Rhinovirus type 14 (RV14) incuced a transient statistically significant stimulation in synthesis of DNA which appeared between 0 and 3 h post-inoculation in the cytoplasm of high density monolayer cultures of KB cells. Newly synthesized DNA was measured by incorporation of [3H] thymidine into acid-insoluble DNAase-sensitive material and the cytoplasmic location established by cell fractionation and electron microscope radioautographic methods. A minimum of 10 plaque-forming units per cell of RV14 was required to stimulate DNA synthesis which did not occur above 34.5 degrees C, a temperature optimal for virus replication. Cytoplasmic DNA taken from RV14-infected or control cells could be differentiated from the bulk of cell (nuclear) DNA by several criteria, including: (1) RV14 induction of synthesis; (2) lower buoyant density and greater heterogeneity in CsCl and ethidium bromide/CsCl gradients; and (3) a different kinetic complexity upon reannealing. The Cot 1/2 value of cytoplasmic DNA, calclated as 50--100 from reassociation profiles, was about 10-fold less complex than the Cot 1/2 value of nuclear DNA (800-1000). These data rule out the possibility that cytoplasmic DNA arises by random breakage of nuclear DNA during cell disruption and extraction and are compatible with the hypothesis that inoculation of KB cells with RV14 results in stimulation of synthesis of a specific class of cell DNA which is detected in the cytoplasm. 相似文献

Nitrogen form influences the response of Deschampsia antarctica to dark septate root endophytes 总被引:1,自引:0,他引:1
Fungi with dematiaceous septate hyphae, termed dark septate endophytes (DSE), are common in plant roots, particularly in cold-stressed
habitats, but their effects on their host plants remain obscure. Here, we report a study that assessed the effects of six
DSE on the growth and nutrient balance of Deschampsia antarctica when plants were supplied with the same amount of nitrogen in organic (casein hydrolysate) or inorganic (ammonium sulphate)
form under controlled conditions. After 60 days, the DSE, that had each been isolated from D. antarctica and which analyses of internal transcribed spacer and large subunit regions indicated were similar to members of the Helotiales
(Oculimacula yallundae, Mollisia and Tapesia spp.) and unassigned anamorphic ascomycetes, typically had no effect on, or reduced by 33–71%, shoot and root dry weights
relative to uninoculated controls when plants had been supplied with nitrogen in inorganic form. In contrast, the DSE usually
enhanced shoot and root dry weights by 51–247% when plants had been supplied with organic nitrogen. In the presence of inorganic
nitrogen, only sporadic effects of DSE were recorded on shoot and root nitrogen or phosphorus concentrations, whereas in the
presence of organic nitrogen, three to six of the DSE isolates increased shoot and root nitrogen and phosphorus contents.
Most of the isolates decreased the phosphorus concentrations of shoots and roots when plants had been supplied with nitrogen
in organic form. Our data suggest that DSE are able to mineralise peptides and amino acids in the rhizosphere, making nitrogen more freely

We review the distributions and functions of mycorrhizas and dark septate root endophytes in polar regions. Arbuscular mycorrhizas (AM) are present in the Arctic and Antarctic to 82 °N and 63 °S, respectively, with fine endophyte being the dominant form of AM in roots at higher latitudes. Ecto- (ECM) and ericoid (ERM) mycorrhizas both occur in the Arctic to 79 °N, owing to the presence of species of Salix, Dryas, Vaccinium and Cassiope to this latitude. ECM and ERM are not present in Antarctic ecosystems, owing to an absence of suitable hosts. Arbutoid and orchid mycorrhizas are infrequent in the Arctic, whilst the latter are present at one location in the sub-Antarctic. Data from studies of AM, ECM and ERM colonisation along a latitudinal transect through the Arctic indicate that the frequency of plant species not colonised by mycorrhizas increases at higher latitudes, largely owing to an increase in non-mycorrhizal and a decrease in obligately mycorrhizal plant families at more northerly locations. A separate group of root- and rhizoid-associated fungi, the dark septate root endophytes (DSE), are widespread to 82 °N and 77 °S, and are apparently more frequent than mycorrhizal fungi in polar regions. The functions of DSE are largely unclear, but studies suggest beneficial effects on plant growth under defined conditions. Although dark septate fungal endophytes (DSE) occur widely in association with plant roots in cold-stressed habitats, little is known of the taxonomic status of DSE in Antarctica. Here we investigate the phylogenetic affinities of DSE colonising the roots of Colobanthus quitensis and Deschampsia antarctica, the two maritime Antarctic vascular plant species. Two hundred and forty-three DSE were isolated from roots collected from 17 sites across a 1 470 km transect through maritime and sub-Antarctica. The ITS1-5.8S-ITS2 nuclear ribosomal gene cluster of representative isolates was sequenced, and the sequences were recovered in 10 sequence groups and sub-groups. Nine of the sequence groupings could be placed in the Helotiales and the remaining one showed high homology to a large number of currently unassigned anamorphic ascomycete sequences. Of the Helotiales, Leptodontidium orchidicola, Rhizoscyphus ericae and species of Tapesia and Mollisia could be confidently identified. This study demonstrates that members of the Helotiales, including several widely-recognised DSE genera, commonly colonise the roots of C. quitensis and D. antarctica in the Antarctic. 相似文献

In a previous paper (Yang et al., Biophys. J. 75:641-645, 1998), we showed a simple, efficient method of recording the diffraction patterns of supramolecular peptide assemblies in membranes where the samples were prepared in the form of oriented multilayers. Here we develop a method of analysis based on the diffraction theory of two-dimensional liquids. Gramicidin was used as a prototype model because its pore structure in membrane in known. At full hydration, the diffraction patterns of alamethicin and magainin are similar to gramicidin except in the scale of q (the momentum transfer of scattering), clearly indicating that both alamethicin and magainin form pores in membranes but of different sizes. When the hydration of the multilayer samples was decreased while the bilayers were still fluid, the in-plane positions of the membrane pores became correlated from one bilayer to the next. We believe that this is a new manifestation of the hydration force. The effect is most prominent in magainin patterns, which are used to demonstrate the method of analysis. When magainin samples were further dehydrated or cooled, the liquid-like diffraction turned into crystal-like patterns. This discovery points to the possibility of investigating the supramolecular structures with high-order diffraction. 相似文献
