Molecular evolution of the mammalian ribosomal protein gene, RPS14   总被引:4,自引:0,他引:4  
Ribosomal protein S14 genes (RPS14) in eukaryotic species from protozoa to primates exhibit dramatically different intron-exon structures yet share homologous polypeptide-coding sequences. To recognize common features of RPS14 gene architectures in closely related mammalian species and to evaluate similarities in their noncoding DNA sequences, we isolated the intron-containing S14 locus from Chinese hamster ovary (CHO) cell DNA by using a PCR strategy and compared it with human RPS14. We found that rodent and primate S14 genes are composed of identical protein-coding exons interrupted by introns at four conserved DNA sites. However, the structures of corresponding CHO and human RPS14 introns differ significantly. Nonetheless, individual intron splice donor, splice acceptor, and upstream flanking motifs have been conserved within mammalian S14 homologues as well as within RPS14 gene fragments PCR amplified from other vertebrate genera (birds and bony fish). Our data indicate that noncoding, intronic DNA sequences within highly conserved, single-copy ribosomal protein genes are useful molecular landmarks for phylogenetic analysis of closely related vertebrate species.   相似文献

To investigate the participation of purinergic P2 receptors in the regulation of renal function in ANG II-dependent hypertension, renal and glomerular hemodynamics were evaluated in chronic ANG II-infused (14 days) and Sham rats during acute blockade of P2 receptors with PPADS. In addition, P2X1 and P2Y1 protein and mRNA expression were compared in ANG II-infused and Sham rats. Chronic ANG II-infused rats exhibited increased afferent and efferent arteriolar resistances and reductions in glomerular blood flow, glomerular filtration rate (GFR), single-nephron GFR (SNGFR), and glomerular ultrafiltration coefficient. PPADS restored afferent and efferent resistances as well as glomerular blood flow and SNGFR, but did not ameliorate the elevated arterial blood pressure. In Sham rats, PPADS increased afferent and efferent arteriolar resistances and reduced GFR and SNGFR. Since purinergic blockade may influence nitric oxide (NO) release, we evaluated the role of NO in the response to PPADS. Acute blockade with N(ω)-nitro-l-arginine methyl ester (l-NAME) reversed the vasodilatory effects of PPADS and reduced urinary nitrate excretion (NO(2)(-)/NO(3)(-)) in ANG II-infused rats, indicating a NO-mediated vasodilation during PPADS treatment. In Sham rats, PPADS induced renal vasoconstriction which was not modified by l-NAME, suggesting blockade of a P2X receptor subtype linked to the NO pathway; the response was similar to that obtained with l-NAME alone. P2X1 receptor expression in the renal cortex was increased by chronic ANG II infusion, but there were no changes in P2Y1 receptor abundance. These findings indicate that there is an enhanced P2 receptor-mediated vasoconstriction of afferent and efferent arterioles in chronic ANG II-infused rats, which contributes to the increased renal vascular resistance observed in ANG II-dependent hypertension.  相似文献

Background

Rift Valley Fever (RVF) is a viral zoonosis that historically affects livestock production and human health in sub-Saharan Africa, though epizootics have also occurred in the Arabian Peninsula. Whilst an effective live-attenuated vaccine is available for livestock, there is currently no licensed human RVF vaccine. Replication-deficient chimpanzee adenovirus (ChAd) vectors are an ideal platform for development of a human RVF vaccine, given the low prevalence of neutralizing antibodies against them in the human population, and their excellent safety and immunogenicity profile in human clinical trials of vaccines against a wide range of pathogens.

Methods

Here, in BALB/c mice, we evaluated the immunogenicity and efficacy of a replication-deficient chimpanzee adenovirus vector, ChAdOx1, encoding the RVF virus envelope glycoproteins, Gn and Gc, which are targets of virus neutralizing antibodies. The ChAdOx1-GnGc vaccine was assessed in comparison to a replication-deficient human adenovirus type 5 vector encoding Gn and Gc (HAdV5-GnGc), a strategy previously shown to confer protective immunity against RVF in mice.

Results

A single immunization with either of the vaccines conferred protection against RVF virus challenge eight weeks post-immunization. Both vaccines elicited RVF virus neutralizing antibody and a robust CD8+ T cell response.

Conclusions

Together the results support further development of RVF vaccines based on replication-deficient adenovirus vectors, with ChAdOx1-GnGc being a potential candidate for use in future human clinical trials.

Flow cytometry sex-sorting technology was developed in 1989. However, it is only the bovine species in which offspring of the desired sex are obtained at a commercial level. The aim of the present work was to evaluate efficiency parameters when using fresh sexed semen in a large-scale equine commercial embryo transfer program. During the 2009, 2010 and 2011 breeding seasons, 938 synchronized cycles were artificially inseminated. One hundred (10.6%) mares failed to ovulate, and for the remaining 838 useable cycles, 887 doses of sexed semen were used, representing 1.06 doses per cycle. In general, 435 (51.9%) out of 838 flushing performed resulted in the recovery of at least one embryo and 496 (59.1%) embryos were recovered, including twins and triplets. Pregnancy rate at 25 days achieved 81.5% (one embryo transferred per recipient). Embryo recovery rate was not statistically different either between preovulatory and postovulatory artificially inseminated mares or when increased quantities of sexed sperm per dose were used (15–45 million) (P > 0.05). A broad variation in embryo recovery rate was observed between the different stallions used in this study. Sex accuracy of the sex sorting assessed by ultrasound fetal sex determination was 90.3%. Finally, overall efficiency (female embryo pregnancies per useable cycles) was 39% (325/838), meaning that to obtain a female pregnancy of at least 75 days it was necessary to perform 2.5 flushing.  相似文献

