Hepatitis B and hepatitis C viruses (HBV and HCV) are both noncytopathic and can cause acute and chronic infections of the liver. Although they share tropism for the same organ, development of chronic hepatitis is much more frequent following HCV infection, suggesting different mechanisms of viral persistence. In this study, we show that circulating HBV- and HCV-specific tetramer-positive CD8 cells during the acute phase of hepatitis B and C belong almost entirely to an effector-memory subset (CCR7(-) CD45RA(-)). Despite this phenotypic similarity, HBV- and HCV-specific CD8 cells show striking functional differences. HBV-specific tetramer-positive CD8 cells express high perforin content ex vivo, expand vigorously, and display efficient cytotoxic activity and gamma interferon (IFN-gamma) production upon peptide stimulation. A comparable degree of functional efficiency is maintained after the resolution of hepatitis B. In contrast, HCV-specific CD8 cells in the acute phase of hepatitis C express significantly lower levels of perforin molecules ex vivo and show depressed CD8 function in terms of proliferation, lytic activity, and IFN-gamma production, irrespective of the final outcome of the disease. This defect is transient, because HCV-specific CD8 cells can progressively improve their function in patients with self-limited hepatitis C, while the CD8 function remains persistently depressed in subjects with a chronic evolution.  相似文献

This work characterizes differences in selection ofTalisia retusa fruits by two sympatric Neotropical primates,Pithecia pithecia (white-faced sakis) andAlouatta seniculus (red howlers). Color appears to be the criterion by which fruits were selected. Greenish fruits were mainly eaten byPithecia, while yellowish fruits byAlouatta. The characteristics of these primates in relation to seed predation and seed dispersal are discussed in the context of theTalisia retusa fruit color spectrum. Furthermore, a possible differential acquisition of chemical components, like tannins, is hypothetically treated considering the variation in fruit color.  相似文献

Nitric oxide reductases (NORs) that are found in bacteria belong to the large enzyme family which includes cytochrome oxidases. Two types of bacterial NORs have been characterised. One is a cytochrome bc-type complex (cNOR) that receives electrons from soluble redox protein donors, whereas the other type (qNOR) lacks the cytochrome c component and uses quinol as the electron donor. The latter enzyme is present in several pathogens that are not denitrifiers. We summarise the current knowledge on bacterial NORs, and discuss the evolutionary relationship between them and cytochrome oxidases in this review.  相似文献

The Ubiquitin-Proteasome System (UPS) and the Autophagy-Lysosome Pathways (ALP) are key mechanisms for cellular homeostasis sustenance and protein clearance. A wide number of Neurodegenerative Diseases (NDs) are tied with UPS impairment and have been also described as proteinopathies caused by aggregate-prone proteins, not efficiently removed by proteasome. Despite the large knowledge on proteasome biological role, molecular mechanisms associated with its impairment are still blur. We have pursued a comprehensive proteomic investigation to evaluate the phenotypic rearrangements in protein repertoires associated with a UPS blockage. Different functional proteomic approaches have been employed to tackle UPS impairment impact on human NeuroBlastoma (NB) cell lines responsive to proteasome inhibition by Epoxomicin. 2-Dimensional Electrophoresis (2-DE) separation combined with Mass Spectrometry and Shotgun Proteomics experiments have been employed to design a thorough picture of protein profile. Unsupervised meta-analysis of the collected proteomic data revealed that all the identified proteins relate each other in a functional network centered on beta-estradiol. Moreover we showed that treatment of cells with beta-estradiol resulted in aggregate removal and increased cell survival due to activation of the autophagic pathway. Our data may provide the molecular basis for the use of beta-estradiol in neurodegenerative disorders by induction of protein aggregate removal.  相似文献

