We studied the mechanism of sphingosylphosphorylcholine (SPC)-induced contraction in feline ileal smooth muscle cells. Western blotting revealed that G protein subtypes of Galpha(i1), Galpha(i3) and Galpha(o) existed in feline ileum. Galpha(i3) antibody penetration into permeabilized cells decreased SPC-induced contraction. In addition, incubation of [35S]guanosine 5'-O-(3-thiotriphosphate) ([35S]GTPgammaS) with membrane fraction increased its binding to Galpha(i3) subtype after SPC treatment, suggesting that the signalling pathways invoked by SPC were mediated by Galpha(i3) protein. MAPK kinase (MEK) inhibitor PD98059 blocked the contraction significantly, but p38 mitogen-activated protein kinase (MAPK) inhibitor SB202190 did not. Chelerythrine and neomycin also inhibited the contraction. However, cotreatment of PD98059 and chelerythrine showed no significant difference. Phosphorylation of p44/42 MAPK was increased by SPC treatment, which was reversed by pretreatment of inhibitors of signalling molecules that decreased SPC-induced contraction previously. The same result was obtained in the assay of MAPK activity.  相似文献

We investigated the alteration of signal transduction after acute esophagitis in cat lower esophageal sphincter (LES). Acute esophagitis (AE) was induced by perfusion with 0.1N HCl at a rate of 1 ml/min for 45 min over three consecutive days. Acetylcholine (ACh)-induced contraction was inhibited by M3> M1 or M2 antagonists in normal LES. In AE, inhibition by M2 antagonists increased significantly, so that contraction was inhibited by M3> M2> M1 antagonists and the expression of M2 and M3 receptors were increased when compared to normal LES. In normal cells, ACh-induced contractions were antagonized by antibody against G(q/11) and the phosphatidylinositol-specific phospholipase C (PI-PLC) antagonist, U73122. The phosphatidylcholine-specific phospholipase C (PC-PLC) inhibitor, D609, or the phospholipase D inhibitor, propranolol had no effects on contraction in normal LES. However, in AE, G(q/11), and G(i3) antibodies reduced ACh-induced contraction and U73122, propranolol and D609 also reduced the contraction. In AE, we found that the expressions of G protein subtypes were increased but the expression of PLCbeta1, and PLCgamma1 were decreased when compared to normal LES. In conclusion, experimental esophagitis may alter the signal transduction by ACh in LES. ACh-induced contraction is mediated by M3 receptor, G(q/11) and PI-PLC in normal LES. However, in AE, the contractions are mediated by M2, M3 receptor, G(q/11) and G(i3). PC-PLC and PLD as PI-PLC are also involved in ACh-induced cell contraction in AE.  相似文献

Adaptive radiation unfolds as selection acts on the genetic variation underlying functional traits. The nature of this variation can be revealed by studying the tips of an ongoing adaptive radiation. We studied genomic variation at the tips of the Darwin's finch radiation; specifically focusing on polymorphism within, and variation among, three sympatric species of the genus Geospiza. Using restriction site‐associated DNA (RAD‐seq), we characterized 32 569 single‐nucleotide polymorphisms (SNPs), from which 11 outlier SNPs for beak and body size were uncovered by a genomewide association study (GWAS). Principal component analysis revealed that these 11 SNPs formed four statistically linked groups. Stepwise regression then revealed that the first PC score, which included 6 of the 11 top SNPs, explained over 80% of the variation in beak size, suggesting that selection on these traits influences multiple correlated loci. The two SNPs most strongly associated with beak size were near genes associated with beak morphology across deeper branches of the radiation: delta‐like 1 homologue (DLK1) and high‐mobility group AT‐hook 2 (HMGA2). Our results suggest that (i) key adaptive traits are associated with a small fraction of the genome (11 of 32 569 SNPs), (ii) SNPs linked to the candidate genes are dispersed throughout the genome (on several chromosomes), and (iii) micro‐ and macro‐evolutionary variation (roots and tips of the radiation) involve some shared and some unique genomic regions.  相似文献

