We have isolated the birch homologue (BP8) for the carrot embryogenic gene DC8 by heterologous hybridization. The birch BP8 gene encodes a putative protein of 53 kDa, showing 52% sequence identity with the DC8 gene at the amino acid level. The putative BP8 protein contains 20 repeats of 11 amino acids and thus belongs to the group of LEA proteins isolated from such plants as carrot, cotton and wheat. Northern hybridization of mRNA isolated from birch cells representing different stages of somatic embryogenesis and non-embryogenetic material with a PB8 probe gave no signals, suggesting a low expression level of the BP8 gene.  相似文献

doi: 10.1111/j.1741‐2358.2011.00495.x Oral health–related quality of life – associations with oral health and conditions in Swedish 70‐year‐old individuals Objective: The aim of this study was to evaluate oral health–related quality of life (OHRQoL) in relation to different chronic oral conditions and perception of oral health among elderly individuals. Background: Health‐related quality of life is important to assess and understand the impact of different diseases and conditions on individuals’ well‐being. Material and methods: Randomly selected 70‐year‐old individuals (N = 561) were included in this analysis. They were clinically examined and answered a questionnaire. Variables used were perceived oral health, satisfaction with appearance of teeth, problems with chewing and dry mouth, burning mouth syndrome, dental anxiety and dental visiting habits. To measure the participants′ perception of the impact of oral conditions on their well‐being, the Oral Health Impact Profile (OHIP‐14) was used. Results: The OHIP‐14 distribution of sum of scores showed a floor effect but revealed acceptable psychometric properties with regard to reliability and validity. Multivariable logistic regression revealed three highly predictive variables for individuals with high OHIP‐14 scores: using dentures, having chewing problems and being dissatisfied with appearance of teeth. Conclusion: Use of dentures, having chewing problems and being dissatisfied with appearance of teeth are associated with high OHIP‐14 sums, indicating strong influence on OHRQoL.  相似文献

Two pentapeptides, Ac-Tyr-Ile-His-Pro-Phe/Ile, were synthesized and shown to have angiotensin II AT2 receptor affinity and agonistic activity. Based on these peptides, a new series of 13 pseudopeptides was synthesized via introduction of five different turn scaffolds replacing the Tyr-Ile amino acid residues. Pharmacological evaluation disclosed subnanomolar affinities for some of these compounds at the AT2 receptor. Substitution of Phe by Ile in this series of ligands enhanced the AT2 receptor affinity of all compounds. These results suggest that the C-terminal amino acid residues can be elaborated on to enhance the AT2 receptor affinity in truncated Ang II analogues.  相似文献

Defects in the mitochondrial protein frataxin are responsible for Friedreich ataxia, a neurodegenerative and cardiac disease that affects 1:40,000 children. Here, we present the crystal structures of the iron-free and iron-loaded frataxin trimers, and a single-particle electron microscopy reconstruction of a 24 subunit oligomer. The structures reveal fundamental aspects of the frataxin mechanism. The trimer has a central channel in which one atom of iron binds. Two conformations of the channel with different metal-binding affinities suggest that a gating mechanism controls whether the bound iron is delivered to other proteins or transferred to detoxification sites. The trimer constitutes the basic structural unit of the 24 subunit oligomer. The architecture of this oligomer and several features of the trimer structure demonstrate striking similarities to the iron-storage protein ferritin. The data reveal how stepwise assembly provides frataxin with the structural flexibility to perform two functions: metal delivery and detoxification.  相似文献

