ABSTRACT. The microsporidium, Enterocytozoon bieneusi , causes a severe, debilitating, chronic diarrhea in patients with the acquired immunodeficiency syndrome. Specific diagnosis of intestinal microsporidiosis, especially due to Enterocytozoon , is difficult and there is no known therapy that can completely eradicate this parasite. Preliminary studies indicate that a short term (about 6 months) in vitro culture of this parasite yielding low numbers of spores, may be established by inoculating human lung fibroblasts and/or monkey kidney cell cultures with duodenal aspirates and or biopsy from infected patients. The cultures may subsequently be used for the isolation and molecular analysis of parasite DNA.  相似文献

The role of plant defence proteins in fungal pathogenesis   总被引:4,自引:0,他引:4  
It is becoming increasingly evident that a plant–pathogen interaction may be compared to an open warfare, whose major weapons are proteins synthesized by both organisms. These weapons were gradually developed in what must have been a multimillion-year evolutionary game of ping-pong. The outcome of each battle results in the establishment of resistance or pathogenesis. The plethora of resistance mechanisms exhibited by plants may be grouped into constitutive and inducible, and range from morphological to structural and chemical defences. Most of these mechanisms are defensive, exhibiting a passive role, but some are highly active against pathogens, using as major targets the fungal cell wall, the plasma membrane or intracellular targets. A considerable overlap exists between pathogenesis-related (PR) proteins and antifungal proteins. However, many of the now considered 17 families of PR proteins do not present any known role as antipathogen activity, whereas among the 13 classes of antifungal proteins, most are not PR proteins. Discovery of novel antifungal proteins and peptides continues at a rapid pace. In their long coevolution with plants, phytopathogens have evolved ways to avoid or circumvent the plant defence weaponry. These include protection of fungal structures from plant defence reactions, inhibition of elicitor-induced plant defence responses and suppression of plant defences. A detailed understanding of the molecular events that take place during a plant–pathogen interaction is an essential goal for disease control in the future.  相似文献

We describe in this paper a new genus and species of cricetid rodent from the Atlantic Forest of Brazil, one of the most endangered eco‐regions of the world. The new form displays some but not all synapomorphies of the tribe Oryzomyini, but a suite of unique characteristics is also observed. This new forest rat possesses anatomical characteristics of arboreal taxa, such as very developed plantar pads, but was collected almost exclusively in pitfall traps. Phylogenetic analyses of morphological (integument, soft tissue, cranial, and dental characters) and molecular [nuclear – Interphotoreceptor retinoid binding protein (Irbp) – and mitochondrial – cytochrome b– genes] datasets using maximum likelihood and cladistic parsimony approaches corroborate the inclusion of the new taxon within oryzomyines. The analyses also place the new form as sister species to Eremoryzomys polius, an Andean rat endemic to the Maranon valley. This biogeographical pattern is unusual amongst small terrestrial vertebrates, as a review of the literature points to few other similar examples of Andean–Atlantic Forest pairings, in hylid frogs, Pionus parrots, and other sigmodontine rodents. © 2011 The Linnean Society of London, Zoological Journal of the Linnean Society, 2011, 161 , 357–390.  相似文献

Knowledge of the role of origin‐related, environmental, sex, and age factors on host defence mechanisms is important to understand variation in parasite intensity. Because alternative components of parasite defence may be differently sensitive to various factors, they may not necessarily covary. Many components should therefore be considered to tackle the evolution of host–parasite interactions. In a population of barn owls (Tyto alba), we investigated the role of origin‐related, environmental (i.e. year, season, nest of rearing, and body condition), sex, and age factors on 12 traits linked to immune responses [humoral immune responses towards sheep red blood cells (SRBC), human serum albumin (HSA) and toxoid toxin TT, T‐cell mediated immune response towards the mitogen phytohemagglutinin (PHA)], susceptibility to ectoparasites (number and fecundity of Carnus haemapterus, number of Ixodes ricinus), and disease symptoms (size of the bursa of Fabricius and spleen, proportion of proteins that are immunoglobulins, haematocrit and blood concentration in leucocytes). Cross‐fostering experiments allowed us to detect a heritable component of variation in only four out of nine immune and parasitic parameters (i.e. SRBC‐ and HSA‐responses, haematocrit, and number of C. haemapterus). However, because nestlings were not always cross‐fostered just after hatching, the finding that 44% of the immune and parasitic parameters were heritable is probably an overestimation. These experiments also showed that five out of these nine parameters were sensitive to the nest environment (i.e. SRBC‐ and PHA‐responses, number of C. haemapterus, haematocrit and blood concentration in leucocytes). Female nestlings were more infested by the blood‐sucking fly C. haemapterus than their male nestmates, and their blood was less concentrated in leucocytes. The effect of year, season, age (i.e. reflecting the degree of maturation of the immune system), brood size, position in the within‐brood age hierarchy, and body mass strongly differed between the 12 parameters. 1. This study utilised an associative analysis (AA) technique named market basket analysis (MBA) to investigate whether particular grasshopper (Orthoptera: Acrididae) species associations are common during outbreaks (>9.6 grasshoppers m?2) that last >3 years. This study is the first of its kind to use MBA on animal communities. 2. A subset of the 17 years of grasshopper density data from the Wyoming Grasshopper Survey Dataset was used to explore associations among grasshopper species. 3. Associations of certain species were found with over 80% confidence. Life‐history traits of those species commonly found together were examined and compared (a posteriori), creating opportunities to hypothesise certain ecological relationships (e.g. interspecific competition, indirect mutualism) for future studies. 4. This case study shows that further MBA analysis of grasshopper assemblages should prove useful in discovering ecological relationships of grasshopper species during outbreaks. Extracellular phenoloxidase enzymes play an important role in the stability of soil carbon storage by contributing to the cycling of complex recalcitrant phenolic compounds. Climate warming could affect peatland functioning through an alteration of polyphenol/phenoloxidase interplay, which could lead them to becoming weaker sinks of carbon. Here, we assessed the seasonal variability of total phenolics and phenoloxidases subjected to 2–3 °C increase in air temperature using open‐top chambers. The measurements were performed along a narrow fen–bog ecological gradient over one growing season. Climate warming had a weak effect on phenoloxidases, but reduced phenolics in both fen and bog areas. Multivariate analyses revealed a split between the areas and also showed that climate warming exacerbated the seasonal variability of polyphenols, culminating in a destabilization of the carbon cycle. A negative relationship between polyphenols and phenoloxidases was recorded in controls and climate treatments suggesting an inhibitory effect of phenolics on phenoloxidases. Any significant decrease of phenolics through repeatedly elevated temperature would greatly impact the ecosystem functioning and carbon cycle through an alteration of the interaction of polyphenols with microbial communities and the production of extracellular enzymes. Our climate treatments did not have the same impact along the fen–bog gradient and suggested that not all the peatland habitats would respond similarly to climate forcing.  相似文献

