REV1 protein, a eukaryotic member of the Y family of DNA polymerases, is involved in the tolerance of DNA damage by translesion DNA synthesis. It is unclear how REV1 is recruited to replication foci in cells. Here, we report that mouse REV1 can bind directly to PCNA and that monoubiquitylation of PCNA enhances this interaction. The interaction between REV1 protein and PCNA requires a functional BRCT domain located near the N terminus of the former protein. Deletion or mutational inactivation of the BRCT domain abolishes the targeting of REV1 to replication foci in unirradiated cells, but not in UV-irradiated cells. In vivo studies in both chicken DT40 cells and yeast directly support the requirement of the BRCT domain of REV1 for cell survival and DNA damage-induced mutagenesis.  相似文献

Alterations in DNA repair lead to genomic instability and higher risk of cancer. DNA base excision repair (BER) corrects damaged bases, apurinic sites, and single-strand DNA breaks. Here, a regulatory mechanism for DNA polymerase beta (Pol beta) is described. Pol beta was found to form a complex with the protein arginine methyltransferase 6 (PRMT6) and was specifically methylated in vitro and in vivo. Methylation of Pol beta by PRMT6 strongly stimulated DNA polymerase activity by enhancing DNA binding and processivity, while single nucleotide insertion and dRP-lyase activity were not affected. Two residues, R83 and R152, were identified in Pol beta as the sites of methylation by PRMT6. Genetic complementation of Pol beta knockout cells with R83/152K mutant revealed the importance of these residues for the cellular resistance to DNA alkylating agent. Based on our findings, we propose that PRMT6 plays a role as a regulator of BER.  相似文献

A well-defined copolymer poly(oligo(ethylene glycol) methacrylate-co-methacrylic acid) P(OEGMA-co-MAA) was studied as a novel water-soluble biocompatible coating for superparamagnetic iron oxide nanoparticles. This copolymer was prepared via a two-step procedure: a well-defined precursor poly(oligo(ethylene glycol) methacrylate-co-tert-butyl methacrylate), P(OEGMA-co-tBMA) (M(n) = 17300 g mol(-1); M(w)/M(n) = 1.22), was first synthesized by atom-transfer radical polymerization in the presence of the catalyst system copper(I) chloride/2,2'-bipyridyl and subsequently selectively hydrolyzed in acidic conditions. The resulting P(OEGMA-co-MAA) was directly utilized as a polymeric stabilizer in the nanoparticle synthesis. Four batches of ultrasmall PEGylated magnetite nanoparticles (i.e., with an average diameter below 30 nm) were prepared via aqueous coprecipitation of iron salts in the presence of variable amounts of P(OEGMA-co-MAA). The diameter of the nanoparticles could be easily tuned in the range 10-25 nm by varying the initial copolymer concentration. Moreover, the formed PEGylated ferrofluids exhibited a long-term colloidal stability in physiological buffer and could therefore be studied in vivo by magnetic resonance (MR) imaging. Intravenous injection into rats showed no detectable signal in the liver within the first 2 h. Maximum liver accumulation was found after 6 h, suggesting a prolongated circulation of the nanoparticles in the bloodstream as compared to conventional MR imaging contrast agents.  相似文献

At a time when tetravalent conjugate vaccines for meningococcal serogroups A/C/Y/W135 are being formulated the O-acetylation status of their respective capsular polysaccharides has not previously been studied in the UK for all components. Although this has been elucidated for serogroup C, little is known about the O-acetylation status of serogroups W135 and Y. Meningococcal serogroup W135 (n=181) and Y (n=90) isolates submitted to the PHLS Meningococcal Reference Unit in 1996, 2000 and 2001 were investigated for O-acetylation capsular status by dot blot assay. Eight per cent of W135 and 79% of Y isolates respectively were found to be O-acetylated with a similar distribution found in both carrier and case isolates. An increase in O-acetylated W135 isolates was noted between 2000 (0%) and 2001 (21%) which was not due to the introduction of the Hajj associated W135 (ET 37 complex; serosubtype P1.5,2) isolates, all of which were de-O-acetylated. Although the biological relevance of O-acetylation status is unknown for these serogroups, an understanding of O-acetylation status of the respective polysaccharides may provide useful insights into the optimal vaccine formulation.  相似文献

NADP-dependent methylene-H(4)MPT dehydrogenase, MtdA, from Methylobacterium extorquens AM1 catalyzes the dehydrogenation of methylene-tetrahydromethanopterin and methylene-tetrahydrofolate with NADP(+) as cosubstrate. The X-ray structure of MtdA with and without NADP bound was established at 1.9 A resolution. The enzyme is present as a homotrimer. The alpha,beta fold of the monomer is related to that of methylene-H(4)F dehydrogenases, suggesting a common evolutionary origin. The position of the active site is located within a large crevice built up by the two domains of one subunit and one domain of a second subunit. Methylene-H(4)MPT could be modeled into the cleft, and crucial active site residues such as Phe18, Lys256, His260, and Thr102 were identified. The molecular basis of the different substrate specificities and different catalytic demands of MtdA compared to methylene-H(4)F dehydrogenases are discussed.  相似文献

