A new insertion sequence (IS), IS 1642 , was identified in a Mycobacterium avium strain isolated from a human patient. IS 1642 had a size of 1642 bp and contained a single ORF encoding a probable transposase of 503 amino acid residues homologous (79% identity) to that of IS 1549 found in Mycobacterium smegmatis . The IS 1642 included imperfect inverted repeats (5'-cctgacttttatca-3', 5'-tgataaaagtcggg-3') on its ends, and was flanked by direct repeats of variable length ranging from 5 to 161 bp. It was suggested that the IS 1642 was widely distributed in many M. avium strains of human patients, and the Southern blot profile of IS 1642 was very diverse among the strains examined. The transposition event of IS 1642 was observed by in vitro repeated passages, showing that the IS 1642 is actually a transposable element. In light of these characteristics, IS 1642 could be a new useful marker when genotyping with high discrimination is required.  相似文献

Asexual development in Aspergillus nidulans begins in superficial hyphae as the programmed emergence of successive pseudohyphal modules, collectively known as the conidiophore, and is completed by a layer of specialized cells (phialides) giving rise to chains of aerial spores. A discrete number of regulatory factors present in hyphae play different stage-specific roles in pseudohyphal modules, depending on their cellular localization and protein-protein interactions. Their multiple roles include the timely activation of a sporulation-specific pathway that governs phialide and spore formation. Such functional versatility provides for a new outlook on morphogenetic change and the ways we should study it.  相似文献

Three ectomycorrhizal (ECM) isolates of Rhizopogon luteolus, R. roseolus and Scleroderma citrinum were found to differ markedly in their in vitro tolerance to adverse conditions limiting fungal growth, i.e. water availability, pH and heavy metal pollution. S. citrinum was the most sensitive, R. luteolus intermediate and R. roseolus the most tolerant species. Pinus radiata D. Don seedlings were inoculated in the laboratory and in a containerised seedling nursery with spore suspensions of the three ECM species. Colonisation percentage was considerably lower under nursery conditions, probably due to competition by native fungi. The effects of nursery ECM inoculation on seedling growth depended on the fungal species. Only R. roseolus-colonised plants showed a significantly higher shoot growth than non-mycorrhizal plants. All three fungi induced significantly higher root dry weights relative to control plants. Despite the low mycorrhizal colonisation, mycorrhization with all three species improved the physiological status of nursery-grown seedlings, e.g. enhanced root enzyme activity, shoot nutrient and pigment content, net photosynthesis rate and water use efficiency. Of the three fungal species, R. roseolus was the most effective; this species was also the most adaptable and showed the greatest range of tolerance to adverse environmental conditions in pure culture. It is, therefore, proposed as a promising fungal species for ECM inoculation of P. radiata in the nursery.  相似文献

Dosidicus gigas (jumbo or Humboldt squid) is a semelparous, major predator of the eastern Pacific that is ecologically and commercially important. In the Gulf of California, these animals mature at large size (>55 cm mantle length) in 1–1.5 years and have supported a major commercial fishery in the Guaymas Basin during the last 20 years. An El Niño event in 2009–2010, was accompanied by a collapse of this fishery, and squid in the region showed major changes in the distribution and life‐history strategy. Large squid abandoned seasonal coastal‐shelf habitats in 2010 and instead were found in the Salsipuedes Basin to the north, an area buffered from the effects of El Niño by tidal upwelling and a well‐mixed water column. The commercial fishery also relocated to this region. Although large squid were not found in the Guaymas Basin from 2010 to 2012, small squid were abundant and matured at an unusually small mantle‐length (<30 cm) and young age (approximately 6 months). Juvenile squid thus appeared to respond to El Niño with an alternative life‐history trajectory in which gigantism and high fecundity in normally productive coastal‐shelf habitats were traded for accelerated reproduction at small size in an offshore environment. Both small and large mature squid, were present in the Salsipuedes Basin during 2011, indicating that both life‐ history strategies can coexist. Hydro‐acoustic data, reveal that squid biomass in this study area nearly doubled between 2010 and 2011, primarily due to a large increase in small squid that were not susceptible to the fishery. Such a climate‐driven switch in size‐at‐maturity may allow D. gigas to rapidly adapt to and cope with El Niño. This ability is likely to be an important factor in conjunction with longerterm climate‐change and the potential ecological impacts of this invasive predator on marine ecosystems.  相似文献