Despite strong empirical evidence of the harmful effects of inbreeding on fecundity, spontaneous recessive deleterious mutations are generally considered as acting on survival only in evolutionary models and population viability analyses. In this study, we modelled a species with separate sexes to assess the effect of selection on fecundity in small populations on the risk of extinction. We showed that the impact of inbreeding on short-term fitness changes and that population dynamics are strongly influenced by phenotypic interactions among males and females during reproduction. In particular, population persistence was found to be highly sensitive to the level at which selection acts (i.e. individual vs. pair) and to asymmetry among sexes (in terms of mutation rates and mutational effects).  © 2005 The Linnean Society of London, Biological Journal of the Linnean Society , 2005, 86 , 467–476.  相似文献

Studies on Differentiation Without Cleavage in Chaetopterus   总被引:2,自引:0,他引:2  
Aphidicolin, a powerful inhibitor of nuclear DNA replication, has been used to establish the level of polyploidisation required for the realization of the late morphogenetic events (segregation, pseudogastrulation and ciliation) of differentiation without cleavage in Chaetopterus -activated eggs. A parallel has been drawn between cytophotometric estimations of the DNA content and cytoplasmic differentiation in eggs treated with aphidicolin under different experimental conditions: either pulses with aphidicolin were followed by culture in sea water or the eggs were treated with aphidicolin after development had taken place in sea water for various lengths of time. The results suggest that a'quantal' monasterial cycle might take place 3 h after activation, corresponding presumably to the fourth or fifth replication cycle. Moreover, early DNA replication seems to be more important for morphogenesis than late DNA replication.  相似文献

Moral status is a vexing topic. Linked for so long to the unending debates about ensoulment and the morality of abortion, it has recently resurfaced in the embryonic stem cell controversy. In this new context, it should benefit from new insights originating in recent scientific advances. We believe that the recently observed capability of somatic cells to return to a pluripotential state (a capability we propose to name 'reversed potency') in a controlled manner requires us to modify the traditional concept of moral status and to consider it as referring not only to intrinsic properties (like 'to possess reason' or 'to be a person'), but also to extrinsic or relational ones.  相似文献

Abstract We expected that in clonal plants with the phalanx growth form, prereproductive ramets function as a meristem bank that helps buffer ramet demography against environmental fluctuations. Specifically, we evaluated four expectations for the forest understorey clonal palm Geonoma brevispatha in a swamp forest in south‐eastern Brazil. Our expectations were are follows: (i) the vital rates of prereproductive ramets are more responsive to genet‐related characteristics (genet size, distance and size of the nearest genet neighbour, and reproductive ramet survivorship) than to environmental variables (canopy openness, basal area of small trees, flooding, and distance and size of the nearest large tree neighbour), whose effects are expected to affect ramet dynamics indirectly through physiological integration; (ii) the progress of prereproductive ramets to later stages is dependent on the death of reproductive ramets; (iii) the population dynamics of G. brevispatha is resistant to significant interannual changes in the vital rates of ramets, (i.e. the occurrence of good and bad years); and (iv) there are large negative covariances among demographic transitions within a given year, as well as between years, reflecting the flexibility of G. brevispatha to adjust to environmental variability. Expectations 1, 2 and 4 were confirmed, while expectation 3 was only partially confirmed. Matrix models predicted that the growth rate of the overall ramet population was significantly greater than unity in the first study year, but it was significantly smaller than 1.0 in the second study year. Ramet population growth rates at the level of individual genets were greater than unity in the majority of genets in the first year, but in the second year genets with growth rates smaller than unity prevailed. Ramet population dynamics seems to be resistant to the succession of good and bad years; in effect, the presence of a meristem bank of ramets helps stabilize λ.  相似文献