Adaptive immune systems are present only in vertebrates. How do all the remaining animals withstand continuous attacks of permanently evolving pathogens? Even in the absence of adaptive immunity, every organism must be able to unambiguously distinguish "self" cells from any imaginable "nonself." Here, we analyzed the function of highly polymorphic gene vCRL1, which is expressed in follicle and blood cells of Ciona intestinalis, pointing to possible recognition roles either during fertilization or in immune reactions. By using segregation analysis, we demonstrate that vCRL1 locus is not involved in the control of self-sterility. Interestingly, genetic knockdown of vCRL1 in all tissues or specifically in hemocytes results in a drastic developmental arrest during metamorphosis exactly when blood system formation in Ciona normally occurs. Our data demonstrate that vCRL1 gene might be essential for the establishment of a functional blood system in Ciona. Presumably, presence of the vCRL1 receptor on the surface of blood cells renders them as self, whereas any cell lacking it is referred to as nonself and will be consequently destroyed. We propose that individual-specific receptor vCRL1 might be utilized to facilitate somatic self/nonself discrimination.  相似文献

Peripheral neuropathy develops in human subjects with prediabetes and metabolic syndrome before overt hyperglycemia. The contributions of impaired glucose tolerance and insulin signaling, hypertriglyceridemia and/or increased nonesterified fatty acids (NEFA), and hypercholesterolemia to this condition remain unknown. Niacin and its derivatives alleviate dyslipidemia with a minor effect on glucose homeostasis. This study evaluated the roles of impaired glucose tolerance versus dyslipidemia in prediabetic neuropathy using Zucker fatty (fa/fa) rats and the niacin derivative acipimox, as well as the interplay of hypertriglyceridemia, increased NEFA, and oxidative-nitrosative stress. Sixteen-week-old Zucker fatty rats with impaired glucose tolerance, obesity, hyperinsulinemia, hypertriglyceridemia, hypercholesterolemia, and increased NEFA displayed sensory nerve conduction velocity deficit, thermal and mechanical hypoalgesia, and tactile allodynia. Acipimox (100 mg kg(-1) day(-1), 4 weeks) reduced serum insulin, NEFA, and triglyceride concentrations without affecting glucose tolerance and hypercholesterolemia. It alleviated sensory nerve conduction velocity deficit and changes in behavioral measures of sensory function and corrected oxidative-nitrosative stress, but not impaired insulin signaling, in peripheral nerve. Elevated NEFA increased total and mitochondrial superoxide production and NAD(P)H oxidase activity in cultured human Schwann cells. In conclusion, hypertriglyceridemia and/or increased NEFA concentrations cause prediabetic neuropathy through oxidative-nitrosative stress. Lipid-lowering agents and antioxidants may find a use in the management of this condition.  相似文献

The concern about climate change has re-vitalised the interest in the relationships between body-size of organisms and temperature both at the intraspecific level (James’ rule, Temperature-Size-Rule) and at the interspecific level (Bergmann's rule). In order to test the expected shifts towards smaller body size under warming conditions, a mesocosm experiment using plankton from the Baltic Sea at three temperature levels was performed in April 2010. The hypothesis was tested, that a tendency well established under monoculture conditions, would also be found in phytoplankton embedded in a semi-natural food-web. Six out of seven phytoplankton species abundant enough for analysis did not show the expected shrinkage of cell volume with increasing temperature, while volume shrinkage of the 7th species (Eutreptiella sp.) could be attributed exclusively to the axis which is affected by cell division. Thus it could not be ruled out, that the populations grown at different temperature levels were just at a different stage of the cell cycle. Similarly, we could not find an indication for a replacement of larger species by smaller ones under higher temperature. As a consequence, mean cell size did not respond to temperature. Therefore, we could not support the hypothesis, that warming should benefit the small.  相似文献

Several land snail species are highly polymorphic regarding their shell colouration. This polymorphism has been related to predatory effects as well as climatic reasons, assuming that dark morphs benefit from being more cryptic and therefore less prone to predation, whereas pale morphs are at an advantage under solar radiation, as they are suspected to heat up less. However, the assumption of different thermal capacities of these morphs is based on experiments with little standardisation or little environmental relevance. In this study, we aimed at measuring thermal capacities of two different morphs (pale versus dark-brown banded) of the Mediterranean land snail Theba pisana, applying a standardised and environmentally relevant test set-up, in order to prove whether darker morphs indeed do heat up more than lighter coloured morphs. We did not find any differences in the thermal capacity of the different morphs and conclude that thermal capacity of the shell is predominantly defined by its material rather than its coloration. These results are discussed with regard to previous studies on thermal characteristics of different land snail morphs and correlations between climate and morph distribution.  相似文献

Mate finding and dispersal from the natal patch in parasitoid Hymenoptera are influenced by the availability of host resources and interactions with other organisms. We compared the mating behavior of three solitary aphid parasitoids, Aphidius ervi Haliday, Lysiphlebus hirticornis Mackauer and Pauesia pini (Haliday) (Hymenoptera: Braconidae: Aphidiinae) that differ in host resource exploitation and ant mutualism. In L. hirticornis, which is obligately ant-attended, the residence time on the natal patch was approximately 4 h compared with less than 2 h in the facultatively ant-attended P. pini; the sexes did not differ in residence time. Females of A. ervi, which is not attended by ants, stayed for slightly more than 2 h on the natal patch while their male siblings remained for only 1 h. In L. hirticornis, 90% of all siblings in a clutch mated on the natal patch but only 13% in A. ervi and 42% in P. pini did so. Off-patch matings (23%) were observed only in A. ervi. Males and females of L. hirticornis were 12-times more likely to mate on the natal patch when aphids and ants were present than when either of the latter species was removed; and patch residence time declined from approximately 4 h to approximately 2.5 h in the absence of either aphids or ants. We propose that, in aphidiine wasps and perhaps other quasigregarious parasitoids, mating behavior is influenced by the availability of resources on the natal patch and the presence or absence of trophobiotic ants. Partial sib mating is expected in species producing large clutches and having a long patch residence time.  相似文献