Mycorrhizal inoculation of conifer roots is a key strategy to optimize establishment and performance of forest tree species under both natural and cultivated conditions and also to mitigate transplantation shock. However, despite being a common practice, inoculation in outdoor nursery conditions has been poorly studied. Here, we have evaluated effectiveness of four fungal species (Lactarius deliciosus, Lactarius quieticolor, Pisolithus arhizus, and Suillus luteus) in the production of mycorrhizal Pinus pinaster seedlings in an outdoor commercial nursery and their ability to improve seedling physiology and field performance. All inoculated seedlings showed a significant increase in growth at the end of the nursery stage and these differences remained after 3 years of growth in the field. Differences observed in the content of malondialdehyde, total chlorophyll, carotenoids, anthocyanins, and phenolic compounds from needles of mycorrhizal and control seedlings may reflect a different sensitivity to photo-oxidative damage. We conclude that ectomycorrhizal inoculation improves adaptability to changeable growing conditions of an outdoor nursery and produces a higher quality nursery stock, thereby enhancing seedling performance after planting.  相似文献

Banana, maize, potato and sagu starches were boiled in the presence or absence of plasticizer (glycerol), producing edible films. In vitro digestibility features, amylose content and amylopectin gel filtration behavior of films and parent starches were evaluated. Available starch contents were lower in glycerol-containing films, due to dilution by the plasticizer. Total resistant starch increased in the maize starch-based film but decreased markedly in those prepared from the other starches. Amylose content of banana starch (40%) was about double those of the other starches. Nonetheless, all starch films exhibited similar retrograded resistant starch content. Although film production led to increased -amylolysis rates, these were further augmented by additional film heating, thereby indicating that film-manufacture did not promote complete starch gelatinization. Gel filtration chromatography suggested amylopectin depolymerization after film-making, which may also increase digestion kinetics. The presence of glycerol in the films slowed down starch digestion, a feature of potential dietetic use.  相似文献

The egg production of the copepod Acartia bifilosa was measured and related to environmental variables and food availability in two estuaries located in the same biogeographic region (Bay of Biscay) but showing very strong differences in abiotic and biotic features: the Gironde estuary (France) and the estuary of Mundaka (Spain). The study was conducted during the spring-summer-autumn period of 1994. Food availability was evaluated by analysing the chlorophyll a (Chl a), the particulate organic carbon (POC) and the easily extractable macromolecular compounds such as proteins, carbohydrates and lipids of the seston. The egg production of copepods was estimated from field incubations with natural water, and phytoplankton feeding of adult females was estimated by means of the gut fluorescence method. The nutritional environment of the Gironde was characterised by high amounts of suspended particulate matter (SPM) with low food value, emphasising the mainly detrital origin of the organic matter (OM). In Mundaka, the higher contribution of phytoplankton to the seston led to marked increases in particulate food value accounting for up to 35% of organic matter. The weight-specific egg production was found to be sharply higher in Mundaka (ranging from 0.2 to 0.63×10−3 day−1) than in the Gironde (ranging from 0 to 0.13×10−3 day−1), but the seasonal trend of variations was similar, the highest weight-specific egg production rates occurring in early summer and the lowest in autumn in both estuaries. Egg production was not correlated linearly with temperature since maximal egg production occurred at intermediate temperatures. In Mundaka, the egg production showed a significant positive correlation with the chlorophyll and the Chl/SPM and the POC/SPM ratios. This coupled with higher values of algal food availability (Chl a/SPM: 10 to 1870 μg g−1) and gut fluorescence (between 0.12 and 0.38 ng Chl a Eq ind−1) indicate that a herbivorous diet could cover the energy requirements of A. bifilosa and support egg production. In the Gironde, the algal food availability and the gut fluorescence were lower (Chl a/SPM: 10 to 80 μg g−1; GF: 0.09 and 0.25 ng Chl a Eq ind−1), and the egg production showed significant positive correlation with the particulate food value, suggesting that other sources of carbon rather than phytoplankton were responsible for the observed changes in egg production. Results indicate that the particular seston properties of each system may be responsible for the noticeable differences in A. bifilosa fertility among estuaries.  相似文献

When treated with sodium bicarbonate in aqueous acetone, the compound closes the E-ring to form two products having the 5′-isopropenyl grouping in the α- and β-configurations. By this cycle, rotenone (5′β-rotenone) gives 5′α-epirotenone as well as rotenone, while d-epirotenone (5′β-epirotenone) gives 5′α-rotenone (the antipode of natural rotenone) as well as d-epirotenone.  相似文献

The mycelium is an organised cellular network that develops according to a functionally coherent plan. As it expands, the mycelium is capable of modulating the relative abundance of different cell types to suit the prevailing environmental conditions. This versatile pattern of multicellular development involves sophisticated environmental sensing and intercellular communication systems that have barely been recognised. This review describes an insight into our current understanding of the signalling molecules and mechanisms that take part in the ordered and timely emergence of various cell types and their biological significance. The prospects that this emerging knowledge may offer for the sustainable control of fungal colonisation or dispersal will also be considered.  相似文献