The artero-venous system is often stressed by accelerative perturbation, not only during exceptional performances, but also in normal life. For example, when the body is subject to fast pressure changes, accelerative perturbations combined with a change in hydrostatic pressure could have severe effects on the circulation. In such cases a preliminary mathematical inquiry, whose results allow qualitative evaluation of the perturbation produced is useful. Pressure variations are studied in this work when the body is subjected both to rectilinear and rotational movements as well as posture change. The dominant modes of the hemodynamic oscillations are emphasized and the numerical simulation results presented. The artery model used for simulation is obviously simplified with respect to the anatomical structure of an artery. Nevertheless, behavior of the main arteries (like the common carotid and aorta) can be approximately described, choosing suitable model parameters. The frequency of blood oscillations strictly depends on the Young modulus of the arterial wall. This connection could be employed for new clinical tests on the state of the arteries.  相似文献

Cobalamin (vitamin B12) is a complex metabolite and essential cofactor required by many branches of life, including most eukaryotic phytoplankton. Algae and other cobalamin auxotrophs rely on environmental cobalamin supplied from a relatively small set of cobalamin-producing prokaryotic taxa. Although several Bacteria have been implicated in cobalamin biosynthesis and associated with algal symbiosis, the involvement of Archaea in cobalamin production is poorly understood, especially with respect to the Thaumarchaeota. Based on the detection of cobalamin synthesis genes in available thaumarchaeotal genomes, we hypothesized that Thaumarchaeota, which are ubiquitous and abundant in aquatic environments, have an important role in cobalamin biosynthesis within global aquatic ecosystems. To test this hypothesis, we examined cobalamin synthesis genes across sequenced thaumarchaeotal genomes and 430 metagenomes from a diverse range of marine, freshwater and hypersaline environments. Our analysis demonstrates that all available thaumarchaeotal genomes possess cobalamin synthesis genes, predominantly from the anaerobic pathway, suggesting widespread genetic capacity for cobalamin synthesis. Furthermore, although bacterial cobalamin genes dominated most surface marine metagenomes, thaumarchaeotal cobalamin genes dominated metagenomes from polar marine environments, increased with depth in marine water columns, and displayed seasonality, with increased winter abundance observed in time-series datasets (e.g., L4 surface water in the English Channel). Our results also suggest niche partitioning between thaumarchaeotal and cyanobacterial ribosomal and cobalamin synthesis genes across all metagenomic datasets analyzed. These results provide strong evidence for specific biogeographical distributions of thaumarchaeotal cobalamin genes, expanding our understanding of the global biogeochemical roles played by Thaumarchaeota in aquatic environments.  相似文献

Epilepsy is characterized by paradoxical patterns of neural activity. They may cause different types of electroencephalogram (EEG), which dynamically change in shape and frequency content during the temporal evolution of seizure. It is generally assumed that these epileptic patterns may originate in a network of strongly interconnected neurons, when excitation dominates over inhibition. The aim of this work is to use a neural network composed of 50 x 50 integrate-and-fire neurons to analyse which parameter alterations, at the level of synapse topology, may induce network instability and epileptic-like discharges, and to study the corresponding spatio-temporal characteristics of electrical activity in the network. We assume that a small group of central neurons is stimulated by a depolarizing current (epileptic focus) and that neurons are connected via a Mexican-hat topology of synapses. A signal representative of cortical EEG (ECoG) is simulated by summing the membrane potential changes of all neurons. A sensitivity analysis on the parameters describing the synapse topology shows that an increase in the strength and in spatial extension of excitatory vs. inhibitory synapses may cause the occurrence of travelling waves, which propagate along the network. These propagating waves may cause EEG patterns with different shape and frequency, depending on the particular parameter set used during the simulations. The resulting model EEG signals include irregular rhythms with large amplitude and a wide frequency content, low-amplitude high-frequency rapid discharges, isolated or repeated bursts, and low-frequency quasi-sinusoidal patterns. A slow progressive temporal variation in a single parameter may cause the transition from one pattern to another, thus generating a highly non-stationary signal which resembles that observed during ECoG measurements. These results may help to elucidate the mechanisms at the basis of some epileptic discharges, and to relate rapid changes in EEG patterns with the underlying alterations at the network level.  相似文献

This article selectively reviews research concerning nicotine's effects on cognition, including the neurobiological mechanism for these effects, task and experimental features that may be important for elucidating these effects, and why these effects may have amplified motivational significance among smokers with cognitive deficit. Nicotine has effects on various cognitive processes, though most studies in humans have focused on the amelioration of cognitive deficits experienced during drug withdrawal. The direct cognitive-enhancing effect of nicotine remains a controversial topic. The relationship between attentional and non-attentional cognitive effects of nicotine is discussed in the context of cognitive self-medication. Further research should include theory-driven examination of cognitive effects of nicotine, and develop targeted smoking cessation programs based on an improved understanding of the role of cognitive self-medication in high-risk individuals.  相似文献

Background  

Clostridial neurotoxins (CNTs) are the most deadly toxins known and causal agents of botulism and tetanus neuroparalytic diseases. Despite considerable progress in understanding CNT structure and function, the evolutionary origins of CNTs remain a mystery as they are unique to Clostridium and possess a sequence and structural architecture distinct from other protein families. Uncovering the origins of CNTs would be a significant contribution to our understanding of how pathogens evolve and generate novel toxin families.  相似文献