Effective visual communication requires signals that are easyto detect, transmit, receive, and discriminate. Animals canincrease the probability that their visual signals would bedetected by evolving signals that contrast with their visualbackground. Animals can further enhance this contrast by behaviorallymodifying the existing visual background. Male golden-collaredmanakins (Manacus vitellinus) clear leaf litter from the groundto form courts, which are used as display arenas. Using reflectancemeasures of the signal (male plumage) and the visual background(cleared court and adjacent litter), the irradiance measuresof ambient light during display, and published measures of photoreceptorsensitivity of a Passerine, we test the hypothesis that court-clearingaugments the contrast between male plumage and the visual background.We find that the chromatic and brightness contrasts of goldenpatches used during courtship are greater against the clearedcourt than against adjacent litter. In addition, we find thatcleared courts provide a less variable background for thesecolor patches, resulting in displays that consistently contrastthe visual background. These results suggest that behavioralmodification of the visual background may act to increase theconspicuousness of colorful male plumage during display, providingan explanation for why golden-collared manakins, and possiblyother species, build or clear display courts.  相似文献

The conspicuousness of animal signals is influenced by their contrast against the background. As such, signal conspicuousness will tend to vary in nature because habitats are composed of a mosaic of backgrounds. Variation in attractiveness could result in variation in conspecific mate choice and risk of predation, which, in turn, may create opportunities for balancing selection to maintain distinct polymorphisms. We quantified male coloration, the absorbance spectrum of visual pigments and the photic environment of Poecilia parae, a fish species with five distinct male color morphs: a drab (i.e., grey), a striped, and three colorful (i.e., blue, red and yellow) morphs. Then, using physiological models, we assessed how male color patterns can be perceived in their natural visual habitats by conspecific females and a common cichlid predator, Aequidens tetramerus. Our estimates of chromatic and luminance contrasts suggest that the three most colorful morphs were consistently the most conspicuous across all habitats. However, variation in the visual background resulted in variation in which morph was the most conspicuous to females at each locality. Likewise, the most colorful morphs were the most conspicuous morphs to cichlid predators. If females are able to discriminate between conspicuous prospective mates and those preferred males are also more vulnerable to predation, variable visual habitats could influence the direction and strength of natural and sexual selection, thereby allowing for the persistence of color polymorphisms in natural environments.  相似文献

Recent evidence suggests that males adjust their sexually selecteddisplay traits in response to female behaviors during courtships.Little is known, however, about whether females signal to influencemale displays and whether females benefit from this interaction.Male courtship displays in the satin bowerbird (Ptilonorhynchusviolaceus) are highly intense and aggressive. Females may usethese displays as indicators of mating benefits, but these displaysoften startle females and disrupt courtship. Previous studieshave shown that successful males decrease female startling byadjusting their display intensity according to female crouchingbehaviors, suggesting that crouching behaviors function as signals.Here we address whether female crouching is a signal by usingobservations of natural courtship behaviors. In addition, weexamine why females differ in signaling and whether femalesbenefit from signaling. First, we find that female crouchingis related to the likelihood that females will be startled bymale displays, suggesting that crouching signals the degreeof display intensity that females will tolerate from a malewithout being startled. Second, we find that female tolerancefor intense display increases during successive courtships asfemales assess potential mates, and that female tolerance mayalso be affected by age and condition. Third, we find evidencethat females that reduce startling by signaling their intensitytolerance are more efficient in mate searching. These resultssuggest that females signal to influence how males display theirsexually selected traits, and by doing so, females may increasetheir benefits in mate choice.  相似文献

A new nitrogen-containing bicyclic spirosesquiterpene designated exiguamide which inhibited cell fate specification during sea urchin embryogenesis has been isolated from the marine sponge Geodia exigua.
