Chilling-sensitive mutants of arabidopsis

Twenty-one mutants ofArabidopsis thaliana were isolated that developed chlorosis or necrosis upon incubation at low temperature (10°C to 15°C). Crosses among mutants in different phenotypic classes showed that mutants in three of four classes were found in a small number of loci. This article is reproduced fromWeeds World, vol. 1. Renal cell culture using autopsy material from children with cystinosis

Renal cell cultures were initiated using fresh autopsy material from two individuals with cystinosis, ages 5 and 8 yr. Cells obtained from collagenase treated autopsy material were grown in a selective kidney medium containing Coon's modified F12, 2.5% fetal bovine serum, transferrin, insulin, selenium, hydrocortisone, PGE1, and fibronectin. These cells had an epithelial appearance, formed domes, and were periodic acid-Schiff positive. Both tight junctions and microvilli were seen by electron microscopy. Fibroblasts had a cloning efficiency of zero in the selective medium and grew poorly compared to their growth in Coon's F12 with 10% fetal bovine serum. The lysosomal cystine content of the renal cells was greatly elevated and comparable to that of fibroblasts from cystinotic patients. Renal cell lysosomal cystine levels were only partially reduced by exposure to either pantethine or the aminothiol, cysteamine. However, exposure to either compound effectively depleted cystinotic cultured fibroblasts of their lysosomal cystine. Study of cultured renal material may have practical significance in pharmacologic considerations.     

Influence of stereotactic hypothalamotomy on alcohol and drug addiction.

2 or 3 years follow-up of 13 hypothalamotomy patients with alcohol and drug addiction reveals that the addicted patients regained their self-control, the severe preoperative social deterioration was removed, and the tendency to social stabilisation was clearly in evidence.     

Analysis of a BrdU-sensitive site in the cactus mouse (Peromyscus eremicus): chromosomal breakage and sister-chromatid exchange

When the thymidine analog BrdU was incorporated into the DNA of a fibroblast cell line derived from the cactus mouse Peromyscus eremicus, a chromosome region with an increased frequency of gaps and breaks was observed. Nearly a third of the chromatid aberrations found at this site were associated with a sister-chromatid exchange (SCE) although this chromosome region showed no increase in sister-chromatid exchange in the absence of a gap or break. SCEs were significantly decreased in the remainder of the chromosome arm when it contained an aberration at the unstable site. This BrdU-sensitive region, unlike others reported, was found not to be late-replicating. — In this chromosome complement, the frequency of sisterchromatid exchange in C-band positive regions was significantly lower than that in C-band negative regions.     

Enhanced rate of ethanol production from D-xylose using recycled or immobilized cells ofPachysolen tannophilus

Summary Enhanced rates of ethanol production byPachysolen tannophilus from D-xylose were obtained by performing the fermentation with recycled cells in suspension culture or immobilized in a Ca-alginate gel. Fermentation under these conditions did not require aeration. Increasing temperature from 30 to 37°C enhanced the amount of ethanol produced in 24 hours from the recycled or the immobilized cells.Issued as National Research Council of Canada Publication Number 19475.
