The coxsackievirus-adenovirus receptor (CAR) and decay-accelerating factor (DAF) have been identified as cellular receptors for coxsackievirus B3 (CVB3). The first described DAF-binding isolate was obtained during passage of the prototype strain, Nancy, on rhabdomyosarcoma (RD) cells, which express DAF but very little CAR. Here, the structure of the resulting variant, CVB3-RD, has been solved by X-ray crystallography to 2.74 Å, and a cryo-electron microscopy reconstruction of CVB3-RD complexed with DAF has been refined to 9.0 Å. This new high-resolution structure permits us to correct an error in our previous view of DAF-virus interactions, providing a new footprint of DAF that bridges two adjacent protomers. The contact sites between the virus and DAF clearly encompass CVB3-RD residues recently shown to be required for binding to DAF; these residues interact with DAF short consensus repeat 2 (SCR2), which is known to be essential for virus binding. Based on the new structure, the mode of the DAF interaction with CVB3 differs significantly from the mode reported previously for DAF binding to echoviruses.  相似文献

This study develops criteria for classifying the actor groups shaping frontier development along the Transamazon Highway colonization project in the Brazilian Amazon, as a basis to improve understanding of their specific contributions to environmental degradation and socio-economic development. Based on an analysis of responses to questionnaires by 93 colonists representing different migration trajectories, production systems, socio-economic strategies and deforestation patterns, actor groups could be statistically classified according to their type of production and level of capitalization. A property size threshold discriminating small and largeholders in the study area is presented and compared with previous attempts to establish such a threshold. Largeholders practicing large-scale cattle ranching and smallholders practicing diversified family agriculture were found to be the two predominant colonist types. Smallholder farming practices were found to be more appropriate to the local environmental conditions than those implemented by cattle ranchers.  相似文献

Bacterial lipases constitute the most important group of biocatalysts for synthetic organic chemistry. Accordingly, there is substantial interest in developing new valuable lipases. Considering the lack of information concerning the lipases of the genus Rhodococcus and taking into account the interest raised by the enzymes produced by actinomycetes, a search for putative lipase-encoding genes from Rhodococcus sp. strain CR-53 was performed. We isolated, cloned, purified, and characterized LipR, the first lipase described from the genus Rhodococcus. LipR is a mesophilic enzyme showing preference for medium-chain-length acyl groups without showing interfacial activation. It displays good long-term stability and high tolerance for the presence of ions and chemical agents in the reaction mixture. Amino acid sequence analysis of LipR revealed that it displays four unique amino acid sequence motifs that clearly separate it from any other previously described family of bacterial lipases. Using bioinformatics tools, LipR could be related only to several uncharacterized putative lipases from different bacterial origins, all of which display the four blocks of consensus amino acid sequence motifs that contribute to define a new family of bacterial lipases, namely, family X. Therefore, LipR is the first characterized member of the new bacterial lipase family X. Further confirmation of this new family of lipases was performed after cloning Burkholderia cenocepacia putative lipase, bearing the same conserved motifs and clustering in family X. Interestingly, all lipases grouping in the new bacterial lipase family X display a Y-type oxyanion hole, a motif conserved in the Candida antarctica lipase clan but never found among bacterial lipases. This observation contributes to confirm that LipR and its homologs belong to a new family of bacterial lipases.  相似文献

Cellular Ca(2+)signaling results from a complex interplay among a variety of Ca(2+) fluxes going across the plasma membrane and across the membranes of several organelles, together with the buffering effect of large numbers of Ca(2+)-binding sites distributed along the cell architecture. Endoplasmic and sarcoplasmic reticulum, mitochondria and even nucleus have all been involved in cellular Ca(2+) signaling, and the mechanisms for Ca(2+) uptake and release from these organelles are well known. In neuroendocrine cells, the secretory granules also constitute a very important Ca(2+)-storing organelle, and the possible role of the stored Ca(2+) as a trigger for secretion has attracted considerable attention. However, this possibility is frequently overlooked, and the main reason for that is that there is still considerable uncertainty on the main questions related with granular Ca(2+) dynamics, e.g., the free granular [Ca(2+)], the physical state of the stored Ca(2+) or the mechanisms for Ca(2+) accumulation and release from the granules. This review will give a critical overview of the present state of knowledge and the main conflicting points on secretory granule Ca(2+) homeostasis in neuroendocrine cells.  相似文献

The temperature-induced misfolding pathway of PDZ3, the third PDZ domain of the PSD95 neuronal protein, is populated by a trimeric β-sheet-rich intermediate state that leads to a stepwise and reversible formation of supramacromolecular structures. Using FTIR, we have found that misfolding of this pathway is not due to different ensembles of a variety of precursors, but comes mainly from the interconversion of a flexible β-sheet of the domain to wormlike fibrils. The appearance of the wormlike fibril FTIR component is also accompanied by a slight decrease of the band that corresponds to loops in the native state, whereas the rest of the regular elements of secondary structure are fairly well maintained upon misfolding. Transmission electron microscope micrographs have confirmed the presence of wormlike fibrils upon heating at 60°C, where the trimeric intermediate is maximally populated. Toxicity assays in the human neuroblastoma cell line SH-SY5Y show that cytotoxicity increases as the aggregation pathway proceeds. NMR analysis of chemical shifts as a function of temperature has revealed, as one of the main conformational aspects of such an interconversion at the residue level, that the β-sheet arrangement around strand β3 promotes the change that drives misfolding of the PDZ3 domain.  相似文献

