This study presents the carbon footprint of a paperback book for which the cover and inside papers were produced in the United States and printed in Canada. The choice of paper mills for both cover and page papers was based on criteria such as percentage of recycled content in the pulp mix, transport distances (pulp mill to paper mill, paper mill to print), and technologies. The cradle‐to‐gate assessment of greenhouse gas (GHG) emissions follows recognized guidelines for carbon footprint assessment. The results show that the production of 400,000 books, mainly distributed in North America, would generate 1,084 tonnes carbon dioxide equivalent (CO2‐eq), or 2.71 kilograms (kg) CO2‐eq per book. The impact of using deinked market pulp (DMP) is shown here to be detrimental, accounting for 54% of total GHG emissions and being 32% higher than reference virgin Kraft pulp. This supports findings that DMP mill GHG emissions strongly correlate with the carbon intensity of the power grid supplying the pulp mill and that virgin Kraft mills that reuse wood residue and black liquor to produce heat and electricity can achieve lower GHG emissions per tonne of pulp produced. Although contrary to common thinking, this is consistent with the Paper Task Force 2002 conclusion for office paper (the closest paper grade to writing paper or fine paper) (EDF 2002a). To get a cradle‐to‐grave perspective, three different end‐of‐life (EOL) scenarios were analyzed, all of which included a harvested wood product (HWP) carbon storage benefit for 25 years. The Italian natural history museums are facing a critical situation, due to the progressive loss of scientific relevance, decreasing economic investments, and scarcity of personnel. This is extremely alarming, especially for ensuring the long-term preservation of the precious collections they host. Moreover, a commitment in fieldwork to increase scientific collections and concurrent taxonomic research are rarely considered priorities, while most of the activities are addressed to public events with political payoffs, such as exhibits, didactic meetings, expositions, and talks. This is possibly due to the absence of a national museum that would have better steered research activities and overall concepts for collection management. We here propose that Italian natural history museums collaborate to instate a “metamuseum”, by establishing a reciprocal interaction network aimed at sharing budgetary and technical resources, which would assure better coordination of common long-term goals and scientific activities. 相似文献

Background
Placental malaria is a disease linked to the sequestration of Plasmodium falciparum infected red blood cells (IRBC) in the placenta, leading to reduced materno-fetal exchanges and to local inflammation. One of the virulence factors of P. falciparum involved in cytoadherence to chondroitin sulfate A, its placental receptor, is the adhesive protein VAR2CSA. Its localisation on the surface of IRBC makes it accessible to the immune system. VAR2CSA contains six DBL domains. The DBL6ε domain is the most variable. High variability constitutes a means for the parasite to evade the host immune response. The DBL6ε domain could constitute a very attractive basis for a vaccine candidate but its reported variability necessitates, for antigenic characterisations, identifying and classifying commonalities across isolates.Methodology/Principal Findings
Local alignment analysis of the DBL6ε domain had revealed that it is not as variable as previously described. Variability is concentrated in seven regions present on the surface of the DBL6ε domain. The main goal of our work is to classify and group variable sequences that will simplify further research to determine dominant epitopes. Firstly, variable sequences were grouped following their average percent pairwise identity (APPI). Groups comprising many variable sequences sharing low variability were found. Secondly, ELISA experiments following the IgG recognition of a recombinant DBL6ε domain, and of peptides mimicking its seven variable blocks, allowed to determine an APPI cut-off and to isolate groups represented by a single consensus sequence.Conclusions/Significance
A new sequence approach is used to compare variable regions in sequences that have extensive segmental gene relationship. Using this approach, the VAR2CSA DBL6 domain is composed of 7 variable blocks with limited polymorphism. Each variable block is composed of a limited number of consensus types. Based on peptide based ELISA, variable blocks with 85% or greater sequence identity are expected to be recognized equally well by antibody and can be considered the same consensus type. Fresh human blood samples were collected from healthy controls and splenectomized and unsplenectomized patients with hereditary spherocytosis due to band 3 or ankyrin and spectrin deficiency. The erythrocytes were separated into age-related fractions using self-forming Percoll density gradients. Membrane proteins were analysed by 2D electrophoresis and identified by mass spectrometry. Annexin VII was present in reticulocytes but was then lost as the cells matured. A different pattern was found in band 3-deficient samples: annexin VII was in fact present in both mature and immature red cell membranes. Cytoskeletal anomalies may then influence the turn-over of annexin VII during erythrocyte maturation. 相似文献

