27 °C) to gain insight into the structural and physiological acclimation controls. Growth at high temperature resulted in larger and thinner leaves with smaller and more densely packed chloroplasts and with lower leaf dry mass per area (MA). High growth temperature also led to lower photosynthetic and respiration rates, isoprene emission rate and leaf pigment content and isoprene substrate dimethylallyl diphosphate pool size per unit area, but to greater stomatal conductance. However, all physiological characteristics were similar when expressed per unit dry mass, indicating that the area‐based differences were primarily driven by MA. Acclimation to high temperature further increased heat stability of photosynthesis and increased activation energies for isoprene emission and isoprene synthase rate constant. This study demonstrates that temperature acclimation of photosynthetic and isoprene emission characteristics per unit leaf area were primarily driven by structural modifications, and we argue that future studies investigating acclimation to growth temperature must consider structural modifications.  相似文献

To date a few sequences regulating expression of conifer seed-specific genes have been reported. To characterize Douglas-fir (Pseudotsuga menziesii [Mirb] Franco) 2S albumin storage protein genes, a genomic DNA sequence containing upstream promoter sequences was isolated by screening a Douglas-fir genomic library. Sequence analysis of the Douglas-fir gPm2S1 promoter revealed the presence of RY-repeated elements (GCATGC), and multiple E-box motifs (CANNTG) and ACGT-core elements, features characteristic of 2S storage protein genes in angiosperms. When fused to the GUS reporter gene, the 1.16 kb Douglas-fir 2S promoter sequence was sufficient to direct transient expression in both developing Douglas-fir embryos and maternally derived haploid megagametophytes. Analysis of this promoter construct in transgenic tobacco showed that expression was restricted to embryo and endosperm in developing seeds and was not detected in vegetative tissues of two-week-old seedlings. These results strongly suggest that both structural and regulatory elements as well as upstream signaling components controlling the expression of 2S albumin genes are highly conserved during evolution.  相似文献

Using the expression of fluorescence originated from the PSII open reaction center in the light by Oxborough and Baker (1997), we obtained a formula that expresses relationships between the quantum efficiency of PSII photochemistry in the dark (Phi(m)= F(v)/F(m)) and in the light Phi'(m)=F'(v)/F'(m):Phi'(m)=Phi(m)+L(NP), where L(NP)(=F(0)/F'(m)) denotes the quantum yield of light induced non-photochemical losses (heat dissipation and fluorescence de-excitation) in PSII. Using L(NP) and other conventional fluorescence parameters, we conducted quenching analyses with leaves of broad bean plants (Vicia faba L.) grown at 700 (high light; HL) and 80 mumol photons m(-2) s(-1) (low light; LL). We also examined whether behavior of q(0) quenching (q(0)=1-F'(0)/F(0)) is related to the reaction center quenching. When the actinic light (AL) was strong, Stern-Volmer quenching [NPQ=(F(m)-F'(m))/F'(m)] and L(NP) increased rapidly and then decreased slowly in HL leaves, while, in LL leaves, they increased slowly. It is probable that rapid formation of a large proton gradient was responsible for sharp rises in both parameters in HL leaves. The steady-state 'excess' parameter [Phi(Ex)= (1 - qP) Phi(m)/(Phi(m)+ L(NP))], fraction of energy migrating to closed PSII centers, increased with the photon flux density of AL in LL leaves. In contrast, in HL leaves, Phi(Ex) did not increase markedly. The examination of the relationship between Phi(Ex) and L(NP) obtained at various AL revealed that in LL leaves the increase in (1 - qP) with the increase in AL prevailed, while, in HL leaves, the increase in L(NP) suppressed the increase in (1 - qP). Using the difference between L(NP) and L(D) (Phi(ND)= L(NP)- L(D), where L(D)= F(0)/F(m)), q(0) and qN (=1-F'(v)/F(v)) were calculated without using measured F'(0). The relationships between q(0) and qN thus obtained for various AL levels were almost identical for both HL and LL leaves, implying no difference in the fluorescence origin between the HL and LL leaves. Usefulness of these equations expressing non-photochemical loss is discussed.  相似文献

Proinflammatory cytokines of the interleukin-36 (IL-36) family are involved in the pathogenesis of different skin diseases in human and mice. Administration of exogenous IL-36 receptor antagonist (IL-36RA) may be an approach to therapy of different dermatitises. For its full biological activity, IL-36RA requires cleavage of N-terminal methionine residue. We created three E. coli strains producing IL-36RA coexpressed with E. coli methionine aminopeptidase under control of different promoters. To test the biological activity of IL-36RA from different strains we transfected А549 cells with plasmid carrying the IL-36 receptor gene (IL1RL2). These cells respond to IL-36g treatment with production of IL-8, which can be quantified with ELISA. IL-36RA treatment disrupts IL-36 receptor activation by IL-36g and production of IL-8. Using this system, we proved that IL-36RA from all three producer strains is fully biologically active.  相似文献

Abstract In normal air, illumination with a low level of blue or red light (40 μmol m?2 s?1) did not induce stomatal opening in maize plantlets. In CO2-free air, 40 μmol m?2 s?1 of blue or red light promoted an enhancement in stomatal opening. At the same quantum flux, blue light was more efficient than red light and stomatal closure occurred more rapidly with a significantly shorter lag phase after blue light. Anoxia inhibited light-dependent stomatal opening, even under 320 μmol m?2 s?1 illumination. However, after 60 min of illumination with 40 μmol m?2 s?1 of blue light in anoxia, transient stomatal opening was observed when the plant was returned to darkness and normal air. This transient stomatal opening was weaker after pretreatment with red light. We conclude that a blue-light-dependent process induced under anoxia leads to stomatal opening provided oxygen is present. Possible mechanisms associated with blue-light-effect and the nature of the oxygen-consuming processes are discussed.  相似文献

The phosphorylation of phospholamban from dog myocardium is shown to correlate with the protein sensitivity to tryptic attack both in undamaged myocardium and under conditions of circulatory hypoxia and experimental infarction. In the absence of "exogenous" protein kinase a decreased phosphorylation of phospholamban is observed in the incubation mixture containing 10(-6) M cAMP both for circulatory hypoxia and myocardial infarction. In the latter case the phosphorylation remains diminished in the presence of "exogenous" protein kinase as well. The sarcoplasmic reticulum from damaged myocardium exhibited changes in the velocity of the oxalate-dependent transport of calcium which correlated with the corresponding degree of the phospholamban phosphorylation.  相似文献