In migratory species, the way in which conspecifics from different breeding populations are distributed during the non‐breeding period is important from and ecological, evolutionary and conservation perspective, but such knowledge is still limited for most species. Migratory and sedentary blackcaps Sylvia atricapilla wintering in southern Spain can occupy two habitat types: forests and shrublands. According to earlier studies, blackcaps prefer forests over shrublands, and residents remain nearly restricted to forests. However, whether migrants with different breeding origin occupy the two habitats differently is unknown. We used morphological and biogeochemical data (hydrogen isotope ratios measured on feathers: δ2Hf), which show variation along the breeding range of the species, to answer this question. Isotope analyses supported the reliability of morphology as a method for distinguishing between migratory and sedentary blackcaps in sympatry, showing that sedentary individuals are rare in shrublands while migratory ones are abundant in both habitat types. However, migratory blackcaps scored similar δ2Hf values in forests and shrublands, and neither did vary in structural size or flight morphology between habitats. Our study suggests that migrants from a wide range of breeding origins end up mixing between forests and shrublands, which may explain the patterns of variation in space and time in the abundance of blackcaps in this area, and supports the view that inequalities may arise among migrants with the same origin but wintering in different habitats. Such inequalities might carry over into other stages of blackcaps’ life cycle contributing to the regulation of its migratory populations.  相似文献

Holm oak (Quercus ilex ssp. ballota [Desf.] Samp) is one of the most important species in forest communities of the western Mediterranean Basin, but is very vulnerable to environmental stress during the first years of its life. In particular, summer drought and winter frosts limit the distribution, survival, and growth of Holm oak. These two stress factors can lead to plasmolysis and/or oxidative stress. We hypothesized that autumn fertilization with nitrogen (N) can improve plant tolerance to these abiotic stress factors and improve plant quality and therefore reforestation success. A 12-week autumn application of 30 or 60 mg N (plus 70 mg N applied to both treatments during the previous 28 weeks in the nursery, i.e. 100 and 130 mg N in total, respectively) improved overall growth, root growth capacity, frost tolerance, and nutritional status of plants relative to plants given 0.0 and 1.5 mg autumnal N, and had no negative effect on seedling response to water stress. A very small increment in N doses during the autumn (1.5 vs. 0.0 mg N) improved some morphological parameters, such as stem diameter (D) and shoot dry weight, and physiological parameters, such as total antioxidant activity. The highest autumnal N dose (60 mg) increased leaf nutrient content without decreasing the concentration, but had a tendency to reduce frost tolerance relative to seedlings given a lower autumnal N dose (30 mg).  相似文献

In this study, we find and characterize the sources of tolerance to drought amongst an oat (Avena sativa L.) germplasm collection of 174 landraces and cultivars. We used multivariate analysis, non-supervised principal component analyses (PCA) and supervised discriminant function analyses (DFA) to suggest the key mechanism/s responsible for coping with drought stress. Following initial assessment of drought symptoms and area under the drought progress curve, a subset of 14 accessions were selected for further analysis. The collection was assessed for relative water content (RWC), cell membrane stability, stomatal conductance (g 1), leaf temperature, water use efficiency (WUE), lipid peroxidation, lipoxygenase activity, chlorophyll levels and antioxidant capacity during a drought time course experiment. Without the use of multivariate approaches, it proved difficult to unequivocally link drought tolerance to specific physiological processes in the different resistant oat accessions. These approaches allowed the ranking of many supposed drought tolerance traits in the order of degree of importance within this crop, thereby highlighting those with a causal relationship to drought stress tolerance. Analyses of the loading vectors used to derive the PCA and DFA models indicated that two traits involved in water relations, temperature and RWC together with the area of drought curves, were important indicators of drought tolerance. However, other parameters involved in water use such as g 1 and WUE were less able to discriminate between the accessions. These observations validate our approach which should be seen as representing a cost-effective initial screen that could be subsequently employed to target drought tolerance in segregating populations.  相似文献

Background

Spinal cord injury is a major cause of long-term disability and has no current clinically accepted treatment. Leptin, an adipocyte-derived hormone, is best known as a regulator of food intake and energy expenditure. Interestingly, several studies have demonstrated that leptin has significant effects on proliferation and cell survival in different neuropathologies. Here, we sought to evaluate the role of leptin after spinal cord injury.

Findings

Based on its proposed neuroprotective role, we have evaluated the effects of a single, acute intraparenchymal injection of leptin in a clinically relevant animal model of spinal cord injury. As determined by quantitative Real Time-PCR, endogenous leptin and the long isoform of the leptin receptor genes show time-dependent variations in their expression in the healthy and injured adult spinal cord. Immunohistochemical analysis of post-injury tissue showed the long isoform of the leptin receptor expression in oligodendrocytes and, to a lesser extent, in astrocytes, microglia/macrophages and neurons. Moreover, leptin administered after spinal cord injury increased the expression of neuroprotective genes, reduced caspase-3 activity and decreased the expression of pro-inflammatory molecules. In addition, histological analysis performed at the completion of the study showed that leptin treatment reduced microglial reactivity and increased caudal myelin preservation, but it did not modulate astroglial reactivity. Consequently, leptin improved the recovery of sensory and locomotor functioning.

Conclusions

Our data suggest that leptin has a prominent neuroprotective and anti-inflammatory role in spinal cord damage and highlights leptin as a promising therapeutic agent.  相似文献
