Several bottlenecks in the alcoholic fermentation process must be overcome to reach a very high and competitive performance of bioethanol production by the yeast Saccharomyces cerevisiae. In this paper, a nutritional strategy is described that allowed S. cerevisiae to produce a final ethanol titre of 19% (v/v) ethanol in 45 h in a fed-batch culture at 30 degrees C. This performance was achieved by implementing exponential feeding of vitamins throughout the fermentation process. In comparison to an initial addition of a vitamin cocktail, an increase in the amount of vitamins and an exponential vitamin feeding strategy improved the final ethanol titre from 126 g l(-1) to 135 g l(-1) and 147 g l(-1), respectively. A maximum instantaneous productivity of 9.5 g l(-1) h(-1) was reached in the best fermentation. These performances resulted from improvements in growth, the specific ethanol production rate, and the concentration of viable cells in response to the nutritional strategy.  相似文献

Previous reports have interpreted hybridization between snake satellite DNA and DNA clones from a variety of distant taxonomic groups as evidence for evolutionary conservation, which implies common ancestry (homology) and/or convergence (analogy) to produce the cross- hybridizing sequences. We have isolated 11 clones from a genomic library of Drosophila melanogaster, using a cloned 2.5-kb snake satellite probe of known nucleotide sequence. We have also analysed published sequence data from snakes, mice, and Drosophila. These data show that (1) all of the cross-hybridization between the snake, fly, and mouse clones can be accounted for by the presence of either of two tandem repeats, [GATA]n and [GACA]n and (2) these tandem repeats are organized differently among the different species. We find no evidence that these sequences are homologous apart from the existence of the simple repeat itself, although their divergence from a common ancestral sequence cannot be ruled out. The sequences contain a variety of homogeneous clusters of tandem repeats of CATA, GA, TA, and CA, as well as GATA and GACA. We suggest that these motifs may have arisen by a self-accelerating process involving slipped-strand mispairing of DNA. Homogeneity of the clusters might simply be the result of a rate of accumulation of tandem repeats that exceeds that of other mutations.   相似文献

A strain of the fission yeast Schizosaccharomyces pombe was aerobically grown in a cell-recycle fermentor under various operating conditions, i.e., different bleeding rates and various separate feed rates of glucose and basal medium. Carbon and energy balances were analyzed during steady-state culture regimes, allowing growth yields and maintenance coefficients to be determined under glucose-limited and glucose-excess environments. Special attention was given to the metabolic shift from purely oxidative to respirofermentative glucose catabolism resulting from a change in the growth-limiting factor. No maintenance requirements for the carbon source and for energy were observed during glucose-limited culture regimes and oxidative catabolism. Under glucose excess and respirofermentative metabolism, the m(G) coefficient was shown to be growth-linked, whereas the enhancement of the apparent m(e) coefficient observed for increased residual glucose concentrations could be assigned to a decline in the ATP yield. Fertilizer N availability impacts photosynthesis and crop performance, although cause–effect relationships are not well established, especially for field-grown plants. Our objective was to determine the relationship between N supply and photosynthetic capacity estimated by leaf area index (LAI) and single leaf photosynthesis using genetically diverse field-grown maize (Zea mays L.) hybrids. We compared a high yield potential commercial hybrid (FR1064 x LH185) and an experimental hybrid (FR1064 x IHP) with low yield potential but exceptionally high grain protein concentration. Plant biomass and physiological traits were measured at tassel emergence (VT) and at the grain milk stage (R3) to assess the effects of N supply on photosynthetic source capacity and N uptake, and grain yield and grain N were measured at maturity. Grain yield of FR1064 x LH185 was much greater than FR1064 x IHP even though plant biomass and LAI were larger for FR1064 x IHP, and single leaf photosynthesis was similar for both hybrids. Although photosynthetic capacity was not related to hybrid differences in productivity, increasing N supply led to proportional increases in grain yield, plant biomass, LAI, photosynthesis, and Rubisco and PEP carboxylase activities for both hybrids. Thus, a positive relationship between photosynthetic capacity and yield was revealed by hybrid response to N supply, and the relationship was similar for hybrids with a marked difference in yield potential. For both hybrids the N response of single leaf CER and initial Rubisco activity was negative when expressed per unit of leaf N. In contrast, PEP carboxylase activity per unit leaf N increased in response to N availability, indicating that PEP carboxylase served as a reservoir for excess N accumulation in field-grown maize leaves. The correlation between CER and initial Rubisco activity was highly significant when expressed on a leaf area or a total leaf basis. The results suggest that regardless of genotypic yield potential, maize CER, and potentially grain yield, could be improved by increasing the partitioning of N into Rubisco.  相似文献

Summary The influence of initial substrate concentration on fermentation kinetics of Zymomonas mobilis on glucose has been studied in batch cultures over the range 50–190 gl-1 glucose. With increasing glucose, parameters relative to growth ( and RGX) are more rapidly and more noticeably affected than those connected with ethanol production (p and RGP).