Insulin can generate oxygen free radicals. Statins, 3-hydroxy-3-methylglutaryl-coenzyme A reductase inhibitors, exert a powerful antioxidant effect. The present study aimed to clarify the mechanisms through which insulin generates free radicals and to assess whether pravastatin modulates such effects. In cultured skin fibroblasts from human volunteers exposed to high insulin concentration, either in the presence or in the absence of pravastatin, insulin induced translocation of the p47(phox) subunit of NAD(P)H oxidase from the cytosol to the membrane and generation of radical oxygen species through a PKC delta-dependent mechanism. The insulin-induced translocation of p47(phox) was PKC delta dependent and attenuated by pravastatin, but independent of the activation of Akt and Rac1. Insulin-induced Akt phosphorylation was increased by pravastatin and ERK1/2 phosphorylation attenuated. The present study demonstrates a novel mechanism by which insulin stimulates the generation of free radicals in human fibroblasts, ex vivo. It involves phosphatidylinositol 3-kinase, PKC delta, and p47(phox) translocation and promotes ERK1/2 phosphorylation. Pravastatin inhibited radical oxygen species production by inhibiting PKC delta. These observations offer a robust explanation for the positive effects of pravastatin treatment in patients with insulin resistance syndrome. 相似文献

Tuber ectomycorrhizae in a Tuber magnatum "truffière", located in Central Italy, were studied using molecular methods. Specifically, RFLP-ITS analyses, ITS sequencing and specific probes hybridization were used to identify 335 Tuber-like ectomycorrhizal morphotypes. Molecular identification was possible even when distinct morphological characteristics were lacking. For the first time, T. magnatum ectomycorrhizae and other coexisting Tuber species collected in the field were analysed using molecular tools for unambiguous identification. Although the "truffière" under investigation yields good harvests of T. magnatum fruiting bodies, the percentage of T. magnatum ectomycorrhizae found was very low (less than 4.4% of the 335 root tips analysed), whereas the percentages of Tuber maculatum and Tuber rufum were considerably higher (48.9% and 19.0%, respectively). 相似文献

Abdominal aortic aneurysm (AAA) is a cardiovascular disease with high incidence among elderly population. Biomechanical computational analyses can provide fundamental insights into AAA pathogenesis and clinical management, but modeling should be sufficiently accurate. Several constitutive models of the AAA wall are present in the literature, and some of them seem to well describe the experimental behavior of the aneurysmatic human aorta. In this work we compare a two (2FF) and a four (4FF) fiber families constitutive models of the AAA wall. Both these models satisfactorily fit literature data from biaxial tests on the aneurysmatic tissue. To investigate the peculiar characteristics of these models, we considered the problem of AAA inflation, and solved it by implementing the constitutive equations in a finite element code. A 20% axial stretch was imposed to the aneurysm ends, to simulate the physiological condition. Although fitted on the same dataset, the two material models lead to considerably different outcomes. In particular, adopting a 4FF strain energy function (SEF), an increase of the circumferential stress values can be observed, while higher axial stresses are recorded for the 2FF model. These differences can be attributed to the intrinsic characteristics of the SEFs and to the effective stress field, with respect to the one experienced in biaxial experimental tests on which the fitting is based. In fact the two SEFs appear similar within the region of the stress-strain experimental data, but become different outside it, as in case of aneurysms, due to the effects of the data extrapolation process. It is suggested that experimental data should be obtained for conditions similar to those of the application for which they are intended. 相似文献