A statistical model is presented in which an ionic polymer is taken as a linear arrangement of flexible segments (similar to the spring-bead model). This model is used to represent semi-flexible linear ionic polysaccharides in solution. The distribution of end-to-end distances is taken from Monte Carlo calculations of the amylosic chain conformation and combined with Manning's counterion condensation theory of linear polyelectrolytes. The excess thermodynamic properties are then calculated as a function of the degree of ionization and of a number of physical variables. The results show that the statistical average of thermodynamic functions taken over the conformational states is not equivalent to the thermodynamic function of the average conformation. This has important implications for the correct comparison of theoretical prediction with the experimental results.  相似文献

We studied the diet, activity budget, vertical ranging, and postural behaviour in relation to weather of the three-toed sloth (Bradypus variegatus flaccidus) in disturbed montane forest remnants (1150 m asl) in northern Venezuela. Sloths spent most (72.9%) of their time resting and had a nearly exclusive (99.4%) leaf diet. While resting they assumed a sitting – not hanging – posture mostly (90.2% of observations). Species of three families, Clethraceae, Cecropiaceae, and Clusiaceae accounted for 77% of feeding records. Young leaves (67.2%) accounted for most of the leaf diet. Activity and posture were dependent on weather conditions. Sloths fed more often during mid-day hours and tended to rest more at dawn and dusk. In northern Venezuela sloths tended to use more frequently the upper strata of the canopy, while in warmer lowland sites they use intermediate levels more often. They adopted postures that maximized exposure of ventral surfaces to incident solar radiation when sunny, but minimized their surface area by huddling when cloudy, foggy or rainy. We propose that sunning behaviour of sloths may speed up their fermentation rate, and ultimately, might have been an important selective factor in the evolution of derived upside-down posture of sloths.  相似文献

Protein-adsorptive properties are a key feature of membranes used for hemodialysis treatment. Protein adsorption is vital to the biocompatibility of a membrane material and influences membrane's performance. The object of the present study is to investigate membrane biocompatibility by correlating the adsorbed proteome repertoire with structural feature of the membrane surfaces. Minidialyzers of identical structural characteristics composed of either cellulose diacetate or ethylenevinyl alcohol materials were employed to develop an ex vivo apparatus to investigate protein adsorption. Adsorbed proteins were eluted by a strong chaotropic buffer condition and investigated by 2-DE coupled to both MALDI-TOF mass spectrometry (MS) mass fingerprinting and fragmentation analysis on a nanoLC-MS/MS hybrid instrument. Membrane surface characterization included evaluation of roughness (atomic force microscopy), elemental chemical composition (X-ray-photoelectron-spectroscopy), and hydrophilicity (pulsed nuclear magnetic resonance). The present study identifies a number of different proteins as common or characteristic of filter material interaction, showing that proteomic techniques are a promising approach for the investigation of proteins surface-adsorbed onto hemodialysis membrane. Proteomic analysis enables the characterization of protein layers of unknown composition.  相似文献

Geophagy has been recorded in an increasing number of New World monkeys (Platyrrhini) over recent years, permitting a tentative analysis of ecological patterns. While geophagy has now been recorded in species representing all 4 platyrrhine families and a majority of genera, there is a marked tendency for it to occur in the larger-bodied Pitheciidae and Atelidae. Howlers (Alouatta) are responsible for almost a third of reports, which are concentrated in the more frugivorous species, Alouatta belzebul and Alouatta seniculus. Geophagy may also be relatively common in the spider monkeys (Ateles) and the pitheciids, which are specialised frugivores and seed predators, respectively. An overview of the available data points to a marked Amazonian bias, allowing for geographical differences in the number of species and field studies. This pattern is demonstrated most emphatically by Alouatta, for which there are almost as many reports as field studies in the Amazon basin, in stark contrast with Central American sites, which have a long tradition of fieldwork, but no published records of geophagy. There are also relatively few reports from the Brazilian Atlantic forest. Despite the growth in reports, and the patterns identified here, the functional aspects of geophagy in the platyrrhines still remain unclear.  相似文献
