1. In ecological webs, net indirect interactions between species are composed of interactions that vary in sign and magnitude. Most studies have focused on negative component interactions (e.g. predation, herbivory) without considering the relative importance of positive interactions (e.g. mutualism, facilitation) for determining net indirect effects. 2. In plant/arthropod communities, ants have multiple top-down effects via mutualisms with honeydew-producing herbivores and harassment of and predation on other herbivores; these ant effects provide opportunities for testing the relative importance of positive and negative interspecific interactions. We manipulated the presence of ants, honeydew-producing membracids and leaf-chewing beetles on perennial host plants in field experiments in Colorado to quantify the relative strength of these different types of interactions and their impact on the ant's net indirect effect on plants. 3. In 2007, we demonstrated that ants simultaneously had a positive effect on membracids and a negative effect on beetles, resulting in less beetle damage on plants hosting the mutualism. 4. In 2008, we used structural equation modelling to describe interaction strengths through the entire insect herbivore community on plants with and without ants. The ant's mutualism with membracids was the sole strong interaction contributing to the net indirect effect of ants on plants. Predation, herbivory and facilitation were weak, and the net effect of ants reduced plant reproduction. This net indirect effect was also partially because of behavioural changes of herbivores in the presence of ants. An additional membracid manipulation showed that the membracid's effect on ant activity was largely responsible for the ant's net effect on plants; ant workers were nearly ten times as abundant on plants with mutualists, and effects on other herbivores were similar to those in the ant manipulation experiment. 5. These results demonstrate that mutualisms can be strong relative to negative direct interspecific interactions and that positive interactions deserve attention as important components of ecological webs.  相似文献

Breast milk is the ideal nutrition for term infants but must be supplemented to provide adequate growth for most premature infants. Human milk oligosaccharides (HMOs) are remarkably abundant and diverse in breast milk and yet provide no nutritive value to the infant. HMOs appear to have at least two major functions: prebiotic activity (stimulation of the growth of commensal bacteria in the gut) and protection against pathogens. Investigations of HMOs in milk from women delivering preterm have been limited. We present the first detailed mass spectrometric analysis of the fucosylation and sialylation in HMOs in serial specimens of milk from 15 women delivering preterm and 7 women delivering at term using nanohigh performance liquid chromatography chip/time-of-flight mass spectrometry. A mixed-effects model with Levene's test was used for the statistical analyses. We find that lacto-N-tetraose, a core HMO, is both more abundant and more highly variable in the milk of women delivering preterm. Furthermore, fucosylation in preterm milk is not as well regulated as in term milk, resulting in higher within and between mother variation in women delivering preterm vs term. Of particular clinical interest, the α1,2-linked fucosylated oligosaccharide 2'-fucosyllactose, an indicator of secretor status, is not consistently present across lactation of several mothers that delivered preterm. The immaturity of HMO production does not appear to resolve over the time of lactation and may have relevance to the susceptibility of premature infants to necrotizing enterocolitis, late onset sepsis, and related neurodevelopmental impairments.  相似文献

Minisatellites are highly variable tandem repeats used for over 20 years in humans for DNA fingerprinting. In prokaryotes fingerprinting techniques exploiting VNTR (variable number of tandem repeats) polymorphisms have become widely used recently in bacterial typing. However although many investigations into the mechanisms underlying minisatellite variation in humans have been performed, relatively little is known about the processes that mediate bacterial minisatellite polymorphism. An understanding of this is important since it will influence how the results from VNTR experiments are interpreted. The minisatellites of Mycobacterium tuberculosis are well characterized since they are some of the few polymorphic loci in what is otherwise a very homogeneous organism. Using VNTR results from a well-defined and characterized set of M. tuberculosis strains we show that the repeats at a locus are likely to evolve by stepwise contraction or expansion in the number of repeats. A stochastic continuous-time population mathematical model was developed to simulate the evolution of the repeats. This allowed estimation of the tendency of the repeats to increase or decrease and the rate at which they change. The majority of loci tend to lose rather than gain repeats. All of the loci mutate extremely slowly, with an average rate of 2.3 x 10(-8), which is 350 times slower than that of a set of VNTR repeats with similar diversity observed experimentally in Escherichia coli. This suggests that the VNTR profile of a strain of M. tuberculosis will be indicative of its clonal lineage and will be unlikely to vary in epidemiologically-related strains.  相似文献

There is considerable debate about whether stability (e.g. inertia) of an assemblage, or of individuals in an assemblage, is positively associated with the number of species or whether there are idiosyncratic effects of particular species. We assessed the general model that the loss of an individual alga, caused by trampling, is greater in monospecific than in multi-species stands but that the responses of algae are idiosyncratic, depending on the morphology of the species. The experiment was done on conspicuous and dominant algae with different morphology on temperate Australian rocky shores: the fucalean algae Hormosira banksii and Sargassum sp. and the coralline alga Corallina officinalis. We assessed the relative and interactive effects of the extent of trampling (number of paths) and the localised intensity of trampling (number of travels per path) on the three algae. The number of paths trampled (the extent of disturbance) had more impact on each alga than the number of times paths were travelled (the intensity of disturbance). As predicted, H. banksii was most susceptible to trampling at each level than were the coarser algae Sargassum sp. and C. officinalis. There was a consistent trend for each alga to be more inert to trampling when in the presence of the other two species than when in monospecific stands, but this was only statistically significant (P < 0.05) for the softer alga H. banksii. The responses of H. banksii and Sargassum sp. to disturbance seemed, in many cases, to be due to the presence of C. officinalis rather than to "diversity" per se. The relationship between the number of species and stability is complex in intertidal habitats, depending on the species and the combinations of species with which it grows.  相似文献