Knowing spatial patterns of functional microbial guilds can increase our understanding of the relationships between microbial community ecology and ecosystem functions. Using geostatistical modeling to map spatial patterns, we explored the distribution of the community structure, size, and activity of one functional group in N cycling, the denitrifiers, in relation to 23 soil parameters over a 44-ha farm divided into one organic and one integrated crop production system. The denitrifiers were targeted by the nirS and nirK genes that encode the two mutually exclusive types of nitrite reductases, the cd1 heme-type and copper reductases, respectively. The spatial pattern of the denitrification activity genes was reflected by the maps of the abundances of nir genes. For the community structure, only the maps of the nirS community were related to the activity. The activity was correlated with nitrate and dissolved organic nitrogen and carbon, whereas the gene pools for denitrification, in terms of size and composition, were influenced by the soil structure. For the nirS community, pH and soil nutrients were also important in shaping the community. The only unique parameter related to the nirK community was the soil Cu content. However, the spatial pattern of the nirK denitrifiers corresponded to the division of the farm into the two cropping systems. The different community patterns, together with the spatial distribution of the nirS/nirK abundance ratio, suggest habitat selection on the nirS- and nirK-type denitrifiers. Our findings constitute a first step in identifying niches for denitrifiers at scales relevant to land management.Soil microorganisms are abundant and diverse (46), drive key processes in biogeochemical cycles, and, thus, play crucial roles in ecosystem functioning (2). They are not randomly distributed but exhibit spatial patterns at different scales (26). Spatial patterns ranging from the micrometer up to the meter scale have been reported (19, 20, 32, 37), and an understanding of such patterns can give clues to how microbial communities are generated and maintained (17). Spatial patterns of microorganisms at the field and landscape scales warrant special attention, since they could be associated with land use and aid in creating knowledge-based management strategies for agricultural production (5, 42). However, our understanding of key habitat-selective factors is limited, and few studies have specified which factors influence the spatial patterns of soil microbial communities at larger scales. Lauber et al. (30) recently demonstrated that pH could predict the community composition of soil bacteria at the continental scale. The importance of pH as a key edaphic driver of bacterial community structure has also been shown in other studies (11, 47). Another major, but complex, factor pointed out in a few studies is the soil type (4, 5, 14). Most studies have included only a limited number of properties that are easy to measure; most often, carbon and nitrogen pools and soil physical factors have been neglected in microbial community ecology. Since these factors delineate soil oxygen and water content, they may exert a stronger impact on microbial communities than other soil resources.Reports on the field or landscape scale spatial distribution of soil bacteria have had a taxon-centered perspective at either the species or total-community level, but there is emerging interest in the biogeography of functional traits possessed by microorganisms (18). Bacterial species composition is likely important for soil ecosystem functions, but species affiliation rarely predicts in which way. In addition, the fuzzy species concept of bacteria makes it all the more difficult to link species to niches. Analysis of functional guilds, i.e., assemblages of populations sharing certain traits, can bridge this gap, and one guild of global concern that has been suggested and recently used as a model in functional ecology is the denitrifiers (39, 49). Denitrification is an anaerobic respiration pathway during which NO3 is reduced to N2 by a wide range of unrelated taxa. The process is an essential route for N loss from agricultural soil and a major source of the greenhouse gas N2O. It was recently shown that the spatial distribution of the relative abundances of denitrifiers with the genetic capacity to perform the last step in the denitrification pathway, reduction of N2O to N2, is linked to areas with high denitrification rates and low N2O emissions (38). Adding field scale predicted patterns of the denitrifier community structure to the abundance and activity would not only give insight into the mechanisms shaping the community, but also deepen our understanding of the relationships between the ecology of denitrifiers, N loss, and the agroecosystems'' impact on climate change.We hypothesize that spatial autocorrelations of the structures, sizes, and activities of communities of denitrifying bacteria is governed by soil-based resources at a scale compatible with land management. To test this, and to elucidate the effects of crop production systems and the importance of soil physical and chemical factors in the denitrifying community, we explored the spatial distribution of community structure, size, and activity in relation to 23 soil parameters at a 44-ha farm divided into one organic and one integrated crop production system. The denitrifier community was described in terms of the signature genes that encode the two different types of nitrite reductases in the denitrification pathway, the cd1 heme-type reductase (NirS), encoded by the nirS gene, and the copper oxidoreductase (NirK), encoded by nirK. In the pursuit towards a systematic analysis of human diseases, array‐based approaches within antibody proteomics offer high‐throughput strategies to discover protein biomarkers in serum and plasma. To investigate the influence of sample preparation on such discovery attempts, we report on a systematic effort to compare serum and plasma protein profiles determined with an antibody suspension bead array. The intensity levels were used to define protein profiles and no significant differences between serum and plasma were observed for 79% of the 174 antibodies (targeting 156 proteins). By excluding 36 antibodies giving rise to differential intensity levels, cluster analysis revealed donor‐specific rather than preparation‐dependent grouping. With a cohort from a clinically relevant medical condition, the metabolic syndrome, the influence of the sample type on a multiplexed biomarker discovery approach was further investigated. Independent comparisons of protein profiles in serum and plasma revealed an antibody targeting ADAMTSL‐4, a protein that would qualify to be studied further in association with the condition. In general, the preparation type had an impact on the results of the applied antibody suspension bead array, and while the technical variability was equal, plasma offered a greater biological variability and allowed to give rise to more discoveries than serum.  相似文献

To predict origins of replication in prokaryotic chromosomes, we analyse the leading and lagging strands of 200 chromosomes for differences in oligomer composition and show that these correlate strongly with taxonomic grouping, lifestyle and molecular details of the replication process. While all bacteria have a preference for Gs over Cs on the leading strand, we discover that the direction of the A/T skew is determined by the polymerase-alpha subunit that replicates the leading strand. The strength of the strand bias varies greatly between both phyla and environments and appears to correlate with growth rate. Finally we observe much greater diversity of skew among archaea than among bacteria. We have developed a program that accurately locates the origins of replication by measuring the differences between leading and lagging strand of all oligonucleotides up to 8 bp in length. Shifting the resource base for chemical production from fossil feedstocks to renewable raw materials provides exciting possibilities for the use of industrial biotechnology-based process tools. This review gives an indication of the current developments in the transition to bio-based production, with a focus on the production of chemicals, and points out some of the challenges that exist in the large-scale implementation of industrial biotechnology. Furthermore, the importance of evaluating the environmental impact of bio-based products with respect to their entire life cycle is highlighted, demonstrating that the choice of the raw material often turns out to be an important parameter influencing the life cycle performance.  相似文献