Objective To determine whether older patients with chronic knee pain should be advised to use topical or oral non-steroidal anti-inflammatory drugs (NSAIDs).Design Randomised controlled trial and patient preference study.Setting 26 general practices.Participants People aged ≥50 with knee pain: 282 in randomised trial and 303 in preference study. Interventions Advice to use topical or oral ibuprofen.Primary outcome measures WOMAC (Western Ontario and McMaster Universities) osteoarthritis index, major and minor adverse effects.Results Changes in global WOMAC scores at 12 months were equivalent. In the randomised trial the difference (topical minus oral) was two points (95% confidence interval −2 to 6); in the preference study, it was one point (−4 to 6). There were no differences in major adverse effects in the trial or study. The only significant differences in secondary outcomes were in the randomised trial. The oral group had more respiratory adverse effects (17% v 7%,95% confidence interval for difference −17% to −2%), the change in serum creatinine was 3.7 mmol/l less favourable (0.9 µmol/l to 6.5 µmol/l); and more participants changed treatments because of adverse effects (16% v 1%, −16% to −5%). In the topical group more participants had chronic pain grade III or IV at three months, and more participants changed treatment because of ineffectiveness.Conclusions Advice to use oral or topical preparations has an equivalent effect on knee pain over one year, and there are more minor side effects with oral NSAIDs. Topical NSAIDs may be a useful alternative to oral NSAIDs.Trial registration ISRCTN 79353052.  相似文献

Limited cell growth and the resulting low volumetric productivity of ethanologenic Escherichia coli KO11 in mineral salts medium containing xylose have been attributed to inadequate partitioning of carbon skeletons into the synthesis of glutamate and other products derived from the citrate arm of the anaerobic tricarboxylic acid pathway. The results of nuclear magnetic resonance investigations of intracellular osmolytes under different growth conditions coupled with those of studies using genetically modified strains have confirmed and extended this hypothesis. During anaerobic growth in mineral salts medium containing 9% xylose (600 mM) and 1% corn steep liquor, proline was the only abundant osmolyte (71.9 nmol ml−1 optical density at 550 nm [OD550] unit−1), and growth was limited. Under aerobic conditions in the same medium, twice the cell mass was produced, and cells contained a mixture of osmolytes: glutamate (17.0 nmol ml−1 OD550 unit−1), trehalose (9.9 nmol ml−1 OD550 unit−1), and betaine (19.8 nmol ml−1 OD550 unit−1). Two independent genetic modifications of E. coli KO11 (functional expression of Bacillus subtilis citZ encoding NADH-insensitive citrate synthase; deletion of ackA encoding acetate kinase) and the addition of a metabolite, such as glutamate (11 mM) or acetate (24 mM), as a supplement each increased the intracellular glutamate pool during fermentation, doubled cell growth, and increased volumetric productivity. This apparent requirement for a larger glutamate pool for increased growth and volumetric productivity was completely eliminated by the addition of a protective osmolyte (2 mM betaine or 0.25 mM dimethylsulfoniopropionate), consistent with adaptation to osmotic stress rather than relief of a specific biosynthetic requirement.  相似文献

Concentrations of heavy metals (zinc, copper and lead) were manipulated experimentally to test the hypotheses about effects on intertidal, soft-sediment assemblages of animals in two sand-flats in Port Hacking, Australia. Hypotheses about changes in the structure and composition of whole assemblages and changes in mean abundance and variability of individual taxa were tested. Specific hypotheses were derived a priori from repeated observations of assemblages in urban and non-urban areas of Port Hacking.After manipulation, concentrations of metals were similar to those in sediments near urban areas. Nevertheless, responses of assemblages to the increased concentrations of metals were weak. Polychaetes, spionids and amphipods responded to experimental treatments. Changes were, however, not consistent among times and places and generally not in agreement with what had been predicted. Significant spatial and temporal variability were detected for all variables investigated. Increased concentrations of metals did not affect variability or overall structure of assemblages. Thus, there was little evidence that increased concentrations of metals caused benthic assemblages in pristine areas to become more similar to those in areas contaminated by human activities.Several potential explanations for the discrepancy between previously observed correlative patterns and the results presented here are discussed. These include a critical assessment of different aspects of the experimental study, such as lack of statistical power, insufficient basis for prediction, artefacts due to experimental procedures and issues to do with the difference between evidence based on correlation and manipulation. Explicit comparisons showed that there were significant effects of physical disturbance due to repeated sampling and that assemblages of animals at the start of the experiment were different from those previously observed in uncontaminated areas. These observations were surprising and made interpretation more difficult. Nevertheless, it is possible that the metals investigated really contribute only marginally to previously observed differences between urban and non-urban areas. Repeated comparisons between observations of patterns and manipulative experiments like these can only improve the basis for prediction and the power of current mechanistic models.  相似文献
